Bone Marrow Transplantation in Mice to Study the Role of Hematopoietic Cells in Atherosclerosis.

Hematopoietic stem cell transplantation or bone marrow transplantation is a common approach to reconstitute the immune system of mice that have been subjected to marrow-ablative doses of radiation. This method can be used in the field of atherosclerosis to assess the contribution of hematopoietic cells of a desired genotype to disease pathogenesis. The engraftment of atherosclerosis-prone mice with donor cells that contain genetic alterations in cells of the innate or adaptive immune system has been invaluable to define the role of multiple gene products in atherosclerosis. Here, we describe t

Xeroderma pigmentosa: three new cases with an in depth review of the genetic and clinical characteristics of the disease.

Xeroderma pigmentosum (XP) is a rare autosomal recessive disorder characterized by hypersensitivity of the skin and eyes to UV-radiation as a result of a defect in one of eight genes. Seven genes (XPA-XPG) have a defect in Nucletoide Excision Repair (NER), while the eighth gene XPV has a defect in polymerase η, which is responsible for replication of UV-damaged DNA to produce corrected daughter strands. We present the varied clinical courses of three African-American female patients with XP. Additionally, we present a review of the literature that focuses on the various clinical manifestations

Cystic fibrosis.

Cystic fibrosis is an autosomal recessive genetic disorder that affects ion transport in exocrine glands. Inadequate ion transport causes dehydration and the production of thick secretions in organs such as the lungs, sinuses, pancreas, intestines, hepatobiliary tree, and vas deferens. Although cystic fibrosis usually is diagnosed through a sweat chloride test, medical imaging is used to monitor pathologic changes caused by the disease. This article discusses the respiratory and nonrespiratory manifestations of cystic fibrosis and describes the diagnosis and treatment of the disease. Because p

Epidemiology of brain tumors.

Brain tumors seemed to have increased in incidence over the past 30 years, but the rise probably results from use of new neuroimaging techniques. Treatments have not improved prognosis for rapidly fatal brain tumors. Established brain tumor risk factors (exposure to ionizing radiation, rare mutations of penetrant genes, and familial history) explain only a small proportion of brain tumors, and only one of these potentially is modifiable. Genetic and environmental characteristics likely play a role in familial aggregation of glioma and these factors are not identified. New concepts in brain tum

Stroke in children.

Childhood stroke, although similar to adult stroke, is characterized by congenital and genetic causes. The evaluation and treatment of the child with stroke requires special considerations. Currently, drug therapy is untested and as a result, therapeutic interventions are problematic. Platelet antagonists are rational prophylactically; warfarin probably has a role in preventing cardiogenic embolus in the older child. However, chronic anticoagulation in the toddler is dangerous due to frequent trauma and is therefore relatively contraindicated. Vascular malformations are surgically repaired but

Influence of Ethnicities and Skin Color Variations in Different Populations: A Review.

BACKGROUND: In the world scientific tradition, skin color is the primary physical characteristic used to divide humans into groups. Human skin has a wide range of tones and colors, which can be seen in a wide range of demographic populations. Many factors influence the color of people's skin, but the pigment melanin is by far the most important. Melanin is produced by cells called melanocytes in the skin and is the primary determinant of skin color in people with darker skin. Indeed, >150 genes have now been identified as having a direct or indirect effect on skin color. Vitamin D has recently

The importance of early diagnosis and treatment of actinic keratosis.

Chronic, long-term sun exposure results in genetic changes in epidermal keratinocytes and the development of various skin lesions ranging from actinic keratosis (AK) to skin cancer. AK lesions may first appear as rough, scaly spots on sun-exposed skin, and, although most individual AK lesions do not become invasive cancers, the majority of invasive squamous cell carcinomas originate from AK. Genetic analysis demonstrates that ultraviolet radiation-induced mutations and changes in gene expression are present in squamous cell carcinoma, AK, and clinically normal-appearing perilesional sun-expose

Malignant gliomas: new translational therapies.

Malignant gliomas are the most common primary brain tumors in adults and carry a dismal prognosis. Despite aggressive therapy with maximal safe surgical resection, radiation and chemotherapy, these tumors invariably are refractory to or become resistant to treatment and recur. Gliomas are highly infiltrative cancers and display remarkable genetic heterogeneity making them challenging to treat. Recent progress has been made in understanding the molecular and genetic composition of these tumors and from this, promising new targets for therapy have emerged. In particular, anti-angiogenesis therap

Cytogenetic investigation of subjects professionally exposed to radiofrequency radiation.

Nowadays, virtually everybody is exposed to radiofrequency radiation (RFR) from mobile phone base station antennas or other sources. At least according to some scientists, this exposure can have detrimental health effects. We investigated cytogenetic effects in peripheral blood lymphocytes from subjects who were professionally exposed to mobile phone electromagnetic fields in an attempt to demonstrate possible RFR-induced genetic effects. These subjects can be considered well suited for this purpose as their RFR exposure is 'normal' though rather high, and definitely higher than that of the 'g

[Selective expression of a mutation of the rad 201 radiosensitivity gene in Drosophila embryogenesis].

Embryonic sensitivity to the lethal effect of gamma-irradiation was compared in a radiosensitive mutant strain rad201G1 and a control radioresistant strain of Drosophila during the first 9 h of embryonic development. Embryonic radiosensitivity was analyzed with regard to the physiological age of embryos at the time of exposure to radiation. In the wild-type embryos, age-dependent variations of radiosensitivity were shown to be closely associated with genetically determined alterations in mitotic cycle structure, synchronism, and duration of separate phases. The rad201 gene mutation, which disr

Epidemiologic aspects of early menopause and ovarian cancer.

The concept of ovarian cancer as a consequence of hypergonadotropic hypogonadism implies that risk factors for early menopause may also be risk factors for ovarian cancer. Gonadal radiation and the oophoritis of mumps are possible examples of environmental risk factors for early menopause and ovarian cancer. Family history is a strong determinant for both early menopause and ovarian cancer, implicating genetic factors in these disorders. Case-control data suggest that galactose consumption through the ingestion of high-lactose dairy foods may be a dietary risk factor and that galactose metabol

A Pound of Cure Requires An Ounce (or More) of Prevention: Survivorship and Complications of Therapy for Hematologic Malignancies.

Patients treated for a hematologic malignancy are at risk for treatment-related complications. As the goal of therapy is frequently curative, treatments are especially intensive and long-term toxicity is common. Chemotherapy and radiation are associated with increased risk for cardiac and pulmonary disease, endocrine disorders, infertility, sexual dysfunction, second cancers, and psychosocial distress. The risk for each complication is dictated by patient characteristics including age, co-morbidities, and genetic predispositions, as well as the specifics of therapy. Survivors of pediatric canc

Multidisciplinary approach of early breast cancer: the biology applied to radiation oncology.

Early breast cancer treatment is based on a multimodality approach with the application of clinical and histological prognostic factors to determine locoregional and systemic treatments. The entire scientific community is strongly involved in the management of this disease: radiologists for screening and early diagnosis, gynecologists, surgical oncologists and radiation oncologists for locoregional treatment, pathologists and biologists for personalized characterization, genetic counselors for BRCA mutation history and medical oncologists for systemic therapies.Recently, new biological tools h

Gastrointestinal toxicity associated to radiation therapy.

Radiation therapy in combination with other treatments, such as surgery and chemotherapy, increases locoregional control and survival in patients with thoracic, abdominal and pelvic malignancies. Nevertheless, significant clinical toxicity with combined treatments may be seen in these patients. With the advent of tridimensional conformal radiotherapy (3D-CRT), dose-volume histograms (DVH) can be generated to assess the dose received by the organs at risk. The possible relationship between these parameters and clinical, anatomical and, more recently, genetic factors has to be considered. Treatm

Xeroderma pigmentosum: a glimpse into nucleotide excision repair, genetic instability, and cancer.

Xeroderma pigmentosum (XP) is a rare DNA repair disorder characterized by extreme sensitivity to sunlight and severe predisposition to UV-induced skin cancer. Seven genes, ranging from XPA to XPG, are defective in XP. These genes are important components of the nucleotide excision repair (NER) system, which removes DNA damage induced by solar radiation, thereby preventing genome instability and carcinogenesis. In addition, XPV patients are defective in a translesion synthesis activity specialized in bypassing UV-induced lesions, and share symptoms with other XP patients. This review will focus

The inherited basis of human radiosensitivity.

Certain individuals cannot tolerate 'conventional' doses of radiation therapy. This is known to be true of patients with ataxia-telangiectasia and ligase IV deficiency. Although in vitro testing may not correlate completely with clinical radiosensitivity, fibroblasts and lymphoblasts from patients with both of these disorders have been clearly shown to be radiosensitive. Using a colony survival assay (CSA) to test lymphoblastoid cells after irradiation with 1 Gy, a variety of other genetic disorders have been identified as strong candidates for clinical radiosensitivity, such as Nijmegen break

Renal cell carcinoma: a rare source of cauda equina metastasis. Case report.

The authors present the case of a patient in whom intradural metastasis from renal cell carcinoma spread to the cauda equina. To the authors' knowledge, this is only the second report of its kind. This male patient had undergone nephrectomy for the treatment of renal cell carcinoma for 5 years and was diagnosed as having metastatic lung disease 1 year prior to admission. The patient presented with lower back pain that radiated to both legs, but he exhibited no sensorimotor deficits. The majority of cauda equina tumors are primary tumors, and metastases are very rare. The literature is reviewed

[Changes in cranial size and configuration in infants].

Abnormalities of cranial size and configuration include macrocephaly, microcephaly and dysmorphic skull. Hydrocephalus, mega-encephalus, abnormal growth of skull bones and chronic subdural and epidural collections are among the most important causes of macrocephaly. Primary microcephaly may be due to deficient neuronal proliferation or abnormalities in neural tube segmentation or cellular migration. Secondary microcephaly may be caused by prenatal, perinatal or early postnatal insults like infections, drugs, radiation, hypoxia, among others. Abnormal cranial forms may be sporadic or secondary
