Functional and structural diversification of the Anguimorpha lizard venom system.

Venom has only been recently discovered to be a basal trait of the Anguimorpha lizards. Consequently, very little is known about the timings of toxin recruitment events, venom protein molecular evolution, or even the relative physical diversifications of the venom system itself. A multidisciplinary approach was used to examine the evolution across the full taxonomical range of this ∼130 million-year-old clade. Analysis of cDNA libraries revealed complex venom transcriptomes. Most notably, three new cardioactive peptide toxin types were discovered (celestoxin, cholecystokinin, and YY peptides).

Interferon-gamma:interleukin 4 ratios and associated type 1 cytokine expression in juvenile rheumatoid arthritis synovial tissue.

OBJECTIVE: To compare synovial tissue cytokine mRNA expression between patients with juvenile rheumatoid arthritis (JRA) and a heterogeneous group of non-autoimmune arthropathies (controls) with respect to type 1/type 2 balance. METHODS: Thirty-five JRA (average 9.1 years' disease duration) and 13 control synovial tissues were studied. As a measure of the type 1/type 2 cytokine balance in a subset of the JRA and control tissues, interferon-gamma (IFN-gamma) and interleukin 4 (IL-4) mRNA levels were measured by competitive fragment reverse transcription-polymerase chain reaction. To quantitate

Nitric oxide and cyclic guanosine monophosphate stimulate apoptosis via activation of the Fas-FasL pathway.

BACKGROUND: Inappropriately exaggerated response of pulmonary vascular cells to inflammatory mediators may be one mechanism that leads to acute (or adult) respiratory distress syndrome. Nitric oxide (NO) is induced following such exaggerated responses and may have a variety of biological effects, including induction of apoptosis. The mechanism by which NO causes apoptosis is unknown; however, Fas (CD95) and Fas ligand (FasL) (CD95L) have been implicated. We hypothesized that NO-induced apoptosis in pulmonary vascular smooth muscle cells is mediated through a Fas-FasL pathway. MATERIALS AND ME

Postnatal regulation of limb proprioception by muscle-derived neurotrophin-3.

To investigate the effects of neurotrophin-3 (NT-3) on postnatal proprioceptive neurons and their targets, transgenic mice were generated that use the myosin light chain 1 (mlc) promoter to overexpress NT-3 in skeletal muscle. Ribonuclease protection assays revealed that NT-3 overexpression in hindlimb skeletal muscle began at embryonic day 14 (E14) and continued throughout adulthood. Overexpression of NT-3 during late embryogenesis resulted in increased numbers of large sensory and small fusimotor axons. Within a week of birth, mlc/NT-3 mice retract their limbs to the torso when lifted by the

Isolation and serum-free culture of epithelial cells derived from epithelial rests of Malassez in human periodontal ligament.

To study biological character and function of epithelial rests of Malassez (ERM) in human periodontal ligament, we have developed a serum-free culture system of epithelial cells (ME) derived from ERM. The mitogenic effects of fibroblast growth factor (FGF)-1, FGF-2, and FGF-7/keratinocyte growth factor (KGF) on ME, human periodontal ligament-derived fibroblasts (PLF), human oral epithelial cells (OE), and human submandibular gland-derived epithelial cells (SGE) were investigated under a serum-free culture condition. FGF-1 and FGF-7/KGF stimulated the growth of both ME and SGE but FGF-2 had no

Effects of RU486 on estrogen, progesterone, oxytocin, and their receptors in the rat uterus during late gestation.

Oxytocin (OT) and its receptor (OTR) are synthesized in the endometrium and myometrium of the pregnant rat during late gestation. Both are regulated by estrogen and progesterone (P4), and tissue concentrations of both increase markedly before parturition. The P4 antagonist RU486 will induce parturition in the rat. The purpose of the present studies was to investigate changes in OT and OTR messenger RNA (mRNA) and peptide synthesis within the pregnant rat uterus during RU486-induced parturition. Pregnant rats were given a single injection of RU486 (2.5 mg/rat in oil) on day 15 of pregnancy (nor

Regulation of glucagon and glucagon-like peptide-1 receptor messenger ribonucleic acid expression in cultured rat pancreatic islets by glucose, cyclic adenosine 3',5'-monophosphate, and glucocorticoids.

Glucagon and glucagon-like peptide-1 (GLP-1) are important regulators of glucose homeostasis, and both are involved in regulating pancreatic islet hormone secretion. Since the sensitivity of the endocrine pancreas to regulatory hormones can be influenced by their receptor number, we have examined the regulation of glucagon receptor and GLP-1 receptor messenger RNA (mRNA) expression in cultured rat pancreatic islets by various factors, including glucose, cAMP, and glucocorticoids. By ribonuclease protection assay we have demonstrated the expression of both glucagon and GLP-1 receptor mRNA in cu

SDJ, a pollen-expressed type III J-protein in Prunus, directly binds S-RNase and promotes recruitment of SLFL6 to an S-RNase-associated complex.

In Prunus, self-incompatibility (SI) is controlled by S-RNases and pollen-expressed F-box proteins, whereas the molecular processes governing S-RNase regulation in pollen remain incompletely understood. Here, we characterized PavSDJ, a novel pollen protein from sweet cherry (Prunus avium), as a candidate factor involved in pollen-side S-RNase-associated processes. Sequence and structural analyses identified PavSDJ as a type III J-protein. Phylogenetic analyses placed PavSDJ within a distinct SDJ-like sublineage of the type III J-protein group, separate from a closely related sister lineage. Co

Mapping the Reactivity of Two Pharmacologically Active Organopalladium Compounds toward the Model Protein RNase A Using a Dual ESI MS and X-ray Diffraction Investigative Approach.

Organopalladium compounds are a new class of metal-based drugs with significant yet largely unexplored therapeutic potential. Previous studies have suggested that these compounds act as prodrugs that can strongly interact with protein targets. However, the underlying protein metalation processes have scarcely been investigated to date. To gain a deeper insight into this issue, we analyzed the interactions of two representative organopalladium compounds with the model protein RNase A using a strategy that combined electrospray ionization mass spectrometry (ESI MS) and X-ray crystallography. Serum RNASE1 as a biomarker for disease activity and lupus nephritis in systemic lupus erythematosus.

INTRODUCTION: Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disease characterized by autoantibody production and immune complex-mediated organ damage, frequently involving the kidneys as lupus nephritis (LN). However, reliable blood-based biomarkers for disease activity and renal involvement remain limited. METHODS: We performed serum proteomic profiling in newly diagnosed SLE patients and integrated these data with renal transcriptomic datasets from SLE mouse models to identify candidate biomarkers. Associations between serum RNASE1 levels and clinical parameters were eval

MCPIP1 modulates host antifungal immunity and exacerbates lethal Candida albicans infection.

Candida albicans (C. albicans) is the leading cause of systemic candidiasis in immunocompromised individuals and is associated with substantial mortality. As current antifungal therapies are limited by suboptimal efficacy and the emergence of resistance, improved understanding of host-pathogen interactions that govern antifungal immunity is essential for identifying new therapeutic strategies. Here, we identify MCPIP1 as a host factor that negatively regulates antifungal immune responses during lethal C. albicans infection. Integrative bioinformatic analyses of transcriptomic profiles from hum

Conserved structural topologies in RNase A-like and trypsin-like serine proteases: a sequence-based folding analysis.

BACKGROUND: Protein folding is a complex process in which amino acid sequences encode the information required for a polypeptide chain to fold into its functional three-dimensional (3D) structure. Many proteins share common substructures and recurring secondary structure elements that contribute to similar 3D folding patterns, even across different protein families. This study examines two distinct groups of proteins, the RNase A-like fold and the trypsin-like serine protease fold, classified by SCOPe. These proteins share only some substructures that contribute to their folding cores. Despite

Intestinal taurine acts as a novel immunometabolic modulator of IBD by degrading redundant mitochondrial RNA.

Anti-tumor necrosis factor (TNF) therapy for inflammatory bowel disease (IBD) is hampered by issues of nonresponse and resistance, highlighting the urgent need for alternative or complementary treatments. Our study revealed significant upregulation of taurine in the intestinal tissues of IBD patients, which was inversely related to the severity of the disease. A key discovery was that TNF directly induced taurine synthesis in intestinal epithelial cells and increased the production of angiogenin, a nuclease that degrades mitochondrial RNA, which is known to amplify inflammatory responses. Sinusoidal Hepatic Growth Pattern of Metastatic DICER1-Mutated Urethral Sarcoma Mimicking Hepatocellular Carcinoma.

Although liver metastases from solid tumors are common and typically present as discrete masses, a sinusoidal pattern of hepatic involvement is rare, especially in sarcomas. We report a case of DICER1-mutated urethral sarcoma exhibiting this unusual metastatic pattern, with features mimicking hepatocellular carcinoma (HCC). Immunohistochemical analysis revealed focal nodular hyperplasia-like changes rather than HCC, attributed to tumor cell infiltration of hepatic sinusoids and consequent alterations in blood flow. Recognizing this distinct metastatic pattern is essential, as it can be overloo

Sinonasal DICER1‑mutated embryonal-like (botryoid-like) rhabdomyosarcoma in an adult: report of the first case.

The DICER1 gene, essential for microRNA biogenesis and posttranscriptional gene regulation, has been implicated in a variety of benign and malignant neoplasms, particularly within the context of the DICER1-related tumor predisposition syndrome. While DICER1-associated rhabdomyosarcomas (RMS) are predominantly documented in the genitourinary tract, we present the first case of a DICER1-mutated embryonal-like (botryoid-like) RMS of the nasal fossa. A 63-year-old woman without relevant family history presented with nasal obstruction, headaches, and epistaxis and underwent resection of a polypoid

Pandemic driven innovation: A pilot evaluation of an alternative respiratory pathogen collection device.

BACKGROUND: The nasopharyngeal swab is the gold standard collection method for COVID-19, but is invasive and painful, subsequently resulting in poor patient acceptance. This investigation explores the process of developing and validating an alternative respiratory pathogen collection device that relies on a nasopharyngeal irrigation mechanic. The primary objective was to determine if sufficient pathological sampling can be achieved by mechanism of nasopharyngeal irrigation that is proportionate to the nasopharyngeal swab method. METHODS: The study device was designed using Shapr3D modeling so
