Salicylsalicylic acid (SS) and salicylic acid (SA) both appear in plasma after the oral ingestion of the former. They can be estimated in the presence of each other after extraction into dichloromethane using high pressure liquid chromatography. SS is unstable in plasma, being converted to SA in vitro. Conversion can be prevented by using ethylene diaminetetraacetic (EDTA) as an anticoagulant and carrying out the extraction in the cold. An aliquot of the extract is dried and dissolved in methanol. Mengovirus, extracted from infected L-cell cultures with Freon 113 and concentrated from the aqueous phase with polyethyleneglycol, was chromatographed on protein-coated controlled pore glass (CPG). The covalent binding of protein to CPG is described. Further purification and concentration of mengovirus was achieved by isopycnic density gradient centrifugation in solutions of either CsCl or salts of iodinated benzoic acid derivatives. A fast and sensitive method by high performance liquid chromatography for the quantification of the methylxanthine metabolites of caffeine in human plasma is described. Plasma is extracted with 20% isopropanol in chloroform and the evaporated residue is redissolved and chromatographed on silica gel using dichloromethane containing 2.2% of a solution of methanol-ammonium formateformic acid (100:0.02:0.017). A simple method is described that permits the direct quantitative determination of carbon disulphide, free diethyldithiocarbamate and disulphides derived from disulfiram in 1 ml of a patient's blood. It is based on a gas chromatographic determination of carbon disulphide produced from diethyldithiocarbamate and disulfiram using the head-space technique and a flame-photometric detector. The method is compared with a recently described spectrophotometric method.

Methotrexate in human plasma at a concentration as low as 0.01 microgram/ml can be assayed with the use of high-pressure-liquid chromatography and a fluorescence detection system. Methotrexate is oxidized stoichiometrically to 2,4-diaminopteridine-6-carboxylic acid, a fluorescent product that is separable from other fluorescent materials in plasma with the use of an octadecylsilane (reversed phase) column. Samples containing 10-150 nmoles of monohexoslceramides are benzoylated by heating for 60 min at 60 degree C in 0.5 ml of 10% (v/v) Benzoyl chloride in pyridine.
