A review on assessment of advances in polymers and their hybrid nanosystem for leukaemia theranostics applications.

Leukaemia, a widespread haematological cancer, possesses particular challenges in theranostics despite the availability of traditional and modern approaches. This article addresses a significant gap in the literature: a lack of focus on polymeric and hybrid nanoparticles (P&HNP) in advancing leukaemia therapy. It explores how to overcome limitations of conventional therapeutic approaches. The rationale for this study stems from several considerations, including the fact that while polymeric nanoparticles show promise in leukaemia theranostics, a comprehensive assessment of how biomaterial choi

Nanocolloidal Hydrogel Coatings With Multiple Functions for Antithrombogenic Biointerfaces.

Hydrogel coatings offer a promising strategy for improving the biocompatibility and functionality of medical devices. However, conventional hydrogel coatings often suffer from swelling-induced delamination, poor durability, and limited resistance to thrombosis and biological adhesion. Here, we report a nanocolloidal hydrogel coating (NHC) that shows an important and highly impactful combination of properties, such as lubrication, antibioadhesion, and antithrombogenicity. The NHC is prepared by anchoring a layer of nanocolloidal hydrogel made from methacryloyl-modified hyaluronate nanoparticles

A Universal Gd3+-based hydrogel matrix for inducing room-temperature phosphorescence.

Room-temperature phosphorescence (RTP) is difficult to achieve in aqueous media, due to the inherent and serious quenching of the excited triplet states by O2 and H2O. Herein, we report RTP induction enabled by a hydrogel matrix assembled from Gd3+ and adenosine monophosphate (AMP). Benefiting from the promoted intersystem crossing (ISC), effective guest confinement, and excellent oxygen shielding, the Gd3+-AMP hydrogel permitted efficient RTP emission from guest molecules. Notably, second-scale afterglow (>1 s) was observed from the thioflavin T (ThT)-encapsulated hydrogel, which has rarely b

Encoded Cell-Material Interactions to Reroute Cytokine Signaling for Regenerative Medicine.

Regenerative engineering harnesses materials science and stem cell biology to develop strategies to repair damaged and diseased tissue. Despite advances in designer materials, few techniques effectively provide auto-regulated feedback mechanisms that govern how cells sense and respond to discrete microenvironmental changes. Here, we demonstrate that the artificial, juxtacrine-like receptor synthetic Notch (synNotch) can be activated by endogenous multimeric cytokines in solution, without immobilizing materials, revealing a previously unreported activation modality and yielding up to 24-fold dy

Engineering bioinspired pH-responsive hydrogels for smart wound repair.

The growing prevalence of chronic and infected wounds represents a major clinical challenge that conventional passive dressings fail to adequately address. Bioinspired pH-responsive hydrogels have emerged as intelligent biomaterials capable of dynamically interacting with the wound microenvironment. However, the field still faces major translational barriers, with many laboratory prototypes not yet progressing to clinically or commercially established products. Unlike descriptive literature compilations, this review provides a critical analytical evaluation focused on synthesizing fundamental

Boolean Logic-Based Control Over Recombinant Protein Biomaterial Degradability and Therapeutic Delivery.

Recombinant protein-based biomaterials offer exciting opportunities in materials design and controlled therapeutic delivery. Though their precursors can be readily synthesized with near-perfect monodispersity and sequence specificity through scalable fermentation processes, recombinant protein materials have yet to achieve the same level of multi-stimuli-responsiveness as their synthetic counterparts. Integrating cutting-edge tools from chemical biology, we autonomously compile topologically specified protein crosslinkers that can be degraded following user-programmable Boolean logic. Covalent

Engineering and Exploring Hydrolytic Degradation in 3D-Printed Liquid Crystalline Elastomers.

Liquid crystalline elastomers (LCEs) are being increasingly explored as biomaterials; however, many LCE properties were not designed with biomedical use in mind. Here, we examine LCE hydrolytic degradation, including investigating approaches to accelerate degradation and whether thermal and mechanical properties change with degradation. Among 3D-printable LCE chemistries, we find that networks formed via thiol-Michael addition followed by thiol-ene photo-cross-linking degrade most rapidly. The integration of hydrophilic chain extenders (e.g., PEG) accelerates LCE degradation and demonstrates t

Laser-Assisted Processing and Modification of Bioactive Glasses: A Review.

Bioactive glasses and lasers have followed a relatively similar path. Lasers and bioactive glasses began their journey in the second half of last century. It was in 1960 when Dr. Theodore H. Maiman constructed the first laser and fascinated the world with that marvellous ray. It was in 1967 when a young associate professor, Larry L. Hench, was addressed by a colonel returned from Vietnam, that asked him about developing a material that could help his soldiers recover from severe bone injuries. Two years later, the first bioactive material, the Bioglass, was launched. From there, the developmen

Functional hydrogel biomaterials for urological diagnosis and treatment-design strategies and application progress: a review.

In urology, the treatment of diseases such as bladder cancer, urethral stricture, and benign prostatic hyperplasia faces challenges such as short drug retention time, unstable therapeutic effects, and drug resistance. Traditional treatments, such as surgery, systemic drug therapy, and local drug instillation, although controlling the condition to some extent, still have many limitations, necessitating new localized treatment and precise diagnostic strategies. Hydrogels have made significant progress in urology, especially in the fields of drug delivery, disease diagnosis, and tissue repair. Th

Harnessing Artificial Intelligence for Regeneration of Endometrium in Asherman's Syndrome.

BACKGROUND: Asherman's Syndrome or intrauterine adhesions develop due to acquired endometrium damage, resulting in partial to complete dysfunction of the endometrium within the uterine cavity. The pathophysiology of these adhesions is not clear. Still, the widely accepted mechanism for the development of these adhesions is attributed to three different causes: i. iatrogenic or mechanical, including curettage; ii. pathophysiological conditions, including infection, miscarriage, and Müllerian malformations; and iii. idiopathic. OBJECTIVE: This review critically evaluates the different therapeut

Designing of porous scaffolds for tissue engineering and regenerative medicine.

The potential of the porous scaffolds lies in its dynamic architectural features, such as porosity, pore size & shape, interconnectivity, and spatial distribution which are highly essential in tissue engineering applications. The porous structure of the scaffolds influences the biological and physicochemical activities in the tissue regeneration process. The porosity also allows better distribution of nutrients and diffusion of biological fluids thereby dispersing the seeding material uniformly. Generally, damaged tissues can be regenerated on their own but many times due to unavoidable factor

A Xerotic Skin-Compatible Neuromotor Electrode Interface for Non-Invasive Muscle Weakness Evaluation in Dermatomyositis.

Dermatomyositis creates a clinical demand for non-invasive, quantitative muscle weakness assessment, which can be achieved by decomposing neuromotor information from surface electromyography (sEMG). However, the absence of a stable electrode interface compatible with pathologically xerotic skin has hindered this approach. Here, we develop a conformable neuromotor electrode interface that overcomes this via molecular engineering, exhibiting an ultrasoft modulus (∼ 2.13 kPa), low impedance (∼ 0.9 kΩ at 1 kHz), and stable electrical retention over 7 days. This interface enables the acquisition of

A super absorbent decellularized human amniotic membrane microgel for chronic wound healing and accelerated skin regeneration.

Chronic wounds present a persistent clinical challenge, often requiring grafts for effective healing. Rich extracellular matrix (ECM) components, cytokines, and growth factors in human amniotic membrane, have demonstrated significant potential in promoting chronic wound healing. However, decellularized human amniotic membrane sheets or hydrogels face challenges in application flexibility and cell infiltration in chronic wound healing. In this study, we developed a novel super absorbent microgel derived from decellularized human amniotic membrane, designed as a biocompatible and injectable alte
