Understanding vulnerability for depression from a cognitive neuroscience perspective: A reappraisal of attentional factors and a new conceptual framework.

We propose a framework to understand increases in vulnerability for depression after recurrent episodes that links attention processes and schema activation to negative mood states, by integrating cognitive and neurobiological findings. Depression is characterized by a mood-congruent attentional bias at later stages of information processing. The basic idea of our framework is that decreased activity in prefrontal areas, mediated by the serotonin metabolism which the HPA axis controls, is associated with an impaired attenuation of subcortical regions, resulting in prolonged activation of the a

DNA damage and repair in translational oncology: an overview.

Unknown to early investigators, DNA damage and repair has been a major focus of anticancer therapy from the beginning of clinical oncology. From the early days of using x-irradiation, to the development of nitrogen mustard analogs, to today's more sophisticated approaches, DNA damage and repair has strongly impacted our ability to successfully treat human malignancy. This area of basic, translational, and clinical science is very broad. The traditional focus of DNA damage and repair has been on diseases such as Xeroderma pigmentosum, and attempting to understand the basic molecular mechanisms

Knowledge Translation of Pharmacogenomics for Pain Management in Patients With Sickle Cell Disease-A Qualitative Study.

BACKGROUND: Health care providers (HCPs) currently prescribe analgesics based on clinical experience, placing Black Americans with sickle cell disease (SCD) at risk for poorly controlled pain, adverse drug effects, and bias. Pharmacogenomics could guide individualized pain management, but many HCPs feel unprepared to use this information in practice. OBJECTIVE: This qualitative research study explored the needs of pediatric hematology HCPs to integrate pharmacogenomics into care for SCD. DESIGN/SETTING: Semistructured interviews and focus groups were conducted at a large urban quaternary-car

Venous Thromboembolism Research Priorities: A Scientific Statement From the American Heart Association and the International Society on Thrombosis and Haemostasis.

Venous thromboembolism is a major cause of morbidity and mortality. The impact of the US Surgeon General's The Surgeon General's Call to Action to Prevent Deep Vein Thrombosis and Pulmonary Embolism in 2008 has been lower than expected given the public health impact of this disease. This scientific statement highlights future research priorities in venous thromboembolism, developed by experts and a crowdsourcing survey across 16 scientific organizations. At the fundamental research level (T0), researchers need to identify pathobiological causative mechanisms for the 50% of patients with unprov

Using knowledge translation as a framework for the design of a research protocol.

Knowledge translation has been defined as the synthesis, dissemination, exchange and ethically sound application of knowledge to improve health, resulting in a stronger health-care system. Using KT activities to aid in the adoption of evidence into practice can address current health-care challenges such as increasing organizational practice standards, alleviating the risk for adverse events and meeting practitioner needs for evidence at the bedside. Two general forms of KT have been identified. These being integrated KT and end-of-grant KT. Integrated KT involves the knowledge users in the re

Cancer nanomedicine and RNA/Lipid nanoparticle platforms in oncology: developmental evolution, translational barriers, and precision-therapy opportunities.

Nanomedicine has become an important translational platform in oncology by enabling altered pharmacokinetics, controlled exposure, reduced systemic toxicity, intracellular delivery of nucleic acids, and improved handling of complex therapeutic payloads. Early cancer nanomedicines, including pegylated liposomal cytotoxics and albumin-bound drug products, demonstrated that nanoscale formulation design could improve the clinical utility of established anticancer agents. More recently, lipid nanoparticles, RNA therapeutics, individualized neoantigen vaccines, gene-regulatory approaches, and immune

Basic and Translational Research Advances in NSCLC.

Treatments for lung cancer have advanced significantly over the last 2 decades, improving survival and long-term outcomes. However, lung cancer mortality remains high, and cures for metastatic disease have remained elusive. Here, we outline recent advances in our understanding of lung cancer biology and areas of interest for future therapeutic development, with a focus on resistance mechanisms, tumor heterogeneity, plasticity, minimal residual disease, and the tumor microenvironment. A comprehensive understanding of how these areas influence tumor biology will be essential for the development

Next-generation intranasal influenza vaccines: mechanisms, platforms, and translational progress.

Seasonal influenza vaccination remains the most effective strategy for reducing influenza burden and preventing severe disease. Despite decades of vaccine development, the seasonal influenza vaccine is administered intramuscularly and provides suboptimal and highly variable effectiveness depending on host factors, pre-existing immunity, and antigenic match between vaccine and circulating strains. Recent advances in vaccine development have highlighted the potential of intranasal vaccine delivery as a strategy to increase protection against influenza virus infection by inducing local and system

Regulatory T cell therapies: biological foundations, engineering strategies, and clinical translation.

Regulatory T cell (Treg) therapy has emerged as a promising strategy to control pathological immune responses in autoimmunity, graft-versus-host disease, and solid organ transplantation. Most clinical studies to date have relied on autologous Tregs expanded ex vivo, an approach that has demonstrated safety and feasibility but remains limited by variable cell quality, restricted scalability, and complex manufacturing requirements. To address these constraints, multiple alternative strategies are being developed, including the induction of regulatory phenotypes in conventional T cells, the engin

Murine models of pulmonary fibrosis: mechanisms, limitations and translational insights.

Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive, irreversible interstitial lung disease characterised by excessive collagen deposition, aberrant tissue remodelling, and impaired gas exchange, ultimately leading to respiratory failure and death. IPF carries a poor prognosis, with limited therapeutic options; currently approved treatments slow disease progression but do not reverse established fibrosis, and lung transplantation remains the only definitive therapy. Moreover, the prevalence and incidence of IPF continue to rise worldwide. These challenges underscore the urgent need t

Drug resistance mechanisms of immunotherapy and translational strategies for reversing resistance for hepatocellular carcinoma: from bench to bedside.

Hepatocellular carcinoma (HCC) is the most common primary malignancy of the liver and remains a major therapeutic challenge. In recent years, immune checkpoint inhibitors (ICIs), represented by PD-1/PD-L1 and CTLA-4 inhibitors, have revolutionized the field of HCC treatment and become the cornerstone of standard immunotherapy regimens, drastically altering the treatment landscape for advanced and unresectable HCC. However, primary and/or acquired drug resistance remains the leading cause of treatment failure, severely limiting the long-term clinical benefits of immunotherapy for HCC patients.

In Vitro-In Vivo Correlation (IVIVC) in Transdermal Drug Delivery Systems: Models, Regulatory Perspectives and Translational Challenges.

Transdermal drug delivery systems (TDDS) represent a clinically established yet mechanistically complex platform for systemic drug administration, offering advantages such as avoidance of hepatic first-pass metabolism, sustained drug input and improved patient adherence. However, their development and regulatory evaluation remain challenging due to the physiological variability of the skin barrier, site-dependent absorption and formulation-skin interactions. In vitro-in vivo correlation (IVIVC) has therefore emerged as a critical translational framework for linking in vitro permeation characte

Ferroptosis in diabetic retinopathy: from pathogenic mechanisms to translational prospects.

Diabetic retinopathy (DR) is a common microvascular complication of diabetes. Despite ongoing revisions in the prevention and treatment of DR, optimal treatment strategies have yet to be established. Revealing the pathological changes and molecular mechanisms of DR is the cornerstone for exploring new therapeutic strategies. Ferroptosis, a new type of programmed cell death proposed in recent years, is characterized mainly by reactive oxygen species and iron-mediated lipid peroxidation. As studies progress, growing evidence has highlighted the involvement of ferroptosis, a newly identified prog

[Artificial intelligence-driven assessment of tertiary lymphoid structures in breast cancer: From microscopic analysis to clinical translation].

Tertiary lymphoid structures (TLSs) are key regulatory components of the breast cancer immune microenvironment. Their microscopic composition (specific immune cell subsets, spatial organization) and functional status (maturity) directly determine their performance in coordinating the anti-tumor immune responses, serving as the microscopic foundation for understanding the efficacy of immunotherapy. However, the high heterogeneity and dynamic evolution of the tumor microenvironment pose significant limitations for immunotherapeutic efficacy prediction models based on traditional histopathologica

Carbonic Anhydrase I and II as Biomarkers and Therapeutic Targets in Human Disease: From Physiology to Clinical Translation.

Carbonic anhydrases (CAs) are zinc-containing metalloenzymes that catalyze the reversible conversion of carbon dioxide and water into bicarbonate and protons, contributing to acid-base balance, pH regulation, and ion transport. Among human cytosolic isoforms, carbonic anhydrase I (CA I) and carbonic anhydrase II (CA II) are abundant and clinically relevant, yet their distinct roles are often obscured within broader discussions of the CA family. This narrative review evaluates CA I and CA II as biomarkers and therapeutic targets in glaucoma, atherosclerosis and vascular calcification, anemia, e

Considerations for enhancing the clinical translational potential of LLM-Based TBI mortality prediction models.

This study explores the use of GPT-5 and traditional machine learning by scholars such as Tu et al. to predict the risk of emergency death in traumatic brain injury (TBI), and affirms the value of their experimental design and method exploration in promoting AI assisted TBI triage. At the same time, it is pointed out that there are still four shortcomings in this study: a lack of clinical experts to validate LLM output, no exploration of subgroup dynamic threshold adaptation, no evaluation of model inference delay, and no sensitivity analysis of prompt words. The study suggests that further re
