[Fecal microbiota transplantation in treatment of inflammatory bowel diseases].

Inflammatory bowel diseases (IBD) are chronic, recurrent disease associated with significant morbidity and disability rates and marked reduction of quality of life. The exact aetiology of these conditions is unknown, however, there is increasing data supporting the influence of gut microbiota in the pathogenesis of IBD. Despite of large number of actively exploring approaches to IBD treatment, some patients remain refractory to standard management or have significant adverse side effects. Given the probably role of the gastrointestinal microbiotain development of IBD, treatments that manipulat…

Intestinal microbiota promote enteric virus replication and systemic pathogenesis.

Intestinal bacteria aid host health and limit bacterial pathogen colonization. However, the influence of bacteria on enteric viruses is largely unknown. We depleted the intestinal microbiota of mice with antibiotics before inoculation with poliovirus, an enteric virus. Antibiotic-treated mice were less susceptible to poliovirus disease and supported minimal viral replication in the intestine. Exposure to bacteria or their N-acetylglucosamine-containing surface polysaccharides, including lipopolysaccharide and peptidoglycan, enhanced poliovirus infectivity. We found that poliovirus binds lipopo…

Whole mouth microbiota effects following subgingival delivery of sanguinarium.

An increased incidence of antibiotic-resistant bacteria and yeast overgrowth has been reported following various periodontal treatments. The objective of this study was to detect possible overgrowth of opportunistic bacteria and fungi as well as changes in normal microbiota after application of a biodegradable delivery system containing 5% sanguinarium (ABDS-S) to one quadrant in a split-mouth study. An oral hygiene quadrant served as a control. The ABDS-S treated and control periodontal sites as well as the saliva of 17 subjects were sampled prior to treatment, immediately after ABDS-S remova…

Fecal microbiota transplantation for vancomycin-resistant Clostridium innocuum infection in inflammatory bowel disease: A pilot study evaluating safety and clinical and microbiota outcome.

BACKGROUND: Clostridium innocuum is a vancomycin-resistant pathobiome associated with poor clinical outcomes in inflammatory bowel disease (IBD). In ulcerative colitis (UC), it correlates with reduced remission rates, while in Crohn's disease (CD), it is linked to creeping fat formation and intestinal strictures. Notably, some patients experience refractory or recurrent C. innocuemailum infections despite metronidazole treatment. This study evaluates the safety and efficacy of single-dose fecal microbiota transplantation (FMT) in IBD patients with refractory or recurrent C. innocuum infections…

Gut microbiota and epigenetic mechanisms in brain physiology and disorders.

The interplay between gut microbiota and epigenetic mechanisms plays a crucial role in brain physiology and disorders, highlighting a novel area of research with significant implications for understanding and treating neurologic conditions. The gut microbiota, the diverse community of microorganisms in the digestive tract, influences brain function and health through the gut-brain axis, a complex communication network linking the gastrointestinal tract and the central nervous system. Epigenetic mechanisms, including DNA methylation, histone modification, and noncoding RNA activity, regulate ge…

L-serine at the crossroads of microbiota, intestinal health, and disorders.

L-serine is an amino acid involved in maintaining intestinal mucosal homeostasis through its role in protein synthesis but also in redox balance, immune function and lipid metabolism. It also contributes to mucus production, the epithelial barrier integrity and gut microbiota composition. Prokaryotic cells, including Escherichia coli, have developed mechanisms to exploit L-serine to support their growth, virulence, inflammatory and/or carcinogenic properties. In this review, we summarize current findings on L-serine metabolism in the intestinal mucosal homeostasis, its interactions with microb…

The role of gut microbiota in the neurobiology and treatment of Alzheimer's disease.

Alzheimer's disease (AD) is the most common cause of dementia in the elderly population and characterized by progressive cognitive decline. The major pathological features of AD are the accumulation of extracellular amyloid-beta protein as neuritic plaques and intracellular hyperphosphorylated tau protein as neurofibrillary tangles. Studies have shown that gut microbiota are involved in several central nervous system disorders through regulation of neurotransmitter production, blood-brain barrier permeability and immune responses. The gut microbiota establishes a two-way communication between …

Safety and efficacy of faecal microbiota transplantation in Parkinson's disease.

The gut microbiota may influence Parkinson's disease's (PD) pathophysiology. Faecal Microbiota Transplantation (FMT) may therefore improve symptoms. We reviewed five randomized controlled trials (RCT), three cohort studies, one case series, and two single case reports (total of 236 subjects). FMT methods varied (donor selection, FMT preparation method, pre-treatment, administration). Mild, self-limiting gastrointestinal symptoms were the most common adverse events (AEs). Constipation improved consistently after FMT. Motor and non-motor outcomes were heterogeneous, with improvements mainly repo…

The Effect of Microbiota and Evaluation of Released Cytokines in Schizophrenia Patients.

Bidirectional communication between the gut microbiota and the brain has been increasingly recognized, with growing evidence suggesting its relevance in schizophrenia. Altered microbiota composition may contribute to gastrointestinal barrier disruption, immune activation, and metabolic dysfunction. Given the multifactorial etiology of schizophrenia involving genetic, epigenetic, and environmental factors, microbiota-related mechanisms may represent an important pathway in disease development. This prospective, controlled study investigated gut microbiota composition and systemic inflammatory m…

Microbiota-Derived Metabolites in the Epigenetic Regulation of Redox Homeostasis.

Redox homeostasis is essential for intestinal and systemic health and is regulated by antioxidant defense systems and redox-sensitive signaling pathways such as the nuclear factor erythroid 2-related factor 2 (Nrf2) and the nuclear factor 'kappa-light-chain-enhancer' of activated B-cells (NF-κB). Disturbances in this balance promote oxidative stress, chronic inflammation, and disease progression. Increasing evidence indicates that microbiota-derived metabolites act as key modulators of redox biology by shaping host gene expression through receptor-mediated signaling, metabolic regulation, and …
