BACKGROUND: removal of incompatible red blood cells (RBCs) or plasma is usually required to avoid hemolysis during infusion of ABO incompatible bone marrow (BM) allogeneic transplants. This process often involves separation of buffy coat (BC) by centrifugation in automated devices. We have evaluated the Spectra Optia™ (Optia) apheresis system to determine its effectiveness in BC concentration, volume reduction and RBCs depletion of ABO-incompatible BM compared with our previous method using Cobe Spectra™ (Cobe).

In our group of newborns we have observed that the rate of occurrence of incompatibility states between blood group 0 mothers and their blood group A or B offspring amounted to 14%. It was in every tenth newborn that this incompatibility became manifest as a haemolytic disease requiring treatment. Antibody-dependent cell-mediated cytotoxicity (ADCMC) was measured in a 51Cr-release assay with antibody-sensitized human red blood cells (RBC) as target cells and nonimmune isologous or autologous mononuclear peripheral blood leukocytes (MPBL) as effector cells. BACKGROUND: A positive flow-cytometry T cell crossmatch (FTXM) has important prognostic implications, even when the complement-dependent cytotoxicity crossmatch is negative. Linkage means a very close association between two adjacent genes, so much so that no crossing-over occurs between them and they are always transmitted together to the next generation, provided there is no mutation. The linkage between nail-patella gene and ABO blood group gene, located on the 9th chromosome, shows the same. This can be understood from the pedigree analysis of an affected family, which is the first step in studying the molecular pathology of the diseased gene.

We studied 526 consecutive post-transplant platelet transfusions to determine the factors associated with response to platelet transfusion in the BMT setting. Poor responses to platelet transfusions occurred frequently, with 310 of the 484 evaluable transfusions (64%) resulting in post-infusion corrected count increments of less than 7500.
