Acute induction of anxiety in humans by delta-9-tetrahydrocannabinol related to amygdalar cannabinoid-1 (CB1) receptors.

Use of Cannabis, the most widely used illicit drug worldwide, is associated with acute anxiety, and anxiety disorders following regular use. The precise neural and receptor basis of these effects have not been tested in man. Employing a combination of functional MRI (fMRI) and positron emission tomography (PET), we investigated whether the effects of delta-9-tetrahydrocannabinol (delta-9-THC), the main psychoactive ingredient of cannabis, on anxiety and on amygdala response while processing fearful stimuli were related to local availability of its main central molecular target, cannabinoid-1 (

Altered regional homogeneity in patients with late monocular blindness: a resting-state functional MRI study.

Many previous studies have demonstrated that the blindness patients have has functional and anatomical abnormalities in the visual and other vision-related cortex. However, changes in the brain function in late monocular blindness (MB) at rest are largely unknown. In this study, we investigated the underlying regional homogeneity (ReHo) of brain-activity abnormalities in patients with late MB and their relationship with clinical features. A total of 32 patients with MB (25 male and seven female) and 32 healthy controls (HCs) (25 male and seven female) closely matched in age, sex, and education

Attention and spatial cognition: Neural and anatomical substrates of visual neglect.

Unilateral spatial neglect (USN) is a neurological disorder often observed following damage to the right cerebral hemisphere. Patients with USN are no longer able to take into account stimuli presented on the left side of space. In this article, we will discuss the neuroanatomical correlates that underlie visuospatial attention and can cause USN, an area of growing research interest in the past 20 years. This syndrome has often been related to cortical damage, notably in the inferior parietal lobule. Other data have also implicated lesions in the inferior frontal gyrus or the superior temporal

Is amyloid-β harmful to the brain? Insights from human imaging studies.

Although the amyloid-β protein associated with the Alzheimer's disease plaque has been detectable in living people for over a decade, its importance in the pathogenesis of Alzheimer's disease is still debated. The frequent presence of amyloid-β in the brains of cognitively healthy older people has been interpreted as evidence against a causative role. If amyloid-β is crucial to the development of Alzheimer's disease, it should be associated with other Alzheimer's disease-like neurological changes. This review examines whether amyloid-β is associated with other biomarkers indicative of early Al

Frequency-specific alternations in the amplitude of low-frequency fluctuations in chronic tinnitus.

Tinnitus, a phantom ringing, buzzing, or hissing sensation with potentially debilitating consequences, is thought to arise from aberrant spontaneous neural activity at one or more sites within the central nervous system; however, the location and specific features of these oscillations are poorly understood with respect to specific tinnitus features. Recent resting-state functional magnetic resonance imaging (fMRI) studies suggest that aberrant fluctuations in spontaneous low-frequency oscillations (LFO) of the blood oxygen level-dependent (BOLD) signal may be an important factor in chronic ti

Widespread hypermetabolism in symptomatic and asymptomatic episodes in Kleine-Levin syndrome.

BACKGROUND: No reliable biomarkers are identified in KLS. However, few functional neuroimaging studies suggested hypoactivity in thalamic and hypothalamic regions during symptomatic episodes. Here, we investigated relative changes in regional brain metabolism in Kleine-Levin syndrome (KLS) during symptomatic episodes and asymptomatic periods, as compared to healthy controls. METHODS: Four drug-free male patients with typical KLS and 15 healthy controls were included. 18-F-fluorodeoxy glucose positron emission tomography (PET) was obtained in baseline condition in all participants, and during …

Chronic neuronopathic type of Gaucher's disease with progressive myoclonic epilepsy in the absence of visceromegaly and bone involvement.

INTRODUCTION: Gaucher's disease is a lysosomal storage disorder caused by the deficiency of glucocerebrosidase. Gaucher's disease has three clinical types: non-neuronopathic (Type 1), Acute Neuropathic (Type 2) and chronic neuronopathic (Type 3). The chronic neuronopathic (Type 3) is characterised by a variety of disease variants with onset in childhood with hepatomegaly, skeletal lesions and later slow horizontal saccades, treatment-resistant generalised tonic-clonic and myoclonic seizures, dementia, progressive spasticity, cognitive deterioration, ataxia and death in the second or third deca

Role of the ventral striatum in developing anorexia nervosa.

Functional imaging data in adult patients with anorexia nervosa (AN) support a dysfunctional signal in the ventral striatum as neural signature of AN. In the present study, development of this signal was investigated with the prediction that a characteristic pattern of ventral-striatal signalling will be shown in response to cues associated with food restriction that reflects the evolvement of starvation dependence over time. The signal was assessed in adolescent patients with AN, whose duration of illness was about five times shorter relative to the adult sample. During functional magnetic re

Knowledge-driven binning approach for rare variant association analysis: application to neuroimaging biomarkers in Alzheimer's disease.

BACKGROUND: Rapid advancement of next generation sequencing technologies such as whole genome sequencing (WGS) has facilitated the search for genetic factors that influence disease risk in the field of human genetics. To identify rare variants associated with human diseases or traits, an efficient genome-wide binning approach is needed. In this study we developed a novel biological knowledge-based binning approach for rare-variant association analysis and then applied the approach to structural neuroimaging endophenotypes related to late-onset Alzheimer's disease (LOAD). METHODS: For rare-var

Cross-Disease Classification of Depression, Bipolar Disorder, and Schizophrenia Using Multiscale Spatiotemporal Network Fusion Features From fNIRS.

BACKGROUND: Major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia (SCZ) are highly prevalent and disabling psychiatric conditions with increasing incidence, significantly affecting individuals' daily functioning, social relationships, and overall quality of life. Current diagnostic practices largely rely on clinical interviews and symptom-based rating scales, which are inherently subjective and time-consuming-posing major obstacles to large-scale screening. METHODS: To address this, we propose a novel multiscale spatiotemporal network fusion (MSNF) feature extraction metho

A Neural Network Approach to Identify Left-Right Orientation of Anatomical Brain MRI.

PURPOSE: This study presents a novel application of deep learning to enhance the accuracy of left-right orientation identification in anatomical brain MRI scans. Left-right orientation misidentification in brain MRIs presents significant challenges due to several factors, including metadata loss or ambiguity, which often occurs during the de-identification of medical images for research, conversion between image formats, software operations that strip or overwrite metadata, and the use of older imaging systems that stored orientation differently. METHOD: A three-dimensional convolutional neur

Motor Cortex Activation During Writing in Focal Upper-Limb Dystonia: An fNIRS Study.

BACKGROUND: Functional imaging studies have associated dystonia with abnormal activation in motor and sensory brain regions. Commonly used techniques such as functional magnetic resonance imaging impose physical constraints, limiting the experimental paradigms. Functional near-infrared spectroscopy (fNIRS) offers a new noninvasive possibility for investigating cortical areas and the neural correlates of complex motor behaviors in unconstrained settings. METHODS: We compared the cortical brain activation of patients with focal upper-limb dystonia and controls during the writing task under natu

Impairments of large-scale functional networks in attention-deficit/hyperactivity disorder: a meta-analysis of resting-state functional connectivity.

Altered resting-state functional connectivity (rsFC) has been noted in large-scale functional networks in attention-deficit/hyperactivity disorder (ADHD). However, identifying consistent abnormalities of functional networks is difficult due to varied methods and results across studies. To integrate rsFC alterations and search for coherent patterns of intrinsic functional network impairments in ADHD, this research conducts a coordinate-based meta-analysis of voxel-wise seed-based rsFC studies comparing rsFC between ADHD patients and healthy controls. A total of 25 datasets from 21 studies inclu

Win for your kin: Neural responses to personal and vicarious rewards when mothers win for their adolescent children.

Mother-child relationships change considerably in adolescence, but it is not yet understood how mothers experience vicarious rewards for their adolescent children. In the current study, we investigated neural responses of twenty mothers winning and losing money for their best friend and for their adolescent child in a gambling task. During the task, functional neuroimaging data were acquired. We examined the activation patterns when playing for or winning for self, adolescent children and friends in four a-priori selected ROIs (nucleus accumbens, dorsomedial prefrontal cortex, precuneus and te…

The rabbit as a behavioral model system for magnetic resonance imaging.

BACKGROUND: fMRI requires that subjects not move during image acquisition. This has been achieved by instructing people not to move, or by anesthetizing experimental animal subjects to induce immobility. We have demonstrated that a surgically implanted headbolt onto the skull of a rabbit allows their brain to be imaged comfortably while the animal is awake. This article provides a detailed method for the preparation. NEW METHOD: We took advantage of the rabbit's tolerance for restraint to image the brain while holding the head at the standard stereotaxic angle. Visual stimulation was produced
