Nephrogenic adenofibroma. A novel kidney tumor of young people.

Nephrogenic adenofibroma is a novel kidney tumor of young people (mean age of presentation, 13 years), who present with polycythemia, hypertension, or hematuria, which resolve following nephrectomy. The typical nephrectomy specimen contains a solitary, nonencapsulated, vaguely circumscribed, irregularly shaped or spherical, firm mass with either tan, gray-white, or pale yellow coloration. Cysts are sometimes present within the tumor. The histologic appearance is distinctive and characterized by a marked proliferation of spindled mesenchymal cells resembling the classical type of congenital mes

Germline duplication of MYCN predisposes to childhood embryonal tumours.

BACKGROUND: Neuroblastoma and Wilms tumour (WT) are common childhood embryonal malignancies. Germline 2p24 duplication has been reported in several cases of neuroblastoma and WT, either as part of a larger 2p duplication or as a microduplication involving just 2p24.3. Although the larger duplications involve many genes, including ALK, the microduplications have been localised to a region including MYCN and DDX1. METHODS: We analysed Whole Genome Sequence data from adults and children sequenced for various indications. We utilised a workflow to extract structural and copy number variants, filt

Transcriptional Heterogeneity Reveals a Synaptic Gene Program in Developing and Adult Human Oligodendrocyte Precursor Cells.

Human oligodendrocyte precursor cells (OPCs) arise early in gestation and expand broadly during cortical development, yet the extent of their heterogeneity remains poorly defined. Here, we isolated >2,300 highly pure OPCs from post-conceptional week (PCW) 17 human cortex using an optimized PDGFRα-based immunopanning and performed single-cell RNA sequencing. Unsupervised clustering revealed four transcriptionally distinct embryonic OPC subsets, including a previously unrecognized population that expressed genes linked to synaptic development, synaptic signaling, and neuromodulation. This subset

High sperm DNA fragmentation increased embryo aneuploidy rate in patients undergoing preimplantation genetic testing.

RESEARCH QUESTION: Is high sperm DNA fragmentation (SDF) associated with a high embryonic aneuploidy rate in patients undergoing intracytoplasmic sperm injection (ICSI)-preimplantation genetic testing (PGT)? DESIGN: This was a retrospective study of 426 couples with normal karyotypes undergoing ICSI-PGT at the authors' centre from March 2017 to March 2021. SDF was assessed using the sperm chromatin structure assay. The population was divided into low and high SDF groups according to cut-off values found by the receiver operating characteristic (ROC) curve. A 1:1 ratio propensity score matchin

Notochord manipulation does not impact oesophageal and tracheal formation from isolated foregut in 3D explant culture.

BACKGROUND: Tracheo-oesophageal malformations result from disturbed foregut separation during early development. The notochord, a specialised embryonic structure, forms immediately adjacent to the dividing foregut. In the Adriamycin mouse model of oesophageal atresia, foregut and notochord abnormalities co-exist, and the site and severity of foregut malformations closely correlate to the position and extent of the notochord defects. Notochord and foregut abnormalities also co-exist in the Noggin Knockout mouse as well in a small number of human cases. The notochord is a source of powerful mole

[Morphogenesis, structure and properties of lymphatic vessels].

In this paper, we present literature results related to structure and various manners of lymphatic vessel formation during embryonic development and in pathological events, such as tumorigenesis, wound healing, and other diseases. The functions of the lymphatic system include the collection of fluids that enter tissues from the circulation, absorption of lipids and lipid-soluble vitamins from the intestine and their subsequent transport, participation in antigen, dendritic cell, and lymphocyte migration. The lymphatic system is also a route for tumor cell and inflammatory cell transport. Nativ

Anatomical Considerations of Embryology and Development of the Musculoskeletal System: Basic Notions for Musculoskeletal Radiologists.

The musculoskeletal (MSK) system begins to form in the third week of intrauterine development. Multiple genes are involved in the complex different processes to form the skeleton, muscles and joints. The embryonic period, from the third to the eighth week of development, is critical for normal development and therefore the time when most structural defects are induced. Many of these defects have a genetic origin, but environmental factors may also play a very important role. This review summarizes the embryology of the different components of the MSK system and their configuration as an organ-

Borrelidin analogues with antimalarial activity: design, synthesis and biological evaluation against Plasmodium falciparum parasites.

Borrelidin, a structurally unique 18-membered macrolide, was found to express antimalarial activity against drug-resistant Plasmodium falciparum malaria parasites, with IC50 value of 0.93 ng/mL. However, it also displays strong cytotoxicity against human diploid embryonic MRC-5 cells. To investigate the issue of the cytotoxicity of borrelidin, borrelidin-based analogues were synthesized and their anti-Plasmodium properties were evaluated. In this communication, we report that a novel borrelidin analogue, bearing the CH2SPh moiety via a triazole linkage, was found to retain a potent antimalaria

[Histological and ultrastructural characteristics of primary malignant liver tumors in children].

Histological and electron microscopic studies of primary malignant liver tumors in children established the differences in the morphological composition of the epithelial and mixed variants of hepatoblastoma. The epithelial component is represented by embryonal and fetal cells at different levels of ultrastructural and functional differentiation. The mixed variant of hepatoblastoma is characterized by islets of stellate and lymphocyte-like cell capable of differentiation into epithelial and mesenchymal components. The latter does not have a regular composition and may include cells differentia

A novel mutation of HOXA10 in a Chinese woman with a Mullerian duct anomaly.

BACKGROUND: Müllerian duct anomalies consist of a set of congenital structural malformations that occur when the Müllerian ducts do not develop properly during embryonic life. Their molecular genetic basis is poorly understood. METHODS: In this study, we conducted mutation analysis of the HOXA10 gene in a cohort of 109 Chinese women with Müllerian duct anomalies. RESULTS: We identified a novel mutation (Y57C) in one patient with a didelphic uterus. The mutation affected the transcriptional regulation capacity of HOXA10. CONCLUSIONS: Our study showed that mutation of HOXA10 gene may contribu

Neuroendocrine neoplasms of the mediastinum.

Neuroendocrine neoplasms of the mediastinum form part of a family of tumors characterized by genotypic, immunophenotypic, and functional properties of neuroendocrine differentiation. Although rare, these tumors have been the source of much attention and controversy in the literature. Their nomenclature and classification, in particular, have continued to evolve over the years. Such tumors comprise lesions derived from neuroendocrine elements within the thymus, from paraganglionic rests, or from misplaced embryonal structures within the mediastinum. The most common neuroendocrine neoplasms of t

Phylo- and -ontogenetically determined local changes in the structure of the arterial wall of vertebrates. Macroscopical findings.

It has been shown, by means of comparative anatomical, phylo- and -ontogenetic studies, that the focal structural changes occuring at seven points in the wall of the aortic arch and brachiocephalic arteries of reptiles, birds, and mammals, and in human-beinmnants of certain vessels which have regressed during embryonic development. They are considered to be loci minoris resistentiae and might therefore represent a site of predilection for pathological lesions, such as atherosclerosis. The existence of these "scars" in human-beings and birds has already been described. This paper presents defin

Evolutionary origins of the reptilian brain: the question of putative homologues of dorsal ventricular ridge. An overview and proposal.

The reptilian brain is characterized by a structure that bulges into the lateral ventricle, called dorsal ventricular ridge (DVR). The DVR was originally considered to be a part of the basal ganglia, although more recent studies indicate that it may correspond to the dorsal part of the hemisphere. The anterior portion of the DVR has several connectional and functional similarities with parts of the mammalian neocortex, for which reason it has been claimed that the two structures can be considered as homologues. In this article I review the evidence supporting and refuting homology of the DVR w

Intraventricular twin fetuses in fetu.

The authors report a rare case of multiple intracranial fetuses in fetu, fulfilling Willis' traditional criteria, which include an axial and appendicular skeleton with surrounding organized tissue. This case was ascertained from studies of a full-term female neonate who presented with ventriculomegaly. A CT scan showed intracranial calcifications that were suggestive of an axial skeleton. Her birth weight was 3.176 kg (50th-75th percentile), length was 52 cm (90th percentile), head circumference was 35 cm (50th-75th percentile), and Apgar scores were 7 and 8 at 1 and 5 minutes, respectively. Oral Organoids: Progress and Challenges.

Oral organoids are complex 3-dimensional structures that develop from stem cells or organ-specific progenitors through a process of self-organization and re-create architectures and functionalities similar to in vivo organs and tissues in the oral and maxillofacial region. Recently, striking advancements have been made in the construction and application of oral organoids of the tooth, salivary gland, and tongue. Dental epithelial and mesenchymal cells isolated from tooth germs or derived from pluripotent stem cells could generate tooth germ-like organoids by self-organization in a specific cu

Identification of pregnancy-associated plasma protein A as a migration-promoting gene in malignant pleural mesothelioma cells: a potential therapeutic target.

UNLABELLED: From the crude ethanol extract of Erythrina caffra leaves, a new compound, stigmasta-5,22-diene 3-O-(6'-tridecanoate)-β-D-glucopyranoside (1), along with stigmasta-5,22-diene (2), lutein (3), and triacontanol (4), was isolated. Their structures were elucidated using spectroscopic (1D and 2D NMR) and spectrometric [High-Resolution Electrospray Ionization Mass Spectrometry, (HRESIMS)] analyses. RP-LC-MS/MS profiling of the ethyl acetate and methanol fractions further identified 22 secondary metabolites, whose proposed structures were consistent with reported fragmentation patterns an

[Regeneration of hair cells and auditory neurons in the ear].

UNLABELLED: Most of deafness have a neuro-sensory origin and are characterized by a loss of hair cells and auditory neurons in the spiral ganglion. At the moment, hearing aids are the only treatment available. To restore hearing in a patient suffering from deafness, it is necessary to study the mechanisms that might lead to the regeneration of neurosensory structures of the inner ear. HAIR CELL REGENERATION: The production of hair cells and supporting cells is terminated during embryonic development of the cochlea in mammals. However, several recent arguments suggest that new hair cells can b

Human MD-2 discrimination of meningococcal lipid A structures and activation of TLR4.

MD-2, a eukaryotic accessory protein, is an essential component for the molecular pattern recognition of bacterial endotoxins. MD-2 interacts with lipid A of endotoxins [lipopolysaccharide (LPS) or lipooligosaccharide (LOS)] to activate human toll-like receptor (TLR) 4. The structure of lipid A influences the subsequent activation of human TLR4 and the immune response, but the basis for the discrimination of lipid A structures is unclear. A recombinant human MD-2 (rMD-2) protein was produced in the Pichia pastoris yeast expression system. Human embryonic kidney (HEK293) cells were transfected

Cell-autonomous and nonautonomous actions of endothelin-A receptor signaling in craniofacial and cardiovascular development.

Craniofacial and cardiac development relies on the proper patterning of the neural crest-derived ectomesenchyme of the pharyngeal arches, from which many craniofacial and great vessel structures arise. One of the intercellular signaling molecules that is involved in this process, endothelin-1 (ET-1), is expressed in the arch epithelium and influences arch development by binding to its cognate receptor, the endothelin A (ET(A)) receptor, found on ectomesenchymal cells. We have previously shown that absence of ET(A) signaling in ET(A)(-/-) mouse embryos disrupts neural crest cell development, re

A descriptive study of early eyelid morphogenesis in a large-scale human embryo collection.

INTRODUCTION: Human eyelid development begins during the embryonic period. Although the general outline of early eyelid development has been described, many structures and developmental phenomena remain insufficiently investigated or controversial mainly due to the scarcity of human fetal specimens. The purpose of this study is to clarify these unresolved issues and to determine the precise developmental timing of each eyelid structure. METHODS: Fifty-six embryos (Carnegie stage (CS) 18-23) and six fetuses (early 9th week of development) from the Kyoto Collection were included in this study.
