Yersinia pestis outer membrane type III secretion protein YscC: expression, purification, characterization, and induction of specific antiserum.

The type III secretion system (YscC) protein of Yersinia pestis plays an essential role in the translocation of Yersinia outer proteins (Yops) into eukaryotic target cells through a type III secretion mechanism. To assess the immunogenicity and potential protective efficacy of YscC against lethal plague challenge, we cloned, overexpressed, and purified YscC using two different bacterial expression and purification systems. The resulting expression plasmids for YscC, pETBlue-2-YscC and pTYB11-YscC, were regulated by robust T7 promoters that were induced with isopropyl-beta-D-thiogalactopyranosi

Pleiotropic effects of signal peptide peptidase A (sppA) gene deletion on membrane homeostasis, gliding motility, and virulence in Flavobacterium columnare.

Columnaris disease, caused by Flavobacterium columnare, represents one of the most economically devastating bacterial infections in global freshwater aquaculture. Despite its significant impact, the molecular mechanisms underlying F. columnare pathogenesis remain largely unexplored. Signal peptide peptidase A (SppA) plays a crucial role in bacterial protein secretion by degrading residual signal peptides after protein translocation, yet its function in F. columnare physiology and virulence has not been characterized. Here, we employed a targeted in-frame gene knockout approach to investigate t

M2b Monocytes Provoke Bacterial Pneumonia and Gut Bacteria-Associated Sepsis in Alcoholics.

Chronic alcohol consumption markedly impairs host antibacterial defense against opportunistic infections. γ-irradiated NOD-SCID IL-2Rγ(null) mice inoculated with nonalcoholic PBMCs (control PBMC chimeras) resisted Klebsiella pneumonia and gut bacteria-associated sepsis, whereas the chimeras created with alcoholic PBMCs (alcoholic PBMC chimeras) were very susceptible to these infections. M1 monocytes (IL-12(+)IL-10(-)CD163(-)CD14(+) cells), major effector cells in antibacterial innate immunity, were not induced by a bacterial Ag in alcoholic PBMC cultures, and M2b monocytes (CCL1(+)CD163(+)CD14

Increased susceptibility to resveratrol of Helicobacter pylori strains isolated from patients with gastric carcinoma.

The increasing therapeutic failures against Helicobacter pylori infection has determined the need to develop new drugs. The susceptibility to resveratrol (1) of twenty-six H. pylori strains representing nine CagA+ strains from patients with gastric carcinoma and eight CagA- and nine CagA+ strains from patients with chronic gastritis only was evaluated. Compound 1 was dissolved in DMSO and double diluted in Brucella broth with 10% BFS; ca. 10(6) organisms were added to each dilution. After incubation, subcultures were performed using Columbia-blood agar plates. The lowest concentration in broth

Quercetin Prevents Escherichia coli O157:H7 Adhesion to Epithelial Cells via Suppressing Focal Adhesions.

The attachment of Escherichia coli O157:H7 to intestinal epithelial cells is indispensable for its pathogenesis. Besides translocated-intimin receptor (Tir), E. coli O157:H7 interacts with host cell surface receptors to promote intimate adhesion. This study showed that integrin β1 was increased in Caco-2 cells upon E. coli O157:H7 infection, while Caco-2 cells subjected to integrin β1 antibody blocking or CRISPR/Cas9 knockout had reduced bacterial attachment. Infection of E. coli O157:H7 inactivated focal adhesion kinase (FAK) and paxillin, increased focal adhesion (FA) and actin polymerizatio

The secreted protein kinase CstK from Coxiella burnetii influences vacuole development and interacts with the GTPase-activating host protein TBC1D5.

The intracellular bacterial pathogen Coxiella burnetii is the etiological agent of the emerging zoonosis Q fever. Crucial to its pathogenesis is type 4b secretion system-mediated secretion of bacterial effectors into host cells that subvert host cell membrane trafficking, leading to the biogenesis of a parasitophorous vacuole for intracellular replication. The characterization of prokaryotic serine/threonine protein kinases in bacterial pathogens is emerging as an important strategy to better understand host-pathogen interactions. In this study, we investigated CstK (for Coxiella Ser/Thr kinas

Mechanism of Antibacterial Activities of a Rice Hull Smoke Extract (RHSE) Against Multidrug-Resistant Salmonella Typhimurium In Vitro and in Mice.

UNLABELLED: The present study tested antibacterial activity of a rice hull smoke extract (RHSE) against a multidrug-resistant strain of Salmonella Typhimurium and examined its mode of suppressive action in vitro and in mice. In vitro studies showed that the minimum inhibitory concentration (MIC) value of RHSE was 1.29% (v/v). The inactivation was confirmed by complete loss of cell viability in the range of 104 to 107 colony forming units of the resistant Salmonella Typhimurium strain. Agarose and sodium dodecyl sulfate-polyacrylamide gel electrophoreses were used to evaluate the integrities of

Abnormal Blood Bacteriome, Gut Dysbiosis, and Progression to Severe Dengue Disease.

Despite a well-known association between gut barrier defect (leaky gut) and several diseases, data on translocation of pathogen molecules, including bacterial DNA (blood bacteriome), lipopolysaccharide (LPS), and serum (1→3)-β-D-glucan (BG), from the gut to the blood circulation (gut translocation) in dengue are still less studied. Perhaps, dengue infection might induce gut translocation of several pathogenic molecules that affect the disease severity. At the enrollment, there were 31 dengue cases in febrile and critical phases at 4.1 ± 0.3 days and 6.4 ± 1.1 days of illness, respectively, wit

Acute-on-chronic liver failure and immune dysregulation: Systemic inflammation and immunoparesis paradox.

Acute-on-chronic liver failure (ACLF) is a severe clinical syndrome characterized by abrupt hepatic decompensation and systemic inflammation in patients with underlying chronic liver disease, frequently progressing to multi-organ failure and high short-term mortality. Although ACLF is increasingly recognized as a distinct clinical entity, its pathogenesis remains incompletely understood, and diagnostic criteria vary across international guidelines. Central to ACLF is immune dysregulation, driven by systemic inflammation triggered by bacterial translocation, pathogen-associated molecular patter…

Gut-liver axis drives the triple mechanism of liver inflammation-carcinogenesis: Microbial-immune-metabolic network.

The gut-liver axis, through intricate microbial-immune-metabolic networks, plays a pivotal role in driving hepatic fibrosis. Chronic liver injury activates hepatic stellate cells, resulting in pathological extracellular matrix deposition. Gut dysbiosis, characterized by altered microbial metabolites and bacterial translocation, significantly modulates this process. Bile acid and short-chain fatty acid metabolism, alongside immune responses involving Toll-like receptors and cytokines, orchestrate inflammatory and fibrogenic cascades. Additionally, metabolic reprogramming, featuring aerobic glyc…

Recent advances in the prevention and treatment of decompensated cirrhosis and acute-on-chronic liver failure (ACLF) and the role of biomarkers.

The progression of cirrhosis with clinically significant portal hypertension towards decompensated cirrhosis remains clinically challenging and the evolution towards acute-on-chronic liver failure (ACLF), with one or more extrahepatic organ failures, is associated with very high mortality. In the last decade, significant progress has been made in the understanding of the mechanisms leading to decompensation and ACLF. As portal hypertension advances, bacterial translocation across an impaired gut barrier culminates in endotoxaemia, systemic inflammation and cirrhosis-associated immune dysfuncti…

Gut microbiome and nonalcoholic fatty liver disease.

Nonalcoholic fatty liver disease (NAFLD) has become the most prevalent chronic liver disease globally and imposed a heavy economic burden on society and individuals. To date, the pathological process of NAFLD is not yet fully elucidated. Compelling evidences have demonstrated the pivotal role of gut microbiota in the pathogenesis of NAFLD, and gut dysbiosis has been commonly observed in patients with NAFLD. Gut dysbiosis impairs gut permeability, allowing the translocation of bacterial products such as lipopolysaccharides (LPS), short-chain fatty acids (SCFAs), and ethanol to the liver via por

Isolated Multifocal Superior Mesenteric Artery Vasculitis With Coexisting Superior Mesenteric Vein Thrombosis: A Unique Coincidence.

Vasculitis involving the gastrointestinal (GI) tract typically occurs in association with multisystem disease. Isolated superior mesenteric artery (SMA) vasculitis is a rare disorder that has a high degree of morbidity and mortality. Patients often present with nonspecific symptoms, and this condition can lead to varying degrees of intestinal ischemia, significant gastrointestinal bleeding, and bowel infarction, leading to perforation, peritonitis, and sepsis from bacterial translocation. Diagnosing this condition can be very challenging. High clinical suspicion and early diagnosis using both …

Assessing placement of nasoduodenal tube and its usefulness in maintaining nutrition in critically ill patients.

Nutritional supplements to the critically ill patients are one of the major issues to be discussed. Enteric feeding is advantageous over parentral feeding because it maintains gut integrity and prevents bacterial translocation. Small intestinal feeding shows significant beneficial results. Nasoduodenal tube placement and its confirmation by serial pH monitoring and by radiological examination was done; and time taken to reach duodenum was assessed. This study was done in 40 critically ill patients, who were divided into two groups (Group 1 and Group2). Early enteric feeding via a nasoduodenal …

Gas chromatography applied to the lactulose-mannitol intestinal permeability test.

Intestinal permeability can be modified by various illnesses, trauma and sepsis. Alterations of the intestinal wall can facilitate the diffusion of potentially harmful substances such as endotoxins, as well as bacterial translocation. We describe the validation of a capillary gas chromatographic method for the determination of mannitol and lactulose, used as intestinal permeability probes. The method is linear up to 3 g/l for mannitol and 300 mg/l for lactulose; recovery from overload samples is between 92 to 110%. Intra-assay coefficients of variation (C.V.s) were 2.7 and 6.8% for mannitol an…

Increment of enterobacterial common antigens in organ extracts from immunosuppressed mice.

This article describes the enterobacterial common antigen (ECA) content in the liver and spleen extracts from immunosuppressed two months ol … hapten shared by almost all Gram-negative bacteria, and CY is an alkylating agent useful to induce bacterial translocation. Therefore, the increments observed in tissular ECA concentration may indicate the extension of the enterobacterial dissemination from intestine to abdominal organs in CY-treated immunosuppressed mice.

Gaps in the wall: understanding cell wall biology to tackle amoxicillin resistance in Streptococcus pneumoniae.

Streptococcus pneumoniae is the most common cause of community-acquired pneumonia, and one of the main pathogens responsible for otitis media infections in children. Amoxicillin (AMX) is a broad-spectrum β-lactam antibiotic, used frequently for the treatment of bacterial respiratory tract infections. Here, we discuss the pneumococcal response to AMX, including the mode of action of AMX, the effects on autolysin regulation, and the evolution of resistance through natural transformation. We discuss current knowledge gaps in the synthesis and translocation of peptidoglycan and teichoic acids, maj
