Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide, largely due to its chronic progression and frequent diagnosis at advanced stages. Although surgical resection remains the gold standard for CRC treatment, adjuvant chemotherapy with 5-fluorouracil (5FU) is commonly required; however, its clinical effectiveness is limited by systemic toxicity and severe side effects. In this study, we evaluated the in vivo performance of a bacterial nanocellulose (BNC)-based 5FU encapsulation system in a murine CRC model induced by azoxymethane/dextran sulfate sodium (AOM/DSS). The healing of diabetic wounds is impeded by a pathological triad of recalcitrant bacterial infections, oxidative stress, and chronic inflammation. While photothermal therapy (PTT) is a promising non-antibiotic alternative, its clinical translation is constrained by a critical efficacy-biosafety paradox: high-temperature ablation (>60 °C) induces collateral tissue damage, whereas biosafe mild-temperature PTT (<50 °C) fails to achieve complete pathogen eradication due to bacterial heat-shock defenses. To reconcile this, we engineered a multifunctional hybrid hydrogel by incorporating flower-lik

Site-specific colon drug delivery is crucial in the treatment of colon-localized diseases. In this study, prodrugs of 5-aminosalicylic acid (5-ASA), the drug of choice in ulcerative colitis (UC), were synthesized by conjugating with chitosan having molecular weights (MW) of 30, 80 and 300 kDa to achieve targeted delivery to the colon. FT-IR and UV-Vis analyses confirmed the successful synthesis and varying loading capacities of 5-ASA, with a maximum loading capacity of 11.32 ± 2.0 %. The obtained conjugates (7-9) exhibited gradual drug release characteristics, with up to 30 % of the drug conte

Dextran 40, a typical high molecular weight carbohydrate drug refined through fermentation, is widely used in the clinical field in an injectable form. The final product obtained through fermentation may contain by-products such as fructose and residual sucrose, which carry a risk of adverse reactions. Current quality standards do not effectively control for the possible presence of carbohydrate residues, and methods for detecting such residues are lacking. This gap in quality control exists in the vast majority of existing carbohydrate drugs. This study established and compared liquid chromat

Ulcerative colitis, a chronic inflammatory condition affecting the rectum and colon to varying degrees, is linked to a dysregulated immune response and the microbiota. Sodium (aS,9R)-3-hydroxy-16,17-dimethoxy-15-oxidotricyclo[12.3.1.12,6]nonadeca-1(18),2,4,6(19),14,16-hexene-9-yl sulfate hydrate (SDH) emerges as a novel diarylheptane compound aimed at treating inflammatory bowel diseases. However, the mechanisms by which SDH modulates these conditions remain largely unknown. In this study, we assessed SDH's impact on the clinical progression of dextran sodium sulfate (DSS)-induced ulcerative c

In this study, a whey protein isolate (WPI)-chitooligosaccharide (COS) stabilized bitter melon (Momordica charantia L.) seed oil emulsions (WC-BSOE) were prepared using the electrostatic layer-by-layer self-assembly technique, and their modulating effects on ulcerative colitis (UC) were investigated in dextran sulfate sodium (DSS)-induced UC mice model. The stability and releasing ability of WC-BSOE under simulated gastrointestinal digestion condition and their acute toxicity were also investigated. The results showed that WC-BSOE was stable to droplet aggregation in the simulated gastric and

In this study, we investigated the regulatory role of curcumin in the differentiation of intestinal stem cells (ISCs) in dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) model mice and explored whether this effect was mediated by the Wnt/β-catenin signaling pathway. We conducted experiments in DSS-induced UC model mice to observe changes in intestinal morphology through HE staining and detect the expression of key proteins in the Wnt/β-catenin signaling pathway. According to these findings, curcumin was found to have a significant impact on the differentiation of ISCs. These result

An excessive inflammatory response of the gastrointestinal tract is recognized as one of the major contributors to ulcerative colitis (UC). Despite this, effective preventive approaches for UC remain limited. Rosmarinic acid (RA), an enriched fraction from Perilla frutescens, has been shown to exert beneficial effects on disease-related inflammatory disorders. However, RA-enriched perilla seed meal (RAPSM) and perilla seed (RAPS) extracts have not been investigated in dextran sulfate sodium (DSS)-induced UC in mice. RAPSM and RAPS were extracted using the solvent-partitioning method and analyz

Sea cucumber peptides have been proven to exhibit a variety of biological activities. Ulcerative colitis (UC) is a chronic disease characterized by diffuse inflammation of the mucosa of the rectum and colon with increasing incidence and long duration, and is difficult to cure. The effect of sea cucumber peptide on UC is currently unknown. In this study, 1.5% dextran sulfate sodium (DSS) was added to the drinking water of mice to induce a UC model, and the daily doses of sea cucumber peptide (SP) solution of 200 mg/kg·BW, 500 mg/kg·BW, and 1000 mg/kg·BW were given to UC mice to detect the relie

PURPOSE: Inflammatory bowel diseases (IBDs) are global health problems that are associated with immune regulation, but clinical IBDs treatment is currently inadequate. Effective preventive or therapeutic methods for immune disorders rely on controlling the function of immune cells. Isosteviol sodium (STV-Na) has antioxidant activity, but the therapeutic effect of STV-Na against IBD remain undocumented. Herein, we investigated the therapeutic effect of STV-Na in mice models with IBDs.
METHODS: Mice received 3.5% DSS for 7 days to establish IBD models. Intraperitoneal STV-Na was given 2 days be

BACKGROUND AND AIMS: 1.4-Dihydroxy-2-naphthoic acid (DHNA), a bifidogenic growth stimulator from Propionibacterium freudenreichii, is thought to have a beneficial effect as a prebiotic; however, its in vivo effect on intestinal inflammation remains unknown. The aim of this study was to determine whether oral administration of DHNA can ameliorate dextran sodium sulphate (DSS) induced colitis and to determine the possible underlying mechanisms.
METHOD: Colitis was induced in mice by treatment with 2.0% DSS for seven days. DHNA (0.6 or 2.0 mg/kg) was given in drinking water prior to (preventive

Twenty healthy pigs weighing 12-17 kg were anesthetized and the small intestines were exteriorized into saline-moistened gauze. During a 2-h period 4% of the animals' body weight was bled through an arterial cannula in six increments and replaced immediately by the fluid tested: hydroxyethyl starch 120 (HES, Plasmafusin, Orion Corp., Mw 120,000), dextran 70 (DEX) and Ringer's acetate (RA). The amount of fluid infused for replacement of blood loss was equal to the amount of blood withdrawn in the colloid groups but fourfold in the RA group. Five non-bled pigs served as controls. After the hemod

Etoposide (ETP), a Biopharmaceutics Classification System class IV drug with poor aqueous solubility, demonstrates limited therapeutic efficacy against colorectal cancer (CRC) because of inferior absorption and off-target effects. To deliver drugs specifically to cancer cells that overexpress CD44, this study developed hyaluronic acid (HA)-functionalized dextran (DEX) polymeric nanoparticles (ETP-DEX-HA-NPs). Optimised nanoparticles (174.7 ± 3.2 nm, -12.83 ± 1.1 mV) demonstrated significant entrapment efficiency (62.75 ± 2.32%) and drug loading (55.64 ± 3.86%), with partial amorphization valid

The development of inflammatory bowel diseases is postulated to be driven by the interaction between genetic susceptibility and environmental factors, resulting in proinflammatory cytokine responses to intestinal dysbiosis. Loss-of-function mutations in the nucleotide-binding oligomerization domain 2 gene (NOD2) are the strongest risk factor for Crohn's disease (CD); however, environmental factors affecting CD development have been poorly defined in patients with NOD2 mutations. In this study, we investigated whether high-fat diet (HFD), one of the possible environmental risk factors for CD, a

BACKGROUND AND PURPOSE: Ulcerative colitis (UC) is a chronic, nonspecific inflammatory bowel disease. The kynurenine pathway (KP) of tryptophan metabolism has been implicated in several autoimmune diseases, including multiple sclerosis and rheumatoid arthritis. However, little is known about the role of KP in UC. This study investigated the role of tryptophan 2,3-dioxygenase (TDO2), a key rate-limiting enzyme of KP, in UC.
EXPERIMENTAL APPROACH: Dextran sodium sulphate (DSS)- and 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis models were used to evaluate the effects of TDO2 defici

BACKGROUND: Ulcerative colitis (UC) arises from complex interactions among genetic predisposition, environmental factors, and immune dysregulation. Promotion of M1-polarized macrophages critically contribute to intestinal inflammation and subsequent tissue damage. Baicalin, a bioactive flavonoid isolated from the roots of Scutellaria baicalensis, exhibits potent anti-inflammatory effects, yet its therapeutic efficacy and regulatory mechanisms of macrophages in the development of UC remain poorly understood.
PURPOSE: This study aimed to investigate the protective effects of baicalin in a murin
