An ovary-targeted nucleic acid delivery system, OT-NADPS, efficiently mediates the generation of transgenic silkworms.

Transgenic technology is crucial for advancing various areas of biological research. In silkworms, the traditional transgenic method relies on embryo microinjection, which is a labor-intensive process with limited efficiency and high technical barriers. To overcome these challenges, we developed an Ovary-Targeted Nucleic Acid Delivery Peptide, OT-NADPS, based on the Receptor-Mediated Ovary Transduction of Cargo system (ReMOT) by innovative integration. OT-NADPS, which consists of a nucleic acid loading domain and an ovarian targeting peptide, demonstrates strong nucleic acid binding capacity a

Designed Ankyrin Repeat Protein-Mediated Peptide Nucleic Acid-Based Pretargeting: A Proof-of-Principle Study.

Designed ankyrin repeat proteins (DARPins) are a class of engineered scaffold proteins (ESPs) with a molecular weight of approximately 15 kDa and a picomolar affinity for tumor antigen targets. Proof-of-concept studies have demonstrated the potential of DARPin radioimmunodiagnostics in humans. However, a high accumulation of activity in the kidneys limits their use in conventional radionuclide therapy. A peptide nucleic acid (PNA)-based pretargeted approach was successfully applied to Affibody molecules, another class of ESP. We hypothesized that this method could also enable the controlled co

Maximising opportunity for therapeutic success: sequential participation in cystic fibrosis nucleic acid-based therapy trials.

Identifying safe and effective therapies that target the underlying cause of cystic fibrosis remains a key priority for the cystic fibrosis community. CFTR modulators are first-in-class, regulatory-approved therapies that improve the function of the protein encoded by the CFTR gene and are associated with dramatic and sustained clinical benefits. Although approximately 90% of the population with cystic fibrosis could benefit from these therapies based on genetic eligibility, a crucial unmet need remains: developing CFTR-directed therapies for the ultra-rare population with cystic fibrosis who

Rapid Detection of Saxitoxin Using a Nucleic Acid Aptamer Biosensor Based on Graphene Oxide as a Fluorescence Quencher.

Saxitoxin (STX) is a toxin with paralyzing and lethal properties, necessitating the development of a simple analytical method. This study developed a nucleic acid aptamer biosensor using graphene oxide (GO) as a fluorescence quencher for STX detection. GO was combined with M30-f, an STX nucleic acid aptamer modification with 5-carboxyfluorescein, which can produce fluorescence absorption under the conditions of an excitation wavelength of 408 nm and emission wavelength of 515 nm. Based on the principle of fluorescence resonance energy transfer, the fluorescence of M30-f was quenched. In the pr

Molecular mechanisms of immune evasion by host protein glycosylation of a bacterial immunogen used in nucleic acid vaccines.

Nucleic acid vaccines, including mRNA platforms, represent a transformative approach by inducing adaptive immunity against in situ-expressed antigens. Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a leading global health threat. The only licensed TB vaccine, Bacillus Calmette-Guérin (BCG), offers limited protection against pulmonary TB in adults and adolescents. Underscoring the urgent need for more effective vaccination strategies. However, clinical trials targeting the Mtb Ag85 complex proteins as nucleic acid vaccines have failed to confer protective immunity. We hy

Benchmarking a massively parallel nucleic acid hybridization platform for monitoring biomarkers of public health significance in wastewater.

Nucleic acid amplification tests (NAATs) are exquisitely sensitive and specific, able to accurately and quickly monitor vanishingly small amounts of RNA or DNA in wastewater-based public health surveillance applications. Yet, multiplexing samples and target assays is difficult and the dependence of NAATs on enzymes increases false negative rates with inhibitor-rich environmental samples. The Nanostring nCounter system (NNS) is a nucleic acid hybridization platform with a capability for high multiplexing (upwards of 800 probes) that directly counts RNA or DNA biomarkers without reverse transcri

Cartilage-Penetrating Framework Nucleic Acid Nanoparticles Ameliorate Osteoarthritis by Promoting Drug Delivery and Chondrocyte Uptake.

Osteoarthritis (OA) is a chronic joint disease that causes a gradual deterioration of articular cartilage. A major challenge in OA treatment is the limited penetration and delivery efficiency of drugs to cartilage and chondrocytes due to the rapid clearance of drugs through synovial fluid in joints and the osmotic barrier of the cartilage extracellular matrix (ECM). To address this issue, a novel tetrahedral framework nucleic acid (tFNA)-based nanomedicine delivery system (tFNA-2WL) is first synthesized with excellent cartilage permeability and perfect chondrocyte endocytosis properties. After

M-phenylenediamine-based color carbon dots open a new chapter in the application of nucleic acid dyes.

The performance of nucleic acid dye for staining and labelling nucleic acid molecules directly determines the effectiveness of nucleic acid detection. The widely used traditional nucleic acid dyes pose economic and environmental challenges due to their high price, high toxicity, and long dyeing time. Therefore, we propose the synthesis of m-phenylenediamine carbon dots for use as nucleic acid dye in agarose gel electrophoresis to achieve efficient visualization of DNA and RNA. The prepared carbon dots emit blue, green, yellow, and white fluorescence, diversifying the nucleic acid dye colour se

A new approach for detecting HPV DNA in cervical swabs: comparison of nucleic acid extraction with direct PCR.

BACKGROUND: Almost all cases of cervical cancer are associated with persistent high-risk HPV infection. WHO prioritizes primary HPV testing for cervical cancer screening. Cervical cancer screening programs require the ability to process a large number of samples in a simple and standardized manner and obtain reliable results. The workload, time, and cost associated with the number of samples to be processed encourage the development of alternative methods to the traditional nucleic acid extraction method for population-based screening. In this study, we aimed to compare the performance of a co

Added Value of Nucleic Acid Testing in Blood Banks: A 15-Year Retrospective Study from India.

Background Viral transfusion-transmissible infections (TTIs), hepatitis B (HBV), hepatitis C (HCV), and human immunodeficiency virus (HIV) pose a major risk to blood safety in India, where most blood banks rely on serology alone for TTI screening. Our blood bank at Max Healthcare, New Delhi, India, has used nucleic acid testing (NAT) alongside serology for TTI screening since 2010. Objectives The aim of this study was to evaluate 15 years of real-world blood bank data to evaluate the impact of NAT alongside serology in improving TTI detection and transfusion safety. Methods This single-centre

Designing a locally produced DENV-1 nucleic acid diagnostic for low resource regions with endemic disease.

Using recombinant techniques, to support local production and availability of active diagnostic ingedients (ADIs) in regions with endemic or neglected disease, a reverse transcriptase (RT) fusion construct is reported (Δ3R5-TGP-RT), having the same modular architecture (R5-fluorescent protein-functional enzyme) as reported previously by us. In tandem with the R5-mCherry-Bst this RT construct completes the enzyme set required for RT-LAMP mediated detection of Dengue. The introduction of the thermophilic green fluorescence protein TGP, allowed a heat purification step at 95 °C to be introduced i
