Miltefosine (hexadecylphosphocholine, HePC) is the first effective oral agent for the treatment of visceral leishmaniasis. This study aimed to determine whether this oral administration alters the integrity and transport capacities of the intestinal barrier. The objectives of this study were: (i) to evaluate the cytotoxicity of HePC, (ii) to investigate the effects of HePC on paracellular and transcellular transport and (iii) to investigate the influence of HePC on three major transporters of the intestinal barrier, namely, P-glycoprotein, the human intestinal peptide transporter (PepT-1) and

BACKGROUND: Based on the concept of "multimodal analgesia", a novel dual drug delivery system was designed to achieve synergistic analgesia between najanajaatra venom protein (αCT) and resveratrol (Res). In order to meet the joint loading of two drugs with different physicochemical properties without affecting each other, an oral Janus nanoparticle (JNP) with a unique cavity structure and synergistic drug delivery was constructed using an improved double emulsion solvent evaporation method, and combined with low-molecular-weight chitosan/sodium alginate and PLGA to achieve its pH-responsive.

Severe COVID-19 is characterized by lung abnormalities, including the presence of syncytial pneumocytes. Syncytia form when SARS-CoV-2 spike protein expressed on the surface of infected cells interacts with the ACE2 receptor on neighboring cells. The syncytia forming potential of spike variant proteins remain poorly characterized. Here, we first assessed Alpha (B.1.1.7) and Beta (B.1.351) spread and fusion in cell cultures, compared with the ancestral D614G strain. Alpha and Beta replicated similarly to D614G strain in Vero, Caco-2, Calu-3, and primary airway cells. However, Alpha and Beta for

Studying viral infections necessitates well-designed cell culture models to deepen our understanding of diseases and develop effective treatments. In this study, we present a readily available ex vivo 3D co-culture model replicating the human intestinal mucosa. The model combines fully differentiated human intestinal epithelium (HIE) with human monocyte-derived macrophages (hMDMs) and faithfully mirrors the in vivo structural and organizational properties of intestinal mucosal tissues. Specifically, it mimics the lamina propria, basement membrane, and the air-exposed epithelial layer, enabling

INTRODUCTION: Ulcerative colitis (UC) involves impaired wound healing processes contributing to sustained immune and microbial interactions that aggravate intestinal injury and may progress to colitis-associated cancer (CAC). Here we investigated whether PDLIM2, a known regulator of both epithelial and immune cell fate, contributes to colitis progression.
METHODS: PDLIM2 knockout mice (-/-) and wildtype littermates (+/+) were assessed for responses to dextran sodium sulphate (DSS)-induced colitis, and to aoxymethane +DSS. Microbiota were assessed using 16s rRNA amplicon sequencing. Mechanisti

Neurosteroids are endogenous steroidal compounds that can modulate neuronal receptors. N-Methyl-D-aspartate receptors (NMDARs) are glutamate-gated, calcium-permeable ion channels that are of particular interest, as they participate in synaptic transmission and are implicated in various processes, such as learning, memory, or long-term neuronal potentiation. Positive allosteric modulators that increase the activity of NMDARs may provide a therapeutic aid for patients suffering from neuropsychiatric disorders where NMDAR hypofunction is thought to be involved, such as intellectual disability, au

Chitosan is object of pharmaceutical research as a candidate permeability enhancer. However, chitosan was recently shown to reduce the oral bioavailability of acyclovir in humans. The effect of chitosan on two processes determining the oral bioavailability of acyclovir, bioaccessibility and intestinal absorption, was now investigated. Acyclovir's bioaccessibility was studied using the dynamic TNO gastro-Intestinal Model (TIM-1). Four epithelial models were used for permeability experiments: a Caco-2 cell model in absence and presence of mucus and both rat and porcine excised intestinal segment

Mesenchymal stem cell-derived exosomes (Exo) have emerged as potent therapeutic agents for skin regeneration; however, their clinical translation is often impeded by poor colloidal stability and the formidable barrier of the stratum corneum. In this study, we developed a novel "bio-shuttle" system by engineering chitosan-shielded exosomes (cExo) via precise electrostatic surface assembly. The optimized cExo exhibited a uniform hydrodynamic diameter of 216 nm and a robust positive surface charge of +30.7 mV, ensuring superior structural integrity. To evaluate their transdermal potential, we emp

In the present study, we developed novel insulin-loaded hyaluronic acid (HA) nanoparticles for insulin delivery. The insulin-loaded HA nanoparticles were prepared by reverse-emulsion-freeze-drying method. This method led to a homogenous population of small HA nanoparticles with average size of 182.2 nm and achieved high insulin entrapment efficiencies (approximately 95%). The pH-sensitive HA nanoparticles as an oral delivery carrier showed advantages in protecting insulin against the strongly acidic environment of the stomach, and not destroying the junction integrity of epithelial cells which

Pomegranate peel, a major by-product of processing, is rich in water-soluble pectin made of D-galacturonic acid units with α (1-4) linkage. Efficiently converting pomegranate peel into safe bioactive principles minimizes waste in the fruit industry and opens new avenues in functional foods. In this work, pomegranate pectin was enzymatically depolymerized with extracellular pectinases from Streptomyces hydrogenans YAM1 to generate pectic oligosaccharides (POS). Products were characterized by TLC and LC-ESI-MS, which indicated unsaturated mono-, di-, and trigalacturonic acids among the main spec

Determining whether potential cancer therapies effectively kill cancer cells is important for informing effective therapeutic choice for patients. Here we describe a rapid label-free method for testing drug efficacy in vitro that evaluates cellular viability from sub-cellular fluctuation imaging (SCFI). We used staurosporine and paclitaxel as known cytotoxic drugs at different concentrations, and four different human cancer-derived cell lines: PC3 (prostate), Caco-2 (colorectal), Calu-3 (lung) and A549 (lung). Both drugs caused a rapid decrease in sub-cellular fluctuations within 1 to 3 h exce

Erythropoietin-producing hepatocellular receptor A1 (EphA1) is one of the Eph receptor family members, the largest group of receptor tyrosine kinases. EphA1 is expressed in various tissues and regulates cellular homeostasis by interacting with its membrane-bound ephrin ligands and other receptors. EphA1 critically correlates with the pathogenesis in several disorders, including Alzheimer's disease and cancers. Therefore, establishing sensitive monoclonal antibodies (mAbs) for EphA1 has been desired for basic research, diagnosis, and treatment. In this study, a novel specific and sensitive anti

The intestinal barrier is crucial for gut health, and its dysfunction can lead to chronic inflammatory conditions. Food contaminants like pesticides, heavy metals, mycotoxins, heterocyclic aromatic amines (HAAs), and polycyclic aromatic hydrocarbons (PAHs) may worsen these conditions. However, research often overlooks the complex interactions between contaminants and the different cells in the intestine. This study explores the effects of repeated exposure to a mixture of 45 food contaminants on a triculture cell model (Caco-2/HT29-MTX monolayers and THP-1 cells) under normal and inflamed cond
