The intake of oleic acid-rich fats, a hallmark of the Mediterranean diet, has well-documented beneficial effects on human metabolic health. One of the key mechanisms underlying these effects is the regulation of gut microbiota structure and function. However, most existing studies focus on gut bacteria, while gut fungi, as a vital component of the gut microbiota, remain largely unexplored. This study compared the effects of regular peanut oil (PO) and high-oleic acid peanut oil (HOPO) on the gut mycobiome and serum metabolome employing ITS high-throughput sequencing and UPLC-MS/MS metabolomics

Because data on the fungal gut community structure of African children are scarce, we aimed to describe it by reanalysing rRNA ITS1 and ITS2 metabarcoding data from a study designed to assess the influence of microbiota in malaria susceptibility in Malian children from the Dogon country. More specifically, we aimed to establish the core gut mycobiome and compare the gut fungal community structure of breastfed children, aged 0-2 years, with other age groups. Briefly, DNA was extracted from 296 children's stool samples. Both rRNA ITS1 and ITS2 genomic barcodes were amplified and subjected to Ill

The morphological diversity of insect mouthparts is closely related to changes in food sources and diets. Research into the structures of insect mouthparts may help to establish a fundamental basis for a better understanding of insect feeding mechanisms. In this study, we examined the fine morphology of the mouthparts of Illeis chinensis using scanning electron microscopy. We paid particular attention to the types, quantities, and distribution of sensilla on the mouthparts. Our results showed that the basic components of the mouthparts of I. chinensis are the same as those in other lady beetle

Marine environments are underexplored terrains containing fungi that produce a diversity of natural products given unique environmental pressures and nutrients. While bacteria are commonly the most studied microorganism for natural products in the marine world, marine fungi are also abundant but remain an untapped source of bioactive metabolites. Given that their terrestrial counterparts have been a source of many blockbuster antitumor agents and anti-infectives, including camptothecin, the penicillins, and cyclosporin A, marine fungi also have the potential to produce new chemical scaffolds a

Anaerobic fungi, first described in 1975 by Orpin, live in close contact with bacteria and other microorganisms in the rumen and caecum of herbivorous animals, where they digest ingested plant food. Seventeen distinct anaerobic fungi belonging to five different genera have been described. They have been found in at least 50 different herbivorous animals. Anaerobic fungi do not possess mitochondria, but instead have hydrogenosomes, which form hydrogen and carbon dioxide from pyruvate and malate during fermentation of carbohydrates. In addition, they are very oxygen- and temperature-sensitive, a

Fungi produce numerous low molecular weight molecules endowed with a multitude of biological activities. However, mining the full-genome sequences of fungi indicates that their potential to produce secondary metabolites is greatly underestimated. Because most of the biosynthesis gene clusters are silent under laboratory conditions, one of the major challenges is to understand the physiological conditions under which these genes are activated. Thus, we cocultivated the important model fungus Aspergillus nidulans with a collection of 58 soil-dwelling actinomycetes. By microarray analyses of both

The objective of this study was to investigate the antifungal activity of the vanillin-derived Schiff base complexed with zinc (II) (SBZ) against fluconazole-resistant strains of Candida spp. and Cryptococcus neoformans. The broth microdilution assay was performed to determine the minimum inhibitory concentration (MIC). The checkerboard assay was used to evaluate the type of pharmacological interaction of the combination of SBZ with azoles. Flow cytometry and alkaline comet assays were performed to investigate the possible action mechanism of SBZ against Candida albicans and Cryptococcus neofo

The functional lactone 2-Methyl-2-propanyl-7-oxo-1,4-oxazepane-4-carboxylate (BocOP), containing a protected secondary amino group, is synthesized through oxone-mediated Baeyer-Villiger oxidation. After that, the homopolymerization kinetics of BocOP catalyzed by immobilized Candida antarctia lipase B (Novozym 435, N435) are investigated. The N435 can efficiently catalyze the conversion of BocOP into poly(β-amino ester) (PAE). Then, the N435 is subsequently employed to synthesize copolymers of BocOP and ε-caprolactone (CL) using methoxypolyethylene glycols (mPEG) as initiators. The study shows

Candida albicans is a major cause of systemic candidiasis, a severe fungal infection with a ∼40% mortality rate. Yck2, a casein kinase 1 (CK1) in C. albicans, is targeted by antifungal inhibitors YK-I-02 (YK) and MN-I-157 (MN). Using multiplexed inhibitor beads and mass spectrometry (MIB/MS), the selectivity of these inhibitors was determined across the fungal kinome. The MIB matrix captured 89% of C. albicans protein kinases, revealing that YK and MN selectively engage three CK1 homologues (Yck2, Yck22, and Hrr25) and a human p38α homologue (Hog1). Chemoproteomics using a custom MN-kinobead c

l-Lysine oxidase (LysOX) is a FAD-dependent homodimeric enzyme that catalyzes the oxidative deamination of l-lysine to produce α-keto-ε-aminocaproate with ammonia and hydrogen peroxide. LysOX shows strict substrate specificity for l-lysine, whereas most l-amino acid oxidases (LAAOs) exhibit broad substrate specificity for l-amino acids. Previous studies of LysOX showed that overall structural similarity to the well-studied snake venom LAAOs. However, the molecular mechanism of strict specificity for l-lysine was still unclear. We here determined the structure of LysOX in complex with l-lysine

The incidence and prevalence of serious fungal infections is rising, especially in immunosuppressed individuals. Moreover, co-administration of antibiotics and immunosuppressants has driven the emergence of new multidrug-resistant pathogens. The significant increase of multidrug-resistant pathogens, together with their ability to form biofilms, is associated with morbidity and mortality. Research on novel synthetically prepared immunomodulators as potential antifungal immunotherapeutics is of serious interest. Our study demonstrated the immunobiological activity of synthetically prepared bioti

A natural furan derivative was isolated from the methanolic extract of the fruit bodies of Fomitiporia ellipsoidea. Its chemical structure was elucidated as methyl 3,5-dioxo- 1,3,5,7-tetrahydrobenzo[1,2-c:4,5-c']difuran-4-carboxylate by means of extensive NMR and MS data analysis, and named as fomitiporiaester A (1). Compound 1 showed significant antitumor activity to hepatoma H(22) in vivo, and the inhibition rates were 42.94%, 49.17%, and 58.15% at concentrations of 5, 10, and 20 mg/kg, respectively. Compound 1 showed weak cytotoxic activities against the human hepatoblastoma (HepG-2) and hu

Alpha-synuclein (alpha-syn), a protein implicated in Parkinson's disease, is structurally diverse. In addition to its random-coil state, alpha-syn can adopt an alpha-helical structure upon lipid membrane binding or a beta-sheet structure upon aggregation. We used yeast biology and in vitro biochemistry to detect how sequence changes alter the structural propensity of alpha-syn. The N-terminus of the protein, which adopts an alpha-helical conformation upon lipid binding, is essential for membrane binding in yeast, and variants that are more prone to forming an alpha-helical structure in vitro a

alpha-Sarcin, mitogillin, and restrictocin are small (approximately 17 kDa) basic robosome-inactivating proteins (RIPs) produced by the Aspergilli that catalytically inactivate the large ribosomal subunits of all organisms tested to date. These three fungal ribotoxins act as specific ribonucleases by hydrolyzing one single phosphodiester bond in the universally conserved alpha-sarcin domain of 23-28S rRNAs and are among the most potent inhibitors of protein synthesis known. Previous molecular studies of ribotoxins indicated that they belong to the superfamily of ribonucleases and analysis of t

Multitrophic interactions are essential for understanding the mechanisms that sustain the local coexistence of diverse soil organisms. Although stable-isotope evidence has revealed clear trophic niche differentiation among mites, co-occurrence network patterns and assembly mechanisms across trophic groups remain poorly understood. Here, we conducted a comprehensive survey of soil mites across four forest types along an elevational gradient in the Shibing World Natural Heritage Site. We aimed to elucidate the community assembly processes and ecological network complexity of predators, fungal fe

Recently, the occurrence of candidiasis has increased dramatically, especially in immunocompromised patients. Additionally, their treatment is often ineffective due to the resistance of yeasts to antimycotics. Therefore, there is a need to search for new antifungals. A series of nine newly synthesized thiazole derivatives containing the cyclopropane system, showing promising activity against Candida spp., has been further investigated. We decided to verify their antifungal activity towards clinical Candida albicans isolated from the oral cavity of patients with hematological malignancies and i

Fungal-microbial interactions drive key ecosystem functions and offer vast, untapped potential for the bio-economy. However, translating these natural consortia into controllable applications is hindered by the structural and metabolic complexity of the interplay. This review navigates the complexity landscape of these interactions, examining the technological trajectory from the mechanistic foundation of 1:1 dialogue to the rational design of synthetic consortia. We discuss the transition from static cultivation toward advanced microfluidic ("fungi-on-a-chip") and biomimetic platforms that re

Dye-sensitized solar cells (DSSCs) using natural pigments offer a low-cost, non-toxic alternative to ruthenium dyes, but fungal sensitizers remain underexplored relative to plant-derived pigments. This study reports, for the first time, the phenoxazine pigments cinnabarin and cinnabaric acid from Pycnoporus sanguineus extract (PSE) as DSSC sensitizers, paired with a ZnTiO3/TiO2 heterojunction photoanode designed to suppress interfacial recombination. The extract was characterized by HPLC-DAD and FTIR, and the semiconductors ZnTiO3/TiO2 (ZTO) and TiO2 (TO) by XRD and SEM. Four device configurat

In disaster situations such as tsunamis, floods, and typhoons, fungal contamination can significantly intensify within 48 h of submersion, rapidly spreading and damaging the value and structural integrity of organic cultural heritage artifacts. Consequently, the application of radiation-based disinfection has been increasingly studied as an effective method for prompt and reliable treatment. In most cases, the fungal species were successfully eradicated at a radiation dose of 5 kGy (gamma rays). Nevertheless, certain fungal strains demonstrated high resistance and survived even at doses rangin
