Potential structure induced effects of the hairpin motif in RNA to form the phase separated droplet like associate in different solution condition.

Reversible associations of RNAs among themselves, or with RNA binding proteins through the process of phase separation, are found to be important in many different cellular contexts, including cellular responses to stress, gene regulation, development, and disease. Short RNA repeat sequences, which are mostly linked with many repeat expansion neurodegenerative diseases, are found to undergo phase separation and yield protein-free biomolecular condensates in vitro and in cells. However, the physicochemical principles governing phase separation of RNAs considering both sequence and structural as

Competitive virus and host RNAs: the interplay of a hidden virus and host interaction.

During virus infection, viral RNAs and mRNAs function as blueprints for viral protein synthesis and possibly as pathogen-associated molecular patterns (PAMPs) in innate immunity. Here, considering recent research progress in microRNAs (miRNAs) and competitive endogenous RNAs (ceRNAs), we speculate that viral RNAs act as sponges and can sequester endogenous miRNAs within infected cells, thus cross-regulating the stability and translational efficiency of host mRNAs with shared miRNA response elements. This cross-talk and these reciprocal interactions between viral RNAs and host mRNAs are termed

A Recurrent 3'UTR Variant in CAPN3 Highlights the Diagnostic Impact of Non-Coding Variants in Limb-Girdle Muscular Dystrophies.

OBJECTIVE: A substantial proportion of individuals with suspected genetic conditions, including neuromuscular disorders, lack a definitive molecular diagnosis. Non-coding variants, particularly those in the 3' untranslated region (3´UTR), which regulate mRNA stability, translation, and localization, may underlie part of this gap. We investigated the clinical, genetic, and functional relevance of a novel 3'UTR variant in CAPN3 in individuals with persistent hyperCKemia, most of whom also presented with proximal muscle weakness. METHODS: Clinical assessment, including muscle biopsy and muscle m…

RNAi: the next wave of green agriculture, challenges, and bridging the translational gap.

Agriculture must achieve substantial yield gains while drastically reducing its environmental footprint to meet the food demands of a growing global population by 2050. Current crop losses to pests and pathogens account for 20-40% of annual production, a challenge predominantly addressed by chemical pesticides, which impose high ecological and health costs. RNA interference (RNAi) has emerged as a transformative, species-specific solution, demonstrating high efficacy (> 90%) against susceptible target pests under controlled laboratory conditions, though responsiveness varies considerably acros

Co-transcriptional R-loops-mediated epigenetic regulation drives growth retardation and docetaxel chemosensitivity enhancement in advanced prostate cancer.

R-loops are prevalent three-stranded nucleic acid structures, comprising a DNA-RNA hybrid and a displaced single-stranded DNA, that frequently form during transcription and may be attributed to genomic stability and gene expression regulation. It was recently discovered that RNA modification contributes to maintain the stability of R-loops such as N6-methyladenosine (m6A). Yet, m6A-modified R-loops in regulating gene transcription remains poorly understood. Here, we demonstrated that insulin-like growth factor 2 mRNA-binding proteins (IGF2BPs) recognize R-loops in an m6A-dependent way. Consequ

Multilayer Timing of Cardiac Circadian Regulation Informs Prevention and Treatment of Cardiovascular Disease.

BACKGROUND: Cardiovascular diseases remain the leading cause of death and economic burden worldwide. Increasing evidence indicates that sleep disturbance and circadian rhythm disruption are major risk drivers for hypertension, coronary artery disease, heart failure, and arrhythmia. Although the classical transcription-translation feedback loop (TTFL) model explains the basic mechanism of rhythm generation, increasing evidence suggests that the heart-an organ with high metabolic demand-maintains circadian stability through coordinated transcriptional, translational, and post-translational regul

The osteogenic regulator THOC5-dependent post-transcriptional control maintains vascular smooth muscle cell homeostasis against chronic kidney disease related vascular calcification.

INTRODUCTION: Osteogenic transdifferentiation of vascular smooth muscle cells (VSMCs) plays a fundamental role in chronic kidney disease (CKD)-associated vascular calcification (VC). Despite its significance, the post-transcriptional control mechanisms driving VSMC osteogenic switching remain poorly understood. Here, we sought to elucidate the THOC5-dependent post-transcriptional regulatory mechanisms that control VSMC transdifferentiation and contribute to CKD-associated VC. METHODS: Von Kossa staining and immunohistochemistry were used to detect calcification and THOC5 expression in arteria
