INTRODUCTION: Chondrocytes in articular cartilage utilize mechanical signals to regulate their metabolic activity. A fundamental step in determining the role of various biophysical factors in this process is to characterize the local mechanical environment of the chondrocyte under physiological loading.
METHODS: A combined experimental and theoretical approach was used to quantify the in-situ mechanical environment of the chondrocyte. The mechanical properties of enzymatically-isolated chondrocytes and their pericellular matrix (PCM) were determined using micropipette aspiration. The values w…

The almost absolute barrier to diffusion of nutrients between articular cartilage and subchondral bone does not exist. These anatomic regions represent a functional unit. Repetitive overloading in degenerative disease leads primarily to lesions in the subchondral region (including vessels), which in turn impede flow of nutrition to articular cartilage. As a result, in degenerative joint disease the subchondral region shows reactive enhanced vascularization and heightened metabolism with insufficient repair. In aging, however, vascularization and metabolism are decreased; no repair takes place.…

We present a new method of treatment of chondral defects of the knee with the usage of carbon fibers. The prospective study describes experience with carbon fibers used as scaffolds in the drilled lesions to enhance ingrowth of regenerative tissue. We treated 35 patients for chondral defects with that method from December 93 to June 97. Average age was 46 years (range, 19-68 years) and average follow-up was 48 months (range, 24-55 months). The results were assessed by HSS knee scale, the Wallgren-Tegner activity score, VAS (visual analogue scale to measure pain) and patients assessment of the

OBJECTIVE: To study the viscoelastic properties of human articular cartilages of patella in 8 cadavers, which were obtained from patients ranged in age from nineteen to fifty-one years old and died of acute head injury.
METHODS: The creep and stress relaxation of articular cartilage of patella were abserved in the condition of articular cartilage loaded of 500 Pa, just as the physiological load of articular cartilage of patella in keeping human body standing position with the knee joint flexed 30 degrees, during the experiment. The deformation-time curve under certain load, the load-time curv…

Endochondral bone formation in vertebrates requires precise coordination between proliferation and differentiation of the participating chondrocytes. We examined the role of perichondrium in this process using an organ culture system of chicken embryonic tibiotarsi. A monoclonal antibody against chicken collagen type X, specifically expressed by hypertrophic chondrocytes, was utilized to monitor the terminal differentiation of chondrocytes. Proliferation of chondrocytes was examined by a BrdU-labeling procedure. The absence of perichondrium is correlated with an extended zone of cartilage expr…

The objective of this study was to examine the effect of a bipolar ablation probe on experimentally roughened articular cartilage and compare it with the traditional mechanical shaving technique using the knee joint of sheep. Twenty-eight skeletally mature ewes were divided randomly into two groups: one group was treated with a rotating shaving device and another group was treated using the bipolar ablation probe (Bipolar Arthroscopic Probe; Electroscope, Inc, Boulder, CO). Animals were killed at 0, 6, 12, and 24 weeks, and histological sections of the experimental limbs were compared with sec…

Three patients with the typical features of Larsen's syndrome are described. All three developed severe respiratory symptoms caused by a congenital subglottic stenosis. Tracheotomy and treatment of the stenosis by means of laryngotracheoplasty resulted in complete collapse of the cricoid cartilage and the proximal tracheal skeleton. Lack of rigidity of the laryngeal and tracheal cartilages in patients with Larsen's syndrome could well be responsible for this failure. Surgical treatment consisted of resection of the stenotic and collapsed areas and end-to-end anastomosis. This therapy was event…

The frequency of concomitant meniscal and articular cartilage lesions in the femorotibial joint was analyzed in a retrospective study of 1740 knee joints examined arthroscopically with the objective of determining possible correlations between the two knee joint abnormalities. Articular cartilage lesions were found in 81.4% (N = 1416) of femorotibial joints examined and meniscal derangements were noted in 72.8% (N = 1268). In the medial compartment, concomitance was noted in 76.3% (821 of 1076) on the femoral condyles and in 48.6% (523 of 1076) on the tibial plateau. In the lateral compartment…

An improved burying method for salvaging an amputated auricle is presented. We elevated a mastoid skin flap and a fascial flap simultaneously and inserted the denuded auricular cartilage between them. This procedure can maintain the auricular cartilage in a more natural form by eliminating undue skin tension and can give a favorable aesthetic result.

Osteoarthritis (OA) is a heterogeneous disease resulting from multiple pathogenic mechanisms. Several forms of OA are inherited in an autosomal Mendelian pattern. Recent studies have identified a variety of mutations in the type II collagen gene, which encodes the main collagenous component of articular cartilage in some forms of familial OA and other heritable diseases of cartilage, including Stickler syndrome and the chondrodysplasias. Mutations in the type X collagen gene, which encodes a major collagen of growth plate cartilage, have been identified in the Schmid type of osteochondrodyspla…

The presence of lysylpyridinoline (LP) as a nonreducible cross-link in appreciable quantities has primarily been limited to the mineralized tissues, bone and dentin. However, the results reported here show that LP is not only present in the hypertrophic cartilage of the tibiotarsus isolated from newly hatched broiler chicks, but it is approx. 4-fold as concentrated as hydroxylysylpyridinoline (HP). Bone and articular cartilage surrounding the hypertrophic cartilage do not contain measurable quantities of LP. Purified LP has a fluorescent scan similar to purified HP and literature values, confi…

In an experimental study in growing rabbits an endolaryngeal injury to the subglottis resulted in the development of a stenosis due to the formation of scar tissue containing ectopic cartilage. For comparison, biopsies taken from the subglottic stenosis in 8 children were studied histologically. In 6 cases ectopic cartilage was observed; all patients had a history of endotracheal intubation. In 3 children the diagnosis hamartoma was made. In the remaining 3 cases the formation of ectopic cartilage might have been a direct reaction to the endolaryngeal intubation. The observations suggest that …

An asymptotic analysis of a lubrication problem is presented for a model of articular cartilage and synovial fluid under the squeeze-film condition. This model is based upon the following constitutive assumptions: (1) articular cartilage is a linear porous-permeable biphasic material filled with a linearly viscous fluid (i.e. Newtonian fluid); (2) synovial fluid is also a linearly viscous fluid. The geometry of the problem is defined by assuming that (1) cartilage is a uniform layer of thickness H; (2) synovial fluid is a very thin layer compared to H; (3) the radius R of the load-supporting a…

A patient complaining of headaches and bone pain at multiple sites had Tc-99m MDP scintigraphy performed for possible bone trauma after a motor vehicle accident. Bone imaging revealed a small focal increase in tracer uptake in the area of the laryngeal cartilage. There was some reluctance to place a label on this abnormality because the findings were so unusual. The initial diagnosis was a probable fracture of the laryngeal cartilage. Computed tomography of the neck also demonstrated a fracture but correctly localized it to the thyroid cartilage.

Islands of mature cartilage or bone were observed in 6 respectively 7 out of 1395 carotid bodies of rats studied. They were found only in spontaneously hypertensive rats of the Okamoto-Aoki stain and situated regularly at the margin of the carotid bodies. The bone islands were bigger than those of cartilage. The presence of such tissue components in the carotid bodies of rats is discussed in connection with the embryology of this peripheral arterial chemoreceptor, especially its mesodermal origin.

The cartilage model of the rat's patella constitutes a formation susceptible of inducing ossification, following its transplantation in a variety of sites (muscle, thyroïd, testis, ovary, anterior chamber of the eye, etc.), including those considered inappropriate in other experiments of induced ossification (liver, kidney). So we have an experimental model allowing the influence on the osteogenesis of various factors, local or general, natural or experimental, to be studied. Ossification occurs in this model only in cases of histocompatibility between donor and recipient. It is constant after…

Degradation of proteoglycans in growth cartilage plays an important role during endochondral ossification. Degradation is controlled by certain enzymes secreted by macrophages. Vitamin D3 metabolites markedly enhance the degradation of proteoglycans by macrophages in vitro. However, this enhancing activity of D3 metabolites is eliminated when the growth cartilage cells are lysed with 0.2N NH4OH. Puromycin (10 micrograms/ml) also inhibits the enhancement effect. These results suggest that D3 metabolites stimulate the synthesis of a D3-dependent substance by growth cartilage cells and that subse…

Responses of cultured cartilage cells to metabolites of vitamin D3 were studied. Cells were obtained from the epiphyseal growth plate of rachitic chicks and were exposed to physiological and pharmacological concentrations of three metabolites of vitamin D3, 25 hydroxyvitamin D3 (25(OH)D3), 24,25-dihydroxyvitamin D3 (24,25(OH)2D3) and 1,25-dihydroxyvitamin D3 (1,25(OH)2D3). 1,25(OH)2D3 was found to reduce L-[U-14C]leucine incorporation into proteins and Na2 35SO4 incorporation into proteoglycans. The synthesis of 24,25(OH)2D3 from 25(OH)D3 was stimulated upon addition of 1,25(OH)2D3 to the cult…

UNLABELLED: In joints with bacterial arthritis, continuing prolonged destruction of cartilage may occur in spite of prompt, effective antibiotic therapy. We measured the extent to which early antibiotic therapy with ceforanide altered the degradation of the cartilage after arthritis due to Staphylococcus aureus had been produced in the knee joint in rabbits. Degradation of the cartilage was quantified by analyses for glycosaminoglycan and collagen. Three weeks after the infection was produced, the cartilage had lost more than half of its glycosaminoglycan whether the antibiotic therapy had bee…

It is reported about the intraseptal implantation of allogeneic cartilago quadrangularis for the correction of saddle nose in the case of eleven patients. The indication for the implantation were non sufficient residuals of the behind septum for a substitution plastic. The sagging of the noses was caused by iatrogenic in 6 and traumatic influences in 5 cases. The conservation of the septal cartilage was carried out in Cialit 1:5000. All the septa healed in without any reaction and obtained satisfactory functional and cosmetic results.
