Linking tissues to phenotypes using gene expression profiles.

Despite great biological and computational efforts to determine the genetic causes underlying human heritable diseases, approximately half (3500) of these diseases are still without an identified genetic cause. Model organism studies allow the targeted modification of the genome and can help with the identification of genetic causes for human diseases. Targeted modifications have led to a vast amount of model organism data. However, these data are scattered across different databases, preventing an integrated view and missing out on contextual information. Once we are able to combine all the e

Description and primary results from an audiometric study of male twins.

OBJECTIVE: The purpose of this study is to present descriptive results from an audiometric examination of a male twin sample, compare the values with normative databases, and explore whether there are age differences in genetic and environmental contributions to variation in hearing. DESIGN: Audiometric and questionnaire data were collected on a subsample of male twins who were identified through the population-based Swedish Twin Registry. Hearing examinations were completed for a total of 557 intact pairs, comprised of 250 identical (monozygotic [MZ] pairs and 307 fraternal (dizygotic [DZ] p

Targeted Therapies for Metabolic Myopathies Related to Glycogen Storage and Lipid Metabolism: a Systematic Review and Steps Towards a 'Treatabolome'.

BACKGROUND: Metabolic myopathies are a heterogenous group of muscle diseases typically characterized by exercise intolerance, myalgia and progressive muscle weakness. Effective treatments for some of these diseases are available, but while our understanding of the pathogenesis of metabolic myopathies related to glycogen storage, lipid metabolism and β-oxidation is well established, evidence linking treatments with the precise causative genetic defect is lacking. OBJECTIVE: The objective of this study was to collate all published evidence on pharmacological therapies for the aforementioned met

Pharmacogenomic landscape of a Moroccan cohort: enhancing global diversity and addressing the North African gap.

Pharmacogenomics (PGx) enables personalized therapy by considering individual genetic variation. However, North African populations remain underrepresented in global PGx databases despite their substantial and unique genetic diversity, which can strongly influence drug response. This study characterizes the pharmacogenomic landscape of 109 Moroccan individuals using whole-genome sequencing data. Across 8289 candidate pharmacogenes, we identified 129,021 novel variants, predominantly within DPYD, CYP2C19, and SLCO1B1, underscoring the region's considerable genomic diversity. Analysis of 17 clin

A dataset of patients with isolated and syndromic optic neuropathies linked to RTN4IP1 genetic variants.

Biallelic pathogenic variants of the Reticulon 4 interacting protein 1 (RTN4IP1) gene are responsible for optic atrophy, either isolated or associated with ataxia, mental retardation, and seizures. They are identified as a cause of hereditary optic neuropathy in 7% of patients diagnosed before the age of 20. We have built a dataset for this gene by collating all the clinical cases available in the literature, and unpublished patients diagnosed at our centre, using standard nomenclature to describe both the molecular and phenotypic features. We performed a comprehensive data analysis, based on

X-linked hypophosphatemic rickets: a new mutation.

Phosphopenic rickets may be caused by mutations in the PHEX gene (phosphate regulating endopeptidase homolog X-linked). Presently, more than 500 mutations in the PHEX gene have been found to cause hypophosphatemic rickets. The authors report a clinical case of a 4-year-old girl with unremarkable family history, who presented with failure to thrive and bowing of the legs. Laboratory tests showed hypophosphatemia, elevated alkaline phosphatase, normal calcium, mildly elevated PTH and normal levels of 25(OH)D and 1.25(OH)D. The radiological study showed bone deformities of the radius and femur. C

A historical, systematic review of handedness origins.

Handedness ontogenesis is still under debate in science. This systematic review analyzed articles regarding the theories and basis of handedness formation, highlighting the historical knowledge path that this literature underwent. Cochrane Library, LILACS, Web of Sciences, Science Direct and PubMed databases were searched. This review included review studies with handedness as the main topic. Only papers written in English with analyses exclusively in neurotypical humans (any age range) were included. Different approaches (genetic, neural, social, and behavioural) were reviewed in light of gro

ENPP1 deficiency: A clinical update on the relevance of individual variants using a locus-specific patient database.

Loss-of-function variants in the ectonucleotide pyrophosphatase/phosphodiesterase family member 1 (ENPP1) cause ENPP1 Deficiency, a rare disorder characterized by pathological calcification, neointimal proliferation, and impaired bone mineralization. The consequence of ENPP1 Deficiency is a broad range of age dependent symptoms and morbidities including cardiovascular complications and 50% mortality in infants, autosomal recessive hypophosphatemic rickets type 2 (ARHR2) in children, and joint pain, osteomalacia and enthesopathies in adults. Recent research continues to add to the growing clini

Novel compound heterozygous GFPT1 mutations in a family with limb-girdle myasthenia with tubular aggregates.

Limb-girdle myasthenia with tubular aggregates, a subtype of congenital myasthenic syndrome, is an extremely rare autosomal recessive genetic disease characterized by prominent limb-girdle weakness and good response to acetylcholinesterase inhibitor therapy. Herein, we reported two novel mutations of GFPT1 gene in a Chinese pedigree. Two siblings presented with fatigue, weakness of limb-girdle and decrement of the muscle action potential with repetitive nerve stimulation. Thus, myasthenia gravis was initially suspected, but anti-AChR antibodies were negative. Two novel missense mutations (p.Ly

Structure-constrained combination-based nonlinear association analysis between incomplete multimodal imaging and genetic data for biomarker detection of neurodegenerative diseases.

Multimodal imaging data are widely applied in imaging genetic studies to identify associations between imaging and genetic data for the biomarker detection of neurodegenerative diseases (NDs). However, the incomplete multimodal imaging data and complex relationships among imaging and genetic data make it difficult to effectively analyze associations between imaging and genetic data and accurately detect disease-related biomarkers. This study proposed a novel structure-constrained combination-based nonlinear association analysis method to exploit associations between incomplete multimodal imagi

Optical genome mapping uncovers clinically relevant structural variants in congenital heart disease with heterotaxy.

INTRODUCTION: The genetic factors underlying congenital heart disease and heterotaxy (CHD/HTX) are complex, including copy number variants, loss-of-function mutations, and missense variants, many of which can be detected by high-throughput sequencing. The screening for chromosomal structural variations (SVs) is another important strategy to understand the genetic etiology of CHD/HTX. METHODS: We employed optical genome mapping (OGM), an innovative technique capable of capturing SVs often missed by traditional cytogenetic methods, to screen for SVs in 12 patients with complex CHD/HTX. Several
