Comparative insights into transition metal mediated nitrite to nitric oxide conversion.

Nitrite (NO2-) is a widespread environmental pollutant that contributes to eutrophication, nitrosative stress, and N-nitrosamine formation. The chemical factors that make copper optimal for nitrite to NO conversion remain central to understanding copper nitrite reductase activity. Here, we compare ascorbate-driven nitrite reduction using transition-metal salts and synthesized ligand-supported Cu(II) complexes under identical aqueous conditions. CuCl2 shows the highest activity, producing NO quantitatively within 8 s with an initial NO-generation rate of 15.6 cm3 s-1 and a TOF of 45 000 h-1. Th

Temporal Neuroimmune Changes During Primary and Persistent Experimental Toxoplasmic Encephalitis.

Toxoplasma gondii persists in the central nervous system and may cause severe toxoplasmic encephalitis, particularly in immunocompromised hosts. Our previous studies demonstrated that T. gondii-induced neuropathology involves oxidative and nitrosative stress, apoptosis, glial activation, blood-brain barrier injury, and GMF-β- and SAP/NF-κB-associated responses. This study investigated the temporal immunoreactivity of CD68, S100B, Foxp3, and IL-5 during early and chronic experimental infection. Twenty-four female Swiss albino mice were assigned to one uninfected control group and three ME49-inf

S-allyl-l-cysteine ameliorates sensorimotor functions after intracerebral hemorrhage in mice concomitantly with prevention of axon tract fragmentation and reduction of brain lesion volume.

Intracerebral hemorrhage (ICH) is a devastating type of stroke with high mortality and limited treatment options. Brain tissue injury featured by neuroinflammation and axonal damage plays a pivotal role in poor outcome. S-allyl-l-cysteine, a garlic-derived amino acid derivative, possesses antioxidant and anti-inflammatory properties and has been shown to afford neuroprotection in ischemic stroke models. In the present study, ICH was induced in male ICR mice by collagenase injection into the striatum. When S-allyl-l-cysteine (300 or 600 mg/kg) was administered intraperitoneally at 1 h before an

Reduction in fungal viability, mycotoxicity, and cytotoxicity in green coffee beans treated with plasma nitric oxide water.

Coffee ranks among the world's most consumed beverages, yet fungal contamination and associated mycotoxins (ochratoxin A (OTA)) remain major safety concerns. This study evaluated cold plasma generated nitric oxide continuously dissolved in water (plasma NO water, P-NOW) as a nonthermal approach for simultaneous fungal inactivation and OTA detoxification for green coffee beans (GCBs). A 15-min P-NOW treatment achieved >2 log reductions in Aspergillus species accompanied by strong inhibition of mycelial growth after 120 h. Fluorescence imaging and flow cytometry confirmed compromised membrane in

Oral Honokiol-loaded solid lipid nanoparticles: A shielding nanotherapeutic strategy to attenuate STZ-induced type 1 diabetes via targeted modulation of oxidative stress and apoptotic signaling.

This study aimed to analyze the protective effects of Honokiol-loaded oral solid lipid nanoparticles (Honokiol-SLNs) on pancreatic beta-cells (β-cells) in an experimental model of type 1 diabetes. Streptozotocin (STZ) injection (150 mg/kg) was used to induce diabetes in male Swiss mice. Honokiol-SLNs were administered daily during the 28-day experimental phase, commencing after the onset of hyperglycemia. After the experiment, β-cell activity, apoptosis, oxidative stress, and inflammation were assessed in pancreatic tissue and blood. In vitro experiments have preliminarily validated the abilit

DAPT Mitigates Osteoarthritic Cartilage Degeneration and Enhances Articular Repair Through Targeted Modulation of the Gucy1a3 and Notch Signaling Axis.

BACKGROUND: While Nitric oxide (NO) and Notch signaling pathways are implicated in osteoarthritis (OA) progression, their functional interplay remains largely unexplored. We hypothesized that Gucy1a3 (sGCα1) functions as a non-canonical soluble sensor, bridging nitrosative stress to pathological Notch activation. METHODS: To test the hypothesis that Gucy1a3 mediates NO-Notch crosstalk, we used both sodium nitroprusside (SNP)-stimulated chondrocytes and a rat medial meniscus resection model. Mechanistic hierarchy was analyzed via Gucy1a3 siRNA knockdown and the Notch inhibitor DAPT. Additional

Evaluation of the Anti-Alzheimer Activity of Lycium barbarum Polysaccharide in Aβ1-42-Induced Neurotoxicity in Rat Model.

As a common neurodegenerative disorder, Alzheimer's disease (AD) manifests as progressive memory loss, cognitive deficits, and dementia in older adults. As the basis of the traditional Chinese medicinal herb Goji berries, Lycium barbarum polysaccharide (LBP) has been proven to exhibit multiple pharmacological activities, including antioxidant, neuroprotective, and anti-inflammatory effects. Evidence supports that LBP can enhance cognitive function and holds promise in counteracting AD. In order to determine the neuroprotective effects of LBP, this study was conducted on an AD rat model induced

CCAAT/Enhancer-Binding Protein Beta Nitration Participates in Hyperhomocysteinemia-Induced Cardiomyocyte Autophagic Flux Blockage by Inhibiting Transcription Factor EB Transcription.

Aims: Autophagy is a protective mechanism of cardiomyocytes. Hyperhomocysteinemia (HHcy) elevates oxidative and nitrosative stress levels, leading to an abnormal increase in nitration protein, possibly leading to abnormal autophagy regulation in cardiomyocytes. However, the regulatory effect of HHcy on autophagy at the post-translational modification level is still unclear. Here, we aimed to explore the regulatory mechanism of HHcy on transcription factor EB (TFEB) and nitration of CCAAT/enhancer-binding protein beta (C/EBPβ), a transcriptional repressor of Tfeb, on autophagy in cardiomyocytes

Biological Mechanisms Involved in Muscle Dysfunction in COPD: An Integrative Damage-Regeneration-Remodeling Framework.

Skeletal muscle dysfunction is a major systemic manifestation of COPD that shapes symptoms, exercise tolerance and mortality. Current evidence can be integrated within a Damage-Regeneration-Remodeling framework linking mechanics and biology to clinical phenotypes. Pulmonary hyperinflation and chest wall geometry chronically load the diaphragm and other respiratory muscles in COPD, whereas inactivity and exacerbation-related disuse underload locomotor muscles. Across muscle compartments, oxidative/nitrosative stress, activation of proteolytic pathways, mitochondrial and endoplasmic reticulum st

Targeted metabolomic evaluation of peripheral blood mononucleated cells from patients with PMM2-CDG.

Phosphomannomutase-2 (PMM2) deficiency represents the most common congenital disorder of glycosylation (CDG). Currently, little is known about cell metabolic alterations occurring in these patients. Here, we quantified compounds connected to protein glycosylation (GDP-mannose, UDP-derivatives), energy metabolism (high-energy phosphates, nicotinic coenzymes, oxypurines), oxidative/nitrosative stress (GSH, nitrite, nitrate) and free amino acids in extracts of peripheral blood mononucleated cells (PBMCs), of seven PMM2-CDG patients and ten control healthy donors. Besides marked GDP-mannose decrea

Unveiling the silent defenders: mycobacterial stress sensors at the forefront to combat tuberculosis.

The global escalation in tuberculosis (TB) cases accompanied by the emergence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains of Mycobacterium tuberculosis (M.tb) emphasizes the critical requirement for novel potent drugs. The M.tb demonstrates extraordinary adaptability, thriving in diverse conditions, and always finds itself in win-win situations regardless of whether the environment is favorable or unfavorable; no matter the magnitude of the challenge, it can endure and survive. This review aims to uncover the role of multiple stress sensors of M.tb that assist bac

Marine Algae and Deriving Biomolecules for the Management of Inflammatory Bowel Diseases: Potential Clinical Therapeutics to Decrease Gut Inflammatory and Oxidative Stress Markers?

The term "inflammatory bowel disease" (IBD) describes a class of relapse-remitting conditions that affect the gastrointestinal (GI) tract. Among these, Crohn's disease (CD) and ulcerative colitis (UC) are two of the most globally prevalent and debilitating conditions. Several articles have brought attention to the significant role that inflammation and oxidative stress cooperatively play in the development of IBD, offering a different viewpoint both on its etiopathogenesis and on strategies for the effective treatment of these conditions. Marine ecosystems may be a significant source of physio

Berberine Attenuates Cerulein-Induced Acute Pancreatitis by Modulating Nrf2/NOX2 Signaling Pathway via AMPK Activation.

AMP-activated protein kinase (AMPK) is the master regulator of cellular energy which gets activated during energy stress and restores tissue homeostasis. AMPK is widely expressed in the pancreas and is involved in protein synthesis. In cerulein-induced acute pancreatitis (AP), diminished AMPK activity in the pancreatic tissue may be associated with pancreatic inflammation and oxidative stress. Our results demonstrated that berberine (BR) treatment produced significant decrease in plasma amylase and lipase levels and improved histopathological features in AP mice model. Melatonin-Nitric Oxide Crosstalk in Plants and the Prospects of NOMela as a Nitric Oxide Donor.

Melatonin regulates vital physiological processes in animals, such as the circadian cycle, sleep, locomotion, body temperature, food intake, and sexual and immune responses. In plants, melatonin modulates seed germination, longevity, circadian cycle, photoperiodicity, flowering, leaf senescence, postharvest fruit storage, and resistance against biotic and abiotic stresses. In plants, the effect of melatonin is mediated by various regulatory elements of the redox network, including RNS and ROS. Similarly, the radical gas NO mediates various physiological processes, like seed germination, flower

Replicative Endothelial Cell Senescence May Lead to Endothelial Dysfunction by Increasing the BH2/BH4 Ratio Induced by Oxidative Stress, Reducing BH4 Availability, and Decreasing the Expression of eNOS.

Vascular aging is associated with the development of cardiovascular complications, in which endothelial cell senescence (ES) may play a critical role. Nitric oxide (NO) prevents human ES through inhibition of oxidative stress, and inflammatory signaling by mechanisms yet to be elucidated. Endothelial cells undergo an irreversible growth arrest and alter their functional state after a finite number of divisions, a phenomenon called replicative senescence. We assessed the contribution of NO during replicative senescence of human aortic (HAEC) and coronary (CAEC) endothelial cells, in which accum

Insight into the on/off switch that regulates expression of the MSMEG-3762/63 efflux pump in Mycobacterium smegmatis.

Drug resistance is one of the most difficult challenges facing tuberculosis (TB) control. Drug efflux is among the mechanisms leading to drug resistance. In our previous studies, we partially characterized the ABC-type MSMEG-3762/63 efflux pump in Mycobacterium smegmatis, which shares high percentage of identity with the Mycobacterium tuberculosis Rv1687/86c pump. MSMEG-3762/63 was shown to have extrusion activity for rifampicin and ciprofloxacin, used in first and second-line anti-TB treatments. Moreover, we described the functional role of the TetR-like MSMEG-3765 protein as a repressor of t

Neuroprotective potential of saroglitazar in 6-OHDA induced Parkinson's disease in rats.

Parkinson's disease (PD) is a neurodegenerative disorder that affects 2%-3% of the population worldwide. Clinical presentation of PD includes motor and non-motor symptoms. The interplay between pathogenic factors such as increased oxidative stress, neuroinflammation, mitochondrial dysfunction and apoptosis are responsible for neurodegeneration in PD. Intrastriatal administration of 6-hydroxy dopamine (6-OHDA) in rat brain provoked oxidative and nitrosative stress by decreasing endogenous antioxidants such as superoxide dismutase, catalase, glutathione, glutathione peroxidase and glutathione re
