Distinct Metabolomic and Proteomic Signatures of Early Brain Damage Triggered by the Entrance Plateau Versus the Spread-Out Bragg Peak of Proton Radiation.

Proton therapy spares normal tissues better than photon therapy, potentially reducing toxicity while maintaining tumor control. However, challenges remain due to variations in proton dose distribution, particularly at the distal edge of the spread-out Bragg peak (SOBP); for organs at risk, such variation is crucial. We evaluated the biological effects by comparing two positions of the proton profile, the entrance plateau (EP) and SOBP, in a murine model and investigated distinct metabolomic and proteomic signatures. Mice exposed to the EP beam segment (LETd = 0.8 keV/µm) exhibited less weight

Integrating transcriptomics and metabolomics reveals the molecular landscape of sperm maturation driven by regional differentiation in the epididymis of Guizhou-Guiqian semi-fine wool sheep.

Epididymal regionalized differentiation is crucial for sperm maturation. However, little is known about the synergistic remodeling mechanisms of different epididymal segments at the transcriptional and metabolic levels during sexual maturation in ruminants (especially sheep). We investigated the caput, corpus, and cauda epididymidis of pre-pubertal (2-month-old) and post-pubertal (7-month-old) Guizhou-Guiqian semi-fine wool sheep using histology, RNA sequencing, and metabolomics. Post-pubertal tissues exhibited increased luminal diameters, cilia lengths, and abundant cauda spermatozoa. Transcr

(p)ppGpp mediates persister formation in Escherichia coli during glucose to fatty acid shift.

Bacterial persistence contributes to antibiotic failure and recurrent infectious disease, yet the metabolic cues that promote persister formation remain poorly understood. Here we investigated Escherichia coli persistence after nutrient downshifts from glucose to various carbons. Compared to shifts to gluconeogenic carbons (pyruvate, malate, succinate, and fumarate), the glucose-to-fatty acid shift induced exceptionally high persister levels, with cells tolerating ampicillin (56%), carbenicillin (22%), and gentamicin (1%) after 24-h treatment. With an RNA-based biosensor and HPLC quantificatio

Cuproptosis in Endodontics: Potential Mechanisms and Therapeutic Applications.

INTRODUCTION: Cuproptosis is a recently identified copper-dependent form of regulated cell death that is distinct from apoptosis, pyroptosis, and ferroptosis. Its relevance to endodontic disease remains poorly understood despite increasing evidence that copper dyshomeostasis influences oxidative stress, mitochondrial dysfunction, and inflammatory tissue destruction. METHODS: A structured PubMed search was conducted up to April 27, 2026, using terms related to cuproptosis, copper, endodontics, pulpitis, and apical periodontitis. Original studies, reviews, and mechanistic reports relevant to th

Acute-on-Chronic Liver Failure: An Eroded Cliff Hit by a Storm-A Narrative Review.

Acute-on-chronic liver failure (ACLF) is a rapidly progressing and highly lethal clinical syndrome characterized by multiorgan failure, driven primarily by a severe systemic inflammatory response. The pathophysiological cascade, triggered by a "cytokine storm," subsequently evolves into profound immune paralysis. This phenomenon is driven by the dysfunction of monocytes, neutrophils, and other immune cells, compounded by their impaired cellular energetics resulting from a metabolic shift toward less efficient energy-yielding mechanisms, mainly aerobic glycolysis, with the pentose phosphate pat

Computational discovery and prioritization of ribose-5-phosphate isomerase A as a mitochondrial target for reversing senescence in mesenchymal stem cells.

Age-related osteoporosis is increasingly recognized as a disease of stem cell dysfunction, hallmarked by the skewed lineage commitment of mesenchymal stem cells (MSCs) away from osteogenesis towards adipogenesis. This functional decline is closely linked to mitochondrial dysregulation. However, the upstream metabolic regulators that orchestrate this detrimental cell-fate switch, particularly the key nodes at the interface of mitochondrial function and cellular senescence, remain largely unknown, hindering the development of targeted therapies. To address this challenge, we implemented a bioinf

Effects of Chinese yam and its peel polysaccharides on blood glucose and lipids in type 2 diabetic mice.

Chinese yam (Dioscorea opposita Thunb.) is a traditional medicinal and edible homologous material, with polysaccharides being its primary bioactive components. In this study, two polysaccharides (CYP and CYPP) were isolated and purified from Chinese yam (cv. Tiegun) and its peel, and their anti-diabetic activities were investigated. Structural characterization revealed that the weight-average molecular weights of CYP and CYPP were 74,746 Da and 118,083 Da, respectively. Monosaccharide composition analysis showed that both polysaccharides had similar compositions, primarily consisting of mannos

Targeting serine metabolism vulnerability in omipalisib-resistant acute myeloid leukemia with phosphoglycerate dehydrogenase inhibitors.

Acute myeloid leukemia (AML) is the most common acute leukemia that primarily affects older adults. Dysregulated PI3K/AKT/mTOR signaling pathway is a common abnormality in AML. Our previous study demonstrated the excellent cytotoxicity of dual PI3K/mTOR inhibitor omipalisib against AML cells. However, its clinical application remains challenging because of potential resistance mechanisms following kinase inhibitor administration. In this study, OCI-AML3-OR, an OCI-AML3 subline that is resistant to omipalisib, was established. Transcriptomics analysis revealed that the significant differentiall

Assessment of the metabolome across the glucose tolerance spectrum in adults with cystic fibrosis hospitalized for pulmonary exacerbations.

BACKGROUND: Cystic fibrosis-related diabetes (CFRD) is one of the most common co-morbidities among adults with cystic fibrosis (CF), yet the mechanisms determining its development remain unclear. This study explored the underlying biological mechanisms associated with CFRD development using plasma high-resolution metabolomics (HRM). METHODS: This cross-sectional study included 52 adults with CF hospitalized for a pulmonary exacerbation. Participants were categorized into three glucose tolerance groups: normal glucose tolerance (n = 26), pre-diabetes (n = 9), and CFRD (n = 17), based on data a

Global transcriptome characterization of peripheral blood mononuclear cells in individuals with chronic HIV infection.

BACKGROUND: Acquired Immune Deficiency Syndrome (AIDS), resulting from Human Immunodeficiency Virus (HIV) infection, is one of the most severe infectious diseases worldwide. The current state of prevention and control remains critical. Recent studies have increasingly highlighted the significant role of cellular metabolism in regulating immune responses and managing infections. However, whether distinct immunometabolic profiles exist among different groups infected with HIV remains to be investigated. In this study, we employed RNA-seq technology to explore the differential characterization of

Label-free isolation of lipid-rich Saccharomyces cerevisiae mutant by high-throughput flow-mode Raman-activated cell sorting and multi-omics analysis for uncovering the mechanism of enhanced lipid accumulation.

BACKGROUND: Palmitoleic acid, a valuable functional fatty acid, is notably scarce in traditional oil crops, with the exception of certain wild plants such as macadamia nuts and sea buckthorn. Recently, the lipid from Saccharomyces cerevisiae was found to contain approximately 50% palmitoleic acid. Consequently, S. cerevisiae has the potential to sustainably produce palmitoleic acid through fermentation, provided that the issue of promoting its lipid content is addressed. RESULTS: In this work, based on the previously isolated oleaginous wild strain of S. cerevisiae, the mutagenesis by zeocin

Glucose priming effect on microbial intercellular metabolic flux diversity in a marine intertidal sediment.

Microbial communities inhabiting intertidal sediment show diverse metabolisms to adapt to hydrodynamic conditions. However, the priming effect of exogenous organic matter on microbial metabolic fluxes remains poorly understood. Here we investigated microbial intercellular activities in surface sandy intertidal sediment (0-5 cm depth, from the Nanhui tidal flat, East China Sea) under anoxic conditions by incubating with position-specific 13C labelled glucose isotopologues. Elevated production of 13C-CO2 and phospholipid-derived fatty acids (PLFAs) was observed after 96-hour incubation, suggesti

Integrative Constraint-Based Modeling and Proteomics Uncover Astrocytic Metabolic Adaptations to the Post-TBI Microenvironment.

Traumatic brain injury (TBI) is a major neurological condition affecting millions of individuals each year. Mild TBI (mTBI) manifests differently, with some individuals experiencing persistent, debilitating symptoms while others recover more rapidly. Despite its classification as "mild," mTBI leads to both short- and long-term neurological effects, many of which occur due to functional changes in the brain. TBI-induced environmental changes within the brain play a critical role in shaping these functional outcomes. The importance of astrocytes in maintaining central nervous system (CNS) homeos

Association of transketolase polymorphisms with diabetic polyneuropathy in the general population: The KORA F4 study.

AIMS: We recently reported that genetic variability in the TKT gene encoding transketolase, a key enzyme in the pentose phosphate pathway, is associated with measures of diabetic sensorimotor polyneuropathy (DSPN) in recent-onset diabetes. Here, we aimed to substantiate these findings in a population-based KORA F4 study. MATERIALS AND METHODS: In this cross-sectional study, we assessed seven single nucleotide polymorphisms (SNPs) in the transketolase gene in 952 participants from the KORA F4 study with normal glucose tolerance (NGT; n = 394), prediabetes (n = 411), and type 2 diabetes (n = 14
