Intriguing olfactory proteins from the yellow fever mosquito, Aedes aegypti.

Four antennae-specific proteins (AaegOBP1, AaegOBP2, AaegOBP3, and AaegASP1) were isolated from the yellow fever mosquito, Aedes aegypti and their full-length cDNAs were cloned. RT-PCR indicated that they are expressed in female and, to a lesser extent, in male antennae, but not in control tissues (legs). AaegOBP1 and AaegOBP3 showed significant similarity to previously identified mosquito odorant-binding proteins (OBPs) in cysteine spacing pattern and sequence. Two of the isolated proteins have a total of eight cysteine residues. The similarity of the spacing pattern of the cysteine residues …

Neurovirulence of yellow fever 17DD vaccine virus to rhesus monkeys.

The yellow fever 17D virus is attenuated and used for human vaccination. Two of its substrains, 17D-204 and 17DD, are used for vaccine production. One of the remarkable properties of this vaccine is limited viral replication in the host but with significant dissemination of the viral mass, yielding a robust and long-lived neutralizing antibody response. The vaccine has excellent records of efficacy and safety and is cheap, used as a single dose, and there are well-established production methodology and quality control procedures which include the monkey neurovirulence test (MNTV). The present …

Vitelline envelope genes of the yellow fever mosquito, Aedes aegypti.

Vitelline envelope genes from the mosquito Aedes aegypti were analyzed with respect to their DNA sequences, genomic representation, temporal and spatial expression profiles and response to 20-hydroxyecdysone. Genomic clones of three vitelline envelope genes, 15a-1, 15a-2 and 15a-3 were isolated. Southern analysis indicates that all three genes are represented by a single copy in the genome. The deduced amino acid sequences of all three vitelline envelope genes contain a conserved region of 46 residues that overlaps with a region that is conserved in four Drosophila melanogaster vitelline envel

Stability of 17D yellow fever virus vaccine using different stabilizers.

To optimize the thermostability of lyophilized 17D vaccine, the authors investigated parameters important for the freeze-drying process. Six different stabilizers with different sugars and amino acids were analysed in a freeze-thaw cycle for their crystallization characteristics and their stabilizing effect under thermal treatment conditions of 37 degrees C for 28 days. This test indicated that three out of six stabilizers (B, C, F) kept the vaccine significantly more stable than the three others (A, D, E). Under storing conditions of 4 degrees C over 96 days stabilizers A, B and C produced th

[Reflection on 2 current viral diseases: yellow fever and dengue].

Yellow fever and dengue are two current viral diseases induced by flaviviruses and usually transmitted by the same mosquito vector, Aedes aegypti. From 1987 to 1991, 18,753 cases of yellow fever, mainly from Africa, have been notified to WHO, leading to 4,522 deaths. On the other hand, WHO estimates that 2.5 billions individuals living in tropical areas are at risk to contract dengue fevers. In fact, 500,000 patients are hospitalized each year for dengue hemorrhagic fever/dengue shock syndrome and 90% of them are children. Nevertheless, the control of these two viral diseases would be reached …

Phenotypic analysis of yellow fever virus derived from complementary DNA.

A thorough phenotypic characterization of yellow fever (YF) virus generated from cDNA is a necessary prerequisite for mapping virulence/attenuation determinants and exploring the potential of YF attenuated virus 17D as a carrier for heterologous protective epitopes. In this study, YF virus was produced from 17D cDNA clones after lipofectin-mediated RNA transfection of certified primary cultures of chicken embryo fibroblasts (YFiv5.2/SL). This virus was passaged once in embryonated chicken eggs according to current YF vaccine manufacture methodology to produce the experimental virus (YFiv5.2/VL…

Yellow fever outbreak in Ipetu-Ijesa: response after mass vaccination.

An outbreak of yellow fever in Ipetu-Ijesa, Nigeria, was confirmed by serological test and virus isolation. Serologically testing of subjects 12 weeks after an emergency mass vaccination following the outbreak showed a high degree of cross reactivity between yellow fever virus and seven other group B flaviviruses. There was no significant difference in the prevalence of neutralizing antibodies to yellow fever vaccine and wild virus among the vaccinated subjects. These findings are attributed to the probable hyperactivity and an all time exposure to yellow fever virus in circulation in the stud…

Morphogenesis of yellow fever virus 17D in infected cell cultures.

The morphogenesis of yellow fever virus replication was examined in infected Vero cell cultures. Penetration and uncoating occurred by endocytosis with the formation of coated vesicles, similar to that demonstrated for other enveloped and unenveloped viruses. Inclusion bodies associated with newly formed nucleocapsids were evident in the perinuclear region during the growth cycle. No evidence of RNA synthesis in the vicinity of the inclusion bodies was obtained by autoradiography, suggesting that genome replication and assembly of viral nucleocapsids occur at separate cytoplasmic sites. An exc…

Studies on yellow fever vaccine. III--Dose response in volunteers.

Standard and stabilized yellow fever (YF) vaccines were compared on the basis of the serological responses of human volunteers to varying doses of vaccine measured as pfu or LD50. The addition of stabilizer substances to bulk vaccine did not affect the immunogenicity and stabilized vaccine gave a consistently good performance. The vaccine fulfilled WHO recommendations in inducing 100% serological conversion in volunteers given about 200 pfu or 600 LD50.

Hormone-controlled cAMP-mediated fluid secretion in yellow-fever mosquito.

Evidence is presented for hormone-controlled adenosine 3',5'-cyclic monophosphate (cAMP)-mediated NaCl diuresis in Malpighian tubules of the blood-feeding yellow-fever mosquito Aedes aegypti. Studies in isolated Malpighian tubules reveal that cAMP added to the peritubular bath selectively stimulates NaCl secretion and not KCl secretion by increasing the Na conductance of the basolateral membrane of primary cells. These effects are duplicated by forskolin and theophylline in parallel with increased intracellular concentrations of endogenous cAMP. Two natriuretic peptides that we have isolated b

Present status of yellow fever: memorandum from a PAHO meeting.

An international seminar on the treatment and laboratory diagnosis of yellow fever, sponsored by the Pan American Health Organization (PAHO) and held in 1984, differed from previous meetings on yellow fever because of its emphasis on the care and management of patients and because the participants included specialists from several branches of medicine, such as hepatology, haematology, cardiology, infectious diseases, pathology and nephrology. The meeting reviewed the current status of yellow fever and problems associated with case-finding and notification; features of yellow fever in individua…

[Yellow fever in Senegal from 1976 to 1980 (author's transl)].

The isolation of a YF strain in Kedougou (eastern region) in December 1976 from Aedes gr. furcifer taylori lead to a mass vaccination campaign in Senegal. In 1977, 67 strains were isolated from different mosquitoes (essentially A. luteocephalus and A. gr. furcifer taylori). Among these strains, 3 were isolated from males and this very important phenomenon might explain the virus conservation during the dry season. In 1978, 55 strains were isolated from mosquitoes of the same species, but not from males, and five from monkeys (E. patas and C. aethiops). But no increase of morbidity or mortality

Evidence of recent jungle Yellow-Fever activity in Eastern Panama.

Outbreaks of jungle yellow fever in man have been recorded twice from eastern Panama of recent years, first in 1948 and again in 1956. Since then, a close surveillance has been maintained on virus activity in eastern Panama. Recent field observations and serological tests on 402 monkey sera indicate that there was an outbreak of yellow fever among monkeys of southern Darién Province some time between 1963 and 1965. It does not appear that the outbreak has spread as yet to other areas. Virus transmission may have been permanently disrupted during the drought which affected the region in 1965. H…
