Pharmacological control of ciliary activity in the young sea urchin larva: chemical studies on the role of cyclic nucleotides.

Distinct peaks in cAMP and cGMP content during early development, partly opposite to each other, may be correlated with the two main phases of gastrulation and ciliary activity. Monoamines increases cAMP formation. A transient or extended decrease follows, presumably reflecting some feedback mechanism. Muscarinic agents and Ca2+ interfere. The developmental variation in cyclic nucleotides may reflect a temporal shift in the role of various signal substances as well as feedback regulation related to Ca2+ influx. The opposite changes in cAMP and cGMP during early gastrulation may reflect a mutua

Concentration of 3', 5' cyclic adenosine monophosphate in ventricular cerebrospinal fluid of patients with prolonged coma after head trauma or intracranial hemorrhage.

A previous study showed that cerebrospinal fluid from the lateral ventricle of patients without disturbance of sensorium or intracranial pressure contains 15 to 30 nm 3', 5' cyclic adenosine monophosphate. We measured the concentration of this cyclic nucleotide by radioimmunoassay in cerebrospinal fluid from the lateral ventricle of six patients with prolonged coma (20 days or longer) after head trauma (four), or spontaneous intracranial hemorrhage (two). Coma was graded IV to I in order of decreasing severity. Fluid was removed at intervals of six to 72 hours from a Rickham reservoir placed i

Effects of arecoline and cholinesterase inhibitors on cyclic guanosine 3',5'-monophosphate and adenosine 3'.5'-monophosphate in mouse brain.

In mice, Arecoline in vivo dose-dependently increased the cGMP concentrations of the cerebellum and the "cereberum" (= parts of cortex, hippocampus, hypothalamus, thalamus, striatum and midbrain) without influencing thecAMP levels. The cholinesterase inhibitors paraoxon and physostigmine caused an elevation only in "cerebrum", whereas the cGMP content of the cerebellum even decreased. Pretreatment with atropine prevented the rise in cGMP levels as well as the symptoms of cholinergic stimulation elicited by arecoline or paraoxon. Diazepam reduced cGMP levels below control values and blocked the

Targeting the cGAS-STING Pathway for Cancer Immunotherapy: From Small-Molecule Agonists to Advanced Nanomaterials.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) signaling pathway has emerged as a pivotal mediator of antitumor immunity, bridging innate and adaptive immune responses through its ability to sense cytoplasmic DNA and orchestrate cytokine production. This review comprehensively explores recent advancements in targeting this pathway for cancer immunotherapy, focusing on two key strategies: small-molecule agonists and nanomaterial-based therapeutics. Small-molecule agonists, including cyclic dinucleotide (CDN) derivatives and non-nucleotide compounds, have been engineered…

Aggressive behavior increases after termination of chronic sildenafil treatment in mice.

Recent reports to the U.S. Food and Drug Administration Adverse Event Reporting System implicate sildenafil citrate in adverse emotional and aggressive behaviors. Sildenafil citrate (Viagra) is widely prescribed for erectile dysfunction and acts by inhibiting phosphodiesterase Type-5, resulting in accumulation of cyclic-guanosine monophosphate (cGMP). Cyclic-GMP is synthesized by guanylyl cyclase that is directly activated by the messenger molecule, nitric oxide (NO), formed throughout the CNS by the enzyme nitric oxide synthase (NOS). Elevated concentrations of cGMP have been associated with

Targeting PKGIα Cys42 attenuates cardiac dysfunction in heart failure with preserved ejection fraction.

Heart failure with preserved ejection fraction (HFpEF) is a highly prevalent condition associated with substantial morbidity and mortality, yet effective therapeutic options remain limited. As oxidation of Cys42 in cyclic guanosine monophosphate (cGMP)-dependent protein kinase Iα (PKGIα) can enhance vessel and diastolic relaxation, processes impaired in HFpEF, we sought to target this mechanism using natural compounds with predicted thiol reactivity. Among these, urolithin A emerged as a compound with a previously unidentified and counterintuitive mode of action, directly modifying Cys42 in PK

2-Phenyl-3-(Phenylselanyl)Benzofuran As a Promising Antidepressant Candidate: Mechanistic Insights Into Nitrergic Modulation and Subchronic Efficacy and Safety Profiling.

The compound 2-phenyl-3-(phenylselanyl)benzofuran (SeBZF1), a selenium-containing molecule based on a benzofuran scaffold, has shown antidepressant-like effects. This study investigated the involvement of the nitric oxide (NO)-cyclic guanosine monophosphate (cGMP) pathway, the efficacy of subchronic treatment in male Swiss mice, and computational analyses. Acute SeBZF1 administration (50 mg/kg, i.g.) showed antidepressant-like effects in the tail suspension test (TST). These effects were reversed by pretreatment with L-arginine (a NO precursor, 750 mg/kg, intraperitoneal, i.p.) or sildenafil (

Retinopathy-associated inosine monophosphate dehydrogenase 1 mutations cause metabolic and filament defects in cones.

Dominant variants in inosine monophosphate dehydrogenase 1 (IMPDH1), a key enzyme in the de novo synthesis of purine bases, cause progressive photoreceptor death, leading to blindness. To investigate the cause of degeneration, we generated the first mutant IMPDH1 animal models and expressed mutant forms of impdh1a in zebrafish cone photoreceptors. Unlike cones expressing exogenous normal impdh1a, cones containing impdh1a with the K238E mutation degenerated. Cones expressing impdh1a with the D226N mutation did not show significant cone loss by 2 years. Steady-state and flux metabolomics in zebr

Nitric oxide promotes rapid development of motility to accelerate biofilm dispersal in Vibrio cholerae.

Bacterial biofilms are resilient multicellular communities that underlie persistent infections and environmental survival. Dispersal from biofilms is a pivotal event for transmission and pathogenesis, yet the host signals and bacterial mechanisms orchestrating this transition remain poorly understood. Here, we show that nitric oxide (NO), a ubiquitous host-derived signaling molecule, acts as a rapid trigger for biofilm dispersal in Vibrio cholerae, a highly motile gram-negative bacterium and the etiologic agent of cholera, by promoting the development of motility. NO exposure induces broad upr

Polymorphism detection of PRKG2 gene and its association with the number of thoracolumbar vertebrae and carcass traits in Dezhou donkey.

BACKGROUND: Previous studies have shown that the protein kinase cGMP-dependent 2 (PRKG2) gene is associated with dwarfism in humans, dogo Argentines, and Angus cattle, as well as with height and osteoblastogenesis in humans. Therefore, the PRKG2 gene was used as the target gene to explore whether this gene is associated with several thoracolumbar vertebrae and carcass traits in Dezhou donkeys. RESULTS: In this study, fifteen SNPs were identified by targeted sequencing, all of which were located in introns of the PRKG2 gene. Association analysis illustrated that the g.162153251 G > A, g.162156

Soluble Guanylyl Cyclase Activators-Promising Therapeutic Option in the Pharmacotherapy of Heart Failure and Pulmonary Hypertension.

Endogenous nitric oxide (NO)-dependent vascular relaxation plays a leading role in the homeostasis of the cardiovascular, pulmonary, and vascular systems and organs, such as the kidneys, brain, and liver. The mechanism of the intracellular action of NO in blood vessels involves the stimulation of the activity of the soluble cytosolic form of guanylyl cyclase (soluble guanylyl cyclase, sGC), increasing the level of cyclic 3'-5'-guanosine monophosphate (cGMP) in smooth muscle and subsequent vasodilation. In recent years, a new group of drugs, soluble guanylyl cyclase stimulators, has found its w

MANP (M-Atrial Natriuretic Peptide) Reduces Blood Pressure and Furosemide-Induced Increase in Aldosterone in Hypertension.

BACKGROUND: cGMP MANP (M-atrial natriuretic peptide) is a best-in-class activator of the pGC-A (particulate guanylyl cyclase A) receptor. Furosemide increases the effectiveness of antihypertensive agents, but activates renin-angiotensin-aldosterone system. We aimed to investigate for the first time cardiorenal and neurohumoral actions of MANP in a genetic model of hypertension in spontaneously hypertensive rats. We also assessed how MANP would potentiate the blood pressure (BP)-lowering actions of furosemide while reducing the production of aldosterone. METHODS: Spontaneously hypertensive rat

The interdependent relationship between the nitric oxide signaling pathway and primary cilia in pulse electromagnetic field-stimulated osteoblastic differentiation.

Pulsed electromagnetic fields (PEMFs) have long been recognized being safe and effective in treating bone fracture nonunion and osteoporosis. However, the mechanism of osteogenic action of PEMFs is still unclear. While primary cilia are reported to be a sensory organelle for PEMFs, and nitric oxide (NO) plays an indispensable role in osteogenic effect of PEMFs, the relationship between NO and primary cilia is unknown. In this study, effects of treatment with 50 Hz 0.6 mT PEMFs on osteogenic differentiation and mineralization, NO secretion, and ciliary location of specific proteins were examine

cGMP homeostasis in malaria parasites-The key to perceiving and integrating environmental changes during transmission to the mosquito.

Malaria-causing parasites are transmitted from humans to mosquitoes when developmentally arrested gametocytes are taken up by a female Anopheles during a blood meal. The changes in environment from human to mosquito activate gametogenesis, including a drop in temperature, a rise in pH, and a mosquito-derived molecule, xanthurenic acid. Signaling receptors have not been identified in malaria parasites but mounting evidence indicates that cGMP homeostasis is key to sensing extracellular cues in gametocytes. Low levels of cGMP maintained by phosphodiesterases prevent precocious activation of game

Chloroquine-induced scratching is mediated by NO/cGMP pathway in mice.

Chloroquine (CQ), a 4-aminoquinoline drug, has long been used in the treatment and prevention of malaria. However its side effect generalized pruritus contributes to treatment failures, and consequently results in the development of chloroquine resistant strains of Plasmodium falciparum. It was proposed that the administration of CQ correlated with increase in nitric oxide (NO) production. Nitric oxide is involved in some pruritic disorders such as atopic dermatitis, psoriasis and scratching behavior evoked by pruritogens like substance P. Therefore, the aim of this study was to investigate th

Pharmacotherapy of the overactive bladder.

Lower urinary tract symptoms (LUTS), the overactive bladder syndrome (OAB), and detrusor overactivity (DO) are all conditions that can have major effects on quality of life and social functioning. Antimuscarinic drugs are first-line treatment--they often have good initial response rates, but adverse effects and decreasing efficacy cause long-term compliance problems, and alternatives are needed. The recognition of the functional contribution of the urothelium, the spontaneous myocyte activity during bladder filling, and the diversity of nerve transmitters involved has sparked interest in both

Crystallization of cyclic nucleotide phosphodiesterases.

Selective inhibitors of cyclic nucleotide phosphodiesterases (PDEs) have been widely studied as therapeutic agents for the treatment of various human diseases. Three-dimensional structures are essential for the design of highly selective inhibitors, but their availability is limited by the speed of crystallization. We describe crystallization of the catalytic domains of the unligated PDE4B2B, rolipram-bound PDE4D2, and 3-isobutyl-1-methylxanthine-bound PDE5A1 using the methods of vapor diffusion and microdialysis. We also briefly describe general methods of protein crystallization to provide a
