Independent origins of plant parasitism in Clavicipitaceae: phylogeny, divergence, and taxonomic revision.

The family Clavicipitaceae (Hypocreales, Ascomycota) includes a wide range of fungal lifestyles and ecological interactions, but the phylogenetic relationships and evolutionary origins of its plant-parasitic taxa have remained controversial. To establish a robust, phylogenetically consistent classification, molecular phylogenetic, fossil-calibrated divergence-time, and likelihood-based ancestral state reconstruction analyses were performed using a multi-locus nuclear dataset. These analyses identified two deeply divergent, monophyletic clades of plant parasites: designated as Clavicipitoideae,

Multiomics analysis of the effects of manure-borne doxycycline combined with oversized fiber microplastics on pak choi growth and the risk of antibiotic resistance gene transmission.

In this study, oversized microplastics (OMPs) were intentionally introduced into soil containing manure-borne doxycycline (DOX). This strategic approach was used to systematically examine the effects of combined OMP and DOX pollution on the growth of pak choi, analyze alterations in soil environmental metabolites, and explore the potential migration of antibiotic resistance genes (ARGs). The results revealed a more pronounced impact of DOX than of OMPs. Slender-fiber OMPs (SF OMPs) had a more substantial influence on the growth of pak choi than did coarse-fiber OMPs (CF OMPs). Conversely, CF O

A new compound, phomaherbarine A, induces cytolytic reactivation in epstein-barr virus-positive B cell lines.

Epstein-Barr virus (EBV), the first virus found to induce cancer in humans, has been frequently detected in various types of B cell lymphomas. During its latent phase, EBV expresses a limited set of proteins crucial for its persistence. Induction of the lytic phase of EBV has shown promise in the treatment of EBV-associated malignancies. The present study assessed the ability of phomaherbarine A, a novel compound derived from the endophytic fungus Phoma herbarum DBE-M1, to stimulate lytic replication of EBV in B95-8 cells. Phomaherbarine A was found to efficiently initiate the expression of bo

Phytoremediation of trichloroethylene in the soil/groundwater environment: Progress, problems, and potential.

Trichloroethylene (TCE) poses a significant environmental threat in groundwater and soil, necessitating effective remediation strategies. Phytoremediation offers a cost-effective and environmentally friendly approach to remediation. However, the mechanisms governing plant uptake, volatilisation, and degradation of TCE remain poorly understood. This review explores the mechanisms of TCE phytoremediation, metabolic pathways, and influencing factors, emphasizing future research directions to improve the understanding of TCE phytoremediation. The results showed that although the proportion of TCE

Screening and characterization of biocontrol bacteria isolated from Ageratum conyzoides against Collectotrichum fructicola causing Chinese plum (Prunus salicina Lindl.) anthracnose.

Chinese plum (Prunus salicina Lindl.) is a nutritionally and economically important stone fruit widely grown around the world. Anthracnose, caused by Collectotrichum spp., is one of the primary biotic stress factors limiting plum production. Medicinal plants may harbor rhizospheric or endophytic microorganisms that produce bioactive metabolites that can be used as anthracnose biocontrol agents. Here, 27 bacterial isolates from the medicinal plant A. conyzoides with diverse antagonistic activities against C. fructicola were screened. Based on morphological, physiological, biochemical, and molec

Biomechanics of the parasite-host interaction of the European mistletoe.

The European mistletoe (Viscum album) is an epiphytic hemiparasite that attaches to its host by an endophytic system. Two aspects are essential for its survival: the structural integrity of the host-parasite interface must be maintained during host growth and the functional integrity of the interface must be maintained during ontogeny and under mechanical stress. We investigated the mechanical properties of the mistletoe-host interaction. Intact and sliced mistletoe-host samples, with host wood as reference, were subjected to tensile tests up to failure. We quantified the rough fractured surfa

Contiguous verrucous proliferations in syringocystadenoma papilliferum: A retrospective analysis with additional evaluation via mutation-specific BRAFV600E immunohistochemistry.

BACKGROUND: Syringocystadenoma papilliferum (SCAP) is an uncommon cutaneous adnexal proliferation. There have been several reports describing collision lesions of SCAP and verruca, although little is known about the frequency of this association. Molecular testing has revealed the BRAFV600E mutation in a large proportion of SCAP cases, although its expression pattern has not been previously evaluated. METHODS: In this retrospective analysis, we explored the potential histopathological association between verruca and SCAP. We also evaluated mutation-specific BRAFV600E expression in these lesio

Synergistic and drug-resistant reversing effects of diorcinol D combined with fluconazole against Candida albicans.

The emergence of drug resistance and limitation of antifungal agents bring enormous challenges to the management of Candida infection. Combination therapy is a useful way to treat the fungal infection, especially for those difficult-to-treat infections. In this research, we investigated the interaction effects between diorcinol D (DD), an antifungal active natural product from lichen endophytic fungus and fluconazole (FLC) against planktonic cells and mature biofilms of Candida albicans in vitro by checkerboard microdilution and time-killing tests. Both fractional inhibitory concentration inde

Evidence of dehydration and electrolyte disturbances in cases of perennial ryegrass toxicosis in Australian sheep.

CASE REPORT: Perennial ryegrass toxicosis (PRGT) is a common disease entity in Australia, presenting as an association of clinical signs including alterations in normal behavioural, ataxia ('staggers'), ill thrift and gastrointestinal dysfunction ('scours'). Clinical signs can range in severity from mild (gait abnormalities and failure to thrive) to severe (seizures, lateral recumbency and death). Presentation across the flock is usually highly variable. PRGT is caused by toxins produced by the endophytic fungus Neotyphodium lolii, a symbiont of perennial ryegrass that is present in pastures a

Urothelial carcinoma of the upper urinary tract: inverted growth pattern is predictive of microsatellite instability.

Urothelial carcinoma of the renal pelvis and ureter may develop as a manifestation of hereditary nonpolyposis colorectal cancer syndrome (HNPCC), a disorder characterized by mutation or inactivation of a number of DNA mismatch repair genes and detectable as microsatellite instability (MSI). Some urothelial carcinomas display areas of endophytic, or inverted, growth. In this study, urothelial cancers of the upper urinary tract (n = 132) from patients treated at 2 tertiary care centers were studied to identify an association between growth pattern and MSI. Thirty-five neoplasms were microsatelli

Eukaryotic metagenome-assembled genomes recovered from deep metagenomic sequencing of the seagrass, Zostera marina, include a novel chytrid in the order Lobulomycetales.

Fungi play pivotal roles in terrestrial ecosystems as decomposers, pathogens and endophytes, yet their significance in marine environments is often understudied. Seagrasses, as globally distributed marine flowering plants, have critical ecological functions but knowledge about their associated fungal communities remains relatively limited. Previous amplicon surveys of the fungal community associated with the seagrass, Zostera marina, have revealed an abundance of potentially novel chytrids. In this study, we employed deep metagenomic sequencing to extract metagenome-assembled genomes (MAGs) fr…

Boereguasins A-D, Cytochalasins from Boeremia exigua with Differential MAPK/NF-κB Inhibition.

Four unprecedented cytochalasins, boereguasins A-D (1-4, respectively), were purified from the endophytic fungus Boeremia exigua. Structurally, compound 1 contains an adenosine moiety and features a novel bridged 5/6/13/5/6/5 ring system. Compound 2 represents the first cytochalasin with an odd-numbered carbon chain (19 carbons), which is presumably assembled via the fungal polyketide synthase pathway from an unusual propionyl-CoA starter unit. Compound 3 incorporates an unusual 15-membered carbocyclic ring, while compound 4 possesses a novel 5/6/14/6 tetracyclic scaffold. The structures were

Additions to Diaporthe (Sordariomycetes, Diaporthaceae) and Neptunomyces (Dothideomycetes, Didymosphaeriaceae) species associated with Itea in Thailand.

Itea is an economically and medicinally important plant, widely distributed across China, Japan and Southeast Asia, noted for producing rare sugars. Despite this, endophytic fungi associated with this plant remain poorly studied. This study focuses on the taxonomy and phylogeny of fungi isolated from healthy tissues of Itea japonica and I. riparia in Northern Thailand. By incorporating morphology, multiloci phylogenetic analyses and nucleotide polymorphism comparison, we establish two novel species, Diaporthe iteae (Diaporthaceae, Sordariomycetes) and Neptunomyces iteae (Didymosphaeriaceae, Do

Taxonomy Identification and Phytotoxic Activities of Pectolytic Bacteria Isolated from Diseased Plants of Phalaenopsis Blume (Orchidaceae).

Orchid plants, due to their high aesthetic qualities of large inflorescences, long flowering period, and ease of care, have high commercial potential; however, when grown industrially in factories, they are susceptible to infectious diseases. In this study, we isolated from Phalaenopsis spp. plants epiphytic, rhizospheric, and endophytic bacteria associated with soft rot symptoms. Twenty-nine isolates exhibiting pectolytic activity were identified as strains of the genera Bacillus, Klebsiella, Microbacterium, Paenibacillus, Paracidovorax, Pseudomonas, and Psychrobacillus based on 16S rRNA anal

Exploring the antifungal activity of the wheat microbiome.

Wheat (Triticum aestivum) is a cornerstone of global food security, yet its productivity is threatened by fungal pathogens such as Fusarium graminearum, Zymoseptoria tritici, and Rhizoctonia solani. These pathogens not only reduce yield but can also compromise grain quality and food safety through the production of mycotoxins. Traditional control strategies, primarily reliant on chemical fungicides and resistant cultivars, face limitations due to evolving pathogen resistance, environmental concerns, and regulatory restrictions. In recent years, the plant microbiome has emerged as a promising f

Dabrafenib-Induced Cutaneous Eruptions in a Patient With Langerhans Cell Histiocytosis.

Langerhans cell histiocytosis (LCH) is a rare malignancy marked by clonal proliferation of Langerhans cells, with BRAF V600E mutations identified in over 50% of the cases. BRAF inhibitors (BRAFi), such as dabrafenib, have shown efficacy in treating BRAF V600E-mutant LCH. However, BRAFi therapy is associated with cutaneous adverse effects, including the development of new melanocytic nevi, verrucae, and keratinocyte carcinoma (KC). This case report describes a 78-year-old woman with BRAF V600E-mutant LCH who developed multiple dermatologic side effects following dabrafenib salvage therapy. Seve

Multiple Hypertrophic Lichen Planus Mimicking Squamous Cell Carcinoma: A Case Report.

INTRODUCTION: Hypertrophic lichen planus (HLP) with pseudoepitheliomatous hyperplasia (PEH) can closely mimic well-differentiated squamous cell carcinoma (SCC), creating a clinically important diagnostic pitfall. Fewer than 26 case reports in the literature describe HLP initially misdiagnosed as SCC, underscoring the need for careful clinicopathologic correlation. CASE PRESENTATION: A 77-year-old woman developed multiple hyperkeratotic nodules and plaques on both legs and feet over 4 months. A punch biopsy led to an initial diagnosis of well-differentiated SCC. An additional elliptical incisi
