Cell Adhesion Factors in the Orbitofrontal Cortex Control Cue-Induced Reinstatement of Cocaine Seeking and Amygdala-Dependent Goal Seeking.

Repeated cocaine exposure causes dendritic spine loss in the orbitofrontal cortex, which might contribute to poor orbitofrontal cortical function following drug exposure. One challenge, however, has been verifying links between neuronal structural plasticity and behavior, if any. Here we report that cocaine self-administration triggers the loss of dendritic spines on excitatory neurons in the orbitofrontal cortex of male and female mice (as has been reported in rats). To understand functional consequences, we locally ablated neuronal β1-integrins, cell adhesion receptors that adhere cells to t

Sigma-1 receptor activation mediates a faster antidepressant-like effect through disinhibition of the DRN5-HT-mPFCpyr neural circuit via S1R/5-HT1AR heterocomplex.

BACKGROUND: The Sigma-1 receptor (S1R) represents a promising target for faster-onset antidepressants. Nevertheless, the mechanism underlying the delayed onset of therapeutic action exhibited by S1R-activating selective serotonin reuptake inhibitors (SSRIs) remains unclear. Thus, elucidating the role of S1R in mediating faster antidepressant-like effects is of considerable significance. OBJECTIVE: This study aimed to elucidate the role of S1R in mediating faster onset antidepressant-like effects and to investigate the underlying mechanism. METHODS: To detect the formation of S1R/5-hydroxytry

Differential roles of EA-TRAPed cells in the anterior cingulate cortex across various intervention times in inflammatory pain.

BACKGROUND: The analgesic effects of multiple electroacupuncture (EA) sessions and single EA sessions differ significantly in pain management. Area 24b (A24b) of the anterior cingulate cortex (ACC) is crucial in pain processing. EA relieves pain by targeting and modulating the neuronal activity within this subregion. However, whether the cumulative effect of EA antinociception is connected to A24b mechanisms has remained unclear. METHODS: In our study, we used the Complete Freund's Adjuvant (CFA) model to induce inflammatory pain and the Spared Nerve Injury (SNI) model to induce neuropathic p

Shuyu capsule improves estrous cycle-dependent depression-like behavior in premenstrual dysphoric disorder (PMDD) mice by increasing GABAergic neuronal activation and downregulating GABAAR δ subunit expression in the dorsal periaqueductal gray (dPAG) region.

Premenstrual dysphoric disorder (PMDD), a subtype of premenstrual syndrome (PMS), involves physical and emotional symptoms that impact patients' daily lives and productivity. A reliable, side-effect-free clinical intervention is needed. Shuyu capsule is an effective traditional Chinese medicine preparation for PMDD used in the clinics, but its therapeutic mechanism remains unclear. Previous research has suggested that the γ-aminobutyric acidergic (GABAergic) system in the periaqueductal gray (PAG) may play a role in treating PMDD with traditional Chinese medicine, but there is a lack of functi

The brainstem BBSome regulates glucose homeostasis and lean mass in a state-dependent manner.

OBJECTIVE: Obesity disrupts metabolic homeostasis through changes in brain function. Hypothalamic cilia and associated proteins, such as the BBSome, a protein complex composed of eight Bardet-Biedl syndrome (BBS) proteins, have been implicated in metabolic regulation and disorders. Here, we investigated the significance of brainstem cilia and the BBSome for energy balance and glucose homeostasis. METHODS: Primary cilia were assessed by immunofluorescence and confocal imaging, and brainstem neuron transcriptomes were analyzed using single-cell RNA sequencing. Mice with Phox2b-specific deletion

Distinct ACC Neural Mechanisms Underlie Authentic and Transmitted Anxiety Induced by Maternal Separation in Mice.

It is known that humans and rodents are capable of transmitting stress to their naive partners via social interaction. However, a comprehensive understanding of transmitted stress, which may differ from authentic stress, thus revealing unique neural mechanisms of social interaction resulting from transmitted stress and the associated anxiety, is missing. We used, in the present study, maternal separation (MS) as a stress model to investigate whether MS causes abnormal behavior in adolescence. A key concern in the analysis of stress transmission is whether the littermates of MS mice who only wi

Heat Shock Protein Family A Member 1 Promotes Intracellular Amplification of Hepatitis B Virus Covalently Closed Circular DNA.

Hepatitis B virus (HBV) contains a partially double-stranded relaxed circular DNA (rcDNA) genome that is converted into a covalently closed circular DNA (cccDNA) in the nucleus of the infected hepatocyte by cellular DNA repair machinery. cccDNA associates with nucleosomes to form a minichromosome that transcribes RNA to support the expression of viral proteins and reverse transcriptional replication of viral DNA. In addition to the de novo synthesis from incoming virion rcDNA, cccDNA can also be synthesized from rcDNA in the progeny nucleocapsids within the cytoplasm of infected hepatocytes vi

Glutamatergic Neurons in the Preoptic Hypothalamus Promote Wakefulness, Destabilize NREM Sleep, Suppress REM Sleep, and Regulate Cortical Dynamics.

Clinical and experimental data from the last nine decades indicate that the preoptic area of the hypothalamus is a critical node in a brain network that controls sleep onset and homeostasis. By contrast, we recently reported that a group of glutamatergic neurons in the lateral and medial preoptic area increases wakefulness, challenging the long-standing notion in sleep neurobiology that the preoptic area is exclusively somnogenic. However, the precise role of these subcortical neurons in the control of behavioral state transitions and cortical dynamics remains unknown. Therefore, in this study

The hypothalamic orexin-medial prefrontal cortex pathway modulates susceptibility to depression in the learned helplessness paradigm in male mice.

Depression is a prevalent psychiatric disorder primarily characterized by behavioral despair and helplessness. The orexinergic system plays a crucial role in stress-induced depression; however, the precise mechanisms remain unclear. The medial prefrontal cortex (mPFC), a major target of orexinergic projections, is essential for emotional regulation. This study suggests that impaired orexinergic input to the mPFC underlies depression-like behaviors in murine models. In the learned helplessness (LH) paradigm, the majority of male mice subjected to inescapable foot shocks developed depression-lik

A modified FLIT-B model of trigeminal neuralgia reveals anterior cingulate cortex involvement in anxiety-like behavior in mice.

BACKGROUND: Trigeminal neuralgia (TN) is a debilitating orofacial pain disorder characterized by paroxysmal, lancinating pain and prominent affective comorbidities. Increasing evidence suggests that chronic TN involves abnormal plasticity in supraspinal pain circuits. However, the lack of animal models that faithfully recapitulate key clinical features of TN has limited mechanistic investigations of these central processes. Here, building upon our previously established foramen lacerum impingement of the trigeminal nerve root (FLIT) model, we developed an improved glass bead-based version (FLI

Opioid- and NMDA-receptor-dependent neural plasticity mediates long-term analgesia from motor cortical stimulation.

Exogenous opioids that activate μ-opioid receptors (MORs) in nociceptive circuits mediate transient pain relief lasting minutes to hours but have more limited utility for treating chronic pain. By comparison, electrical or magnetic stimulation of the motor cortex can induce pain relief lasting weeks, for which the underlying mechanisms have remained unclear. Here we report an unconventional role for endogenous opioidergic signaling in the rapid induction of long-lasting analgesia from motor cortical stimulation, which triggers opioid-peptide-dependent neural plasticity in the rostral ventromed

Ultrasound-driven piezoelectric nanomaterial platform enables wireless neuromodulation for Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dysfunction of the basal ganglia circuitry, leading to both motor and cognitive impairments. Current neuromodulation strategies for deep brain targets remain highly invasive and lack precise spatiotemporal controllability. For example, traditional deep brain stimulation offers substantial relief from PD's movement-related symptoms by surgically placing electrodes in the subthalamic nucleus (STN), but this procedure carries substantial risks including intracranial hemorrhage and infection. Herein, we report an ultrasound-driven piezoelect

Ventromedial hypothalamus IL-6 signaling mediates the excessive aggressive behavior induced by late-gestational sleep deprivation in male mice.

BACKGROUND: Aggressive behavior is an evolutionarily conserved trait that supports resource acquisition and survival; however, excessive or dysregulated aggression is closely associated with neuropsychiatric disorders and social dysfunction. Maternal sleep deprivation (SD) during late gestation is a common yet underexplored environmental stressor that may disrupt offspring neurodevelopment through immune-mediated pathways. The present study investigated whether SD during late pregnancy alters aggressive behavior in offspring during early adulthood, explored the underlying neuroimmune mechanism

Estrogen receptor-α signaling in tanycytes lies at the crossroads of fertility and metabolism.

BACKGROUND: Estrogen secretion by the ovaries regulates the hypothalamic-pituitary-gonadal axis during the reproductive cycle, influencing gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) secretion, and also plays a role in regulating metabolism. Here, we establish that hypothalamic tanycytes-specialized glia lining the floor and walls of the third ventricle-integrate estrogenic feedback signals from the gonads and couple reproduction with metabolism by relaying this information to orexigenic neuropeptide Y (NPY) neurons. METHODS: Using mouse models, including mice floxed fo

α-Synuclein aggregation decreases cortico-amygdala connectivity and impairs social behavior in mice.

Abnormal accumulation of insoluble α-synuclein (α-Syn) inclusions in neurons, neurites, and glial cells is the defining neuropathology of synucleinopathies, including Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy. Accumulation of α-Syn inclusions in the amygdala has been well-documented in post-mortem studies of PD and DLB brains, as well as preclinical animal models of these conditions. Though α-Syn pathology is closely associated with neurodegeneration, there is a poor correlation between neuronal loss in the amygdala and the clinical features of PD a

Interictal spikes in Alzheimer's disease: Preclinical evidence for dominance of the dentate gyrus and cholinergic control by the medial septum.

Interictal spikes (IIS) are a common type of abnormal electrical activity in Alzheimer's disease (AD) and preclinical models. The brain regions where IIS are largest are not known but are important because such data would suggest sites that contribute to IIS generation. Because hippocampus and cortex exhibit altered excitability in AD models, we asked which areas dominate the activity during IIS along the cortical-CA1-dentate gyrus (DG) dorso-ventral axis. Because medial septal (MS) cholinergic neurons are overactive when IIS typically occur, we also tested the novel hypothesis that silencing

Dysfunction of the Hippocampal-Lateral Septal Circuit Impairs Risk Assessment in Epileptic Mice.

Temporal lobe epilepsy, a chronic disease of the brain characterized by degeneration of the hippocampus, has impaired risk assessment. Risk assessment is vital for survival in complex environments with potential threats. However, the underlying mechanisms remain largely unknown. The intricate balance of gene regulation and expression across different brain regions is related to the structure and function of specific neuron subtypes. In particular, excitation/inhibition imbalance caused by hyperexcitability of glutamatergic neurons and/or dysfunction of GABAergic neurons, have been implicated i

Activity of Tachykinin1-Expressing Pet1 Raphe Neurons Modulates the Respiratory Chemoreflex.

Homeostatic control of breathing, heart rate, and body temperature relies on circuits within the brainstem modulated by the neurotransmitter serotonin (5-HT). Mounting evidence points to specialized neuronal subtypes within the serotonergic neuronal system, borne out in functional studies, for the modulation of distinct facets of homeostasis. Such functional differences, read out at the organismal level, are likely subserved by differences among 5-HT neuron subtypes at the cellular and molecular levels, including differences in the capacity to coexpress other neurotransmitters such as glutamat

The central melanocortin system regulates growth hormone secretion.

The central melanocortin system is crucial for controlling food intake and metabolism. It includes neurons that produce pro-opiomelanocortin (POMC), a prohormone that generates the melanocortin receptor (MCR) agonist alpha-melanocyte-stimulating hormone (α-MSH), and neurons that produce agouti-related protein (AgRP), an endogenous MCR antagonist. This system also influences neuroendocrine neurons, thereby impacting pituitary hormone secretion. However, it remains unclear whether the central melanocortin system also modulates growth hormone (GH) secretion. Here we showed that arcuate nucleus (A

Impaired WNT3/IGF-1 Signaling in Dorsal Dentate Gyrus Contributes to Chronic Pain-Related Cognitive Impairment.

BACKGROUND: Neuropathic pain is frequently accompanied by cognitive deficits, but the neural circuits and molecular mechanisms linking nociceptive hypersensitivity to cognitive dysfunction remain incompletely understood. The hippocampal dentate gyrus (DG), a critical hub for adult neurogenesis and memory encoding, is emerging as a key substrate integrating pain processing and cognitive impairment, but how peripheral nerve injury disrupts DG homeostasis remains unclear. METHODS: In this study, we used the spared nerve injury (SNI) mouse model to investigate the cellular and molecular mechanism
