Cross-cohort DNA methylation analysis shows CpG island promoter hypermethylation features associated with cervical lesion grade.

BACKGROUND: Cervical lesion progression is accompanied by epigenomic alterations, but reproducible methylation features across public cohorts require cautious re-analysis. STUDY DESIGN AND METHODS: We performed a secondary analysis of public Illumina 450K methylation cohorts: GSE46306 as discovery (20 normal, 17 CIN3, six carcinoma), GSE99511 and GSE186835 as validation, and TCGA-CESC tumors for methylation-RNA expression association. Shared autosomal CpGs were screened for monotonic grade-associated change, cross-cohort directional consistency, promoter annotation, regional aggregation, samp

Proteomic Analysis of TCGA Data Reveals Limited Prognostic Value of p53 but Suggests BAX as a Potential Survival Marker in Cervical Carcinoma.

Cervical carcinoma is predominantly driven by high-risk human papillomavirus (HPV) infection, in which functional inactivation of p53 occurs mainly through viral oncoproteins rather than mutations in the TP53 gene. This raises questions concerning the prognostic relevance of TP53 gene status and p53 protein levels in this tumor type. This study aimed to systematically evaluate the prognostic significance of TP53 mutation status, p53 protein abundance, and selected downstream p53-regulated proteins in cervical carcinoma, with particular focus on tumors carrying wild-type TP53. Clinical and prot

Systems-level proteomic profiling reveals modular control of human cervical remodeling during pregnancy†.

Cervical remodeling (CR) is essential for pregnancy maintenance and successful parturition, yet its molecular regulation in humans remains incompletely defined. This study aimed to present a high-resolution, longitudinal proteomic atlas of the human CR across normal gestation. A retrospective study was conducted using cervical tissue samples collected during nonpregnancy and at three trimesters. The study cohort included 10 healthy controls and 31 pregnant women with uncomplicated singleton pregnancies. Proteomic analyses were performed using data-independent acquisition mass spectrometry. Effectiveness of regional diffusion MRI measures in distinguishing multiple sclerosis abnormalities within the cervical spinal cord.

INTRODUCTION: Multiple sclerosis (MS) is an inflammatory disorder of the central nervous system. Although conventional magnetic resonance imaging (MRI) is widely used for MS diagnosis and clinical follow-up, quantitative MRI has the potential to provide valuable intrinsic values of tissue properties that can enhance accuracy. In this study, we investigate the efficacy of diffusion MRI in distinguishing MS lesions within the cervical spinal cord, using a combination of metrics extracted from diffusion tensor imaging and Ball-and-Stick models. METHODS: We analyzed spinal cord data acquired from
