Sex determination in the GIFT strain of tilapia is controlled by a locus in linkage group 23.

BACKGROUND: Tilapias (Family Cichlidae) are the second most important group of aquaculture species in the world. They have been the subject of much research on sex determination due to problems caused by early maturation in culture and their complex sex-determining systems. Different sex-determining loci (linkage group 1, 20 and 23) have been detected in various tilapia stocks. The 'genetically improved farmed tilapia' (GIFT) stock, founded from multiple Nile tilapia (Oreochromis niloticus) populations, with some likely to have been introgressed with O. mossambicus, is a key resource for tilap

A DNA polymorphism from five inbred strains of the mouse identifies a functional class of domesticus-type Y chromosome that produces the same phenotypic distribution of gonadal hermaphrodites.

The wild-derived CLA inbred strain of the house mouse contains a domesticus-type Y chromosome that lacks a 2.3-kb TaqI band with fragment 1 of the AC11 probe. The CLA Y chromosome also causes a low frequency of XY gonadal hermaphrodites when backcrossed to the C57BL/6J strain (F. G. Biddle and Y. Nishioka, 1988. Genome, 30:870-878). A similar domesticus-type Y chromosome, lacking the 2.3-kb TaqI band has now been found in the four historical inbred strains AKR/J, MA/MyJ, PL/J, and RF/J. When backcrossed to C57BL/6J, these four Y chromosomes cause low frequencies of gonadal hermaphrodites simil

Nix alone is sufficient to convert female Aedes aegypti into fertile males and myo-sex is needed for male flight.

A dominant male-determining locus (M-locus) establishes the male sex (M/m) in the yellow fever mosquito, Aedes aegyptiNix, a gene in the M-locus, was shown to be a male-determining factor (M factor) as somatic knockout of Nix led to feminized males (M/m) while transient expression of Nix resulted in partially masculinized females (m/m), with male reproductive organs but retained female antennae. It was not clear whether any of the other 29 genes in the 1.3-Mb M-locus are also needed for complete sex-conversion. Here, we report the generation of multiple transgenic lines that express Nix under

Whole-genome Resequencing Reveals the Sex Determination System and Develops a Sex-specific Marker in Fat Greenling (Hexagrammos otakii).

Teleost fishes display remarkable diversity in sex determination mechanisms, yet this aspect remains uninvestigated in the family Hexagrammidae. In this study, we performed whole-genome resequencing of Hexagrammos otakii to characterize its sex determination system. Genome-wide association studies and population differentiation index analysis identified chromosome 9 as the sex chromosome, supporting a male heterogametic (XY) system. A 3.11 Mb sex-determining region (SDR) was delineated, containing the potential candidate gene gdf6a. Comparative genomic analysis revealed that the H. otakii sex

Regulation of the amh gene promoter activity by SF-1 in sex-reversing Cheilinus undulatus and its adaptive evolution.

BACKGROUND: The humphead wrasse (Cheilinus undulatus) is a protogynous hermaphroditic fish and a top predator in coral reef ecosystems. However, overfishing has led to a significant decline in its population, making it challenging to study its adaptive evolution and sex reversal mechanisms. RESULTS: In this study, comparative genomics analyses revealed adaptive evolutionary signals associated with feeding capacity, digestive function, and color pattern transformation in C. undulatus. Notably, positive selection signals were detected in the key masculinization pathway gene amh in C. undulatus,

Evolution and regulation of animal sex chromosomes.

Animal sex chromosomes typically carry the upstream sex-determining gene that triggers testis or ovary development and, in some species, are regulated by global dosage compensation in response to functional decay of the Y chromosome. Despite the importance of these pathways, they exhibit striking differences across species, raising fundamental questions regarding the mechanisms underlying their evolutionary turnover. Recent studies of non-model organisms, including insects, reptiles and teleosts, have yielded a broad view of the diversity of sex chromosomes that challenges established theories

SRY+ Derivative X Chromosome in a Female With Apparently Typical Sexual Development.

BACKGROUND: When the SRY gene is present in a 46,XX fetus, some degree of testicular development is expected. Our laboratory performed prenatal genetic testing for a fetus that had screened positive for Y chromosome material by noninvasive prenatal screening (NIPS) but that had apparently typical female development by ultrasound imaging. The aim of this study was to determine the clinical relevance of the NIPS results. METHODS: We analyzed fetal material obtained via amniocentesis procedure by G-banding, microarray, and fluorescence in situ hybridization (FISH). Optical genome mapping (OGM) w

Methods for identifying and interpreting sex-linked SNP markers and carrying out sex assignment: application to thornback ray (Raja clavata).

Sex-determining modes remain unknown in numerous species, notably in fishes, in which a variety of modalities have been reported. Additionally, noninvasive individual sexing is problematic for species without external sex attributes or for early life stages, requiring cytogenetic or molecular analyses when sex chromosomes or sex-linked markers have been characterized. Genomics now provide a means to achieve this. Here, we review common sex-determination systems and corresponding statistical methods for identifying sex-linked genetic markers and their use for sex assignment, focusing on single

Gene Duplication, Translocation, and Molecular Evolution of Dmrt1 and Related Sex-Determining Genes in Anurans.

Sex determination, the developmental process that directs embryos toward male or female fates, is controlled by master sex-determining genes whose origins and evolutionary dynamics remain poorly understood outside of a few model systems. In contrast to the highly differentiated sex chromosomes of mammals, birds, and Drosophila, most anurans (frogs and toads) maintain homomorphic sex chromosomes that exhibit a rapid turnover, even among closely related species. Master sex-determining genes evolve via gene duplication or via allelic diversification, and sex chromosome turnover is driven by gene

A transgenic tool to assess Anopheles mating competitiveness in the field.

BACKGROUND: Malaria parasites, transmitted by the bite of an anopheline mosquito, pose an immense public health burden on many tropical and subtropical regions. The most important malaria vectors in sub-Saharan Africa are mosquitoes of the Anopheles gambiae complex including An. gambiae (sensu stricto). Given the increasing rates of insecticide resistance in these mosquitoes, alternative control strategies based on the release of genetically modified males are being evaluated to stop transmission by these disease vectors. These strategies rely on the mating competitiveness of release males, ho

46, XX man with SRY gene translocation: cytogenetic characteristics, clinical features and management.

This report describes a well-masculinized 33-year-old man with infertility and primary hypogonadism in whom chromosomal evaluation revealed a 46, XX karyotype. This syndrome is a rare but important cause of hypergonadotropic hypogonadism in which the diagnosis can be delayed or missed. A review of the cytogenetic basis and clinical features is presented to raise awareness of this entity among clinicians and to emphasize the importance of appropriate laboratory testing when indicated.

Multilevel transcriptional regulation of embryonic sex determination and dosage compensation by the X-signal element sex-1.

The Caenorhabditis elegans nuclear hormone receptor sex-1 is an embryonic X-signal element that represses xol-1, the sex-switch gene that is the master regulator of sex determination and dosage compensation. Several prior studies on sex-1 function have suggested that sex-1 may have additional downstream roles beyond the regulation of xol-1 expression. In this study, we characterize some of these additional roles of sex-1 in regulating the dual processes of sex determination and dosage compensation during embryogenesis. Our study reveals that sex-1 acts on many of the downstream targets of xol-

Development of a pheasant interspecies primordial germ cell transfer to chicken embryo: effect of donor cell sex on chimeric semen production.

This study was conducted to evaluate whether the sex of donor primordial germ cells (PGCs) influences production of chimeric semen from recipient hatchlings produced by interspecies transfer between pheasant (Phasianus colchicus) and chicken (Gallus gallus). Pheasant PGCs were retrieved from 7-d-old embryos and subsequently transferred into circulatory blood of 2.5-d-old (Stage 17) embryos. The sex of embryos was discerned 3 to 6 days after laying, and in preliminary study, overall rate of embryo survival after sexing was 74.6% with male-to-female ratio of 0.49 to 0.51. In Experiment 1, magnet

Brave new world?

Recent developments in biology and medicine are raising new problems in the prevention and treatment of birth defects, and in research on these diseases. The problems include immediate issues such as genetic counseling, abortion for birth defects, the withholding of complex treatments from individuals in some situations, screening for genetic and other diseases, artificial insemination, and fertilization in vitro. Other problems, such as the dysgenic effects of modern medicine and the possibilities of cloning and gene therapy, are more remote. Each of these issues should be considered on its o

Hormonal induction of supermale golden rosy barb and isolation of Y-chromosome specific markers.

Viable supermales of the golden rosy barb Puntius conchonius are generated through hormonal sex reversal and progeny testing. Discrete immersion (3 h/day on the second, fourth, and sixth day after hatching) of the fry in Estradiol-17beta (E2) at doses of 400-600 microg/L ensures greater than 40% survival and production of more than 98% F1 females; of these 50% are heterogametic females, that when crossed with normal males, sire 25% YY supermales. Supermales sire female progenies at the frequencies between 0 and 8%. Reproductive performance of hormonally sex-reversed females and androgenetic fe

Bridging Biology and Society: Molecular Insights into Social Determinants of Vascular Disease.

BACKGROUND: Vascular diseases (VDs)-such as arterial aneurysms, chronic venous disease, and peripheral artery disease-are influenced by a complex interplay of genetic, biological, and social factors. Integrating the assessment of both molecular and social determinants offers a novel framework that connects factors like socioeconomic status, chronic stress, and environmental exposures to the molecular pathways that drive vascular pathophysiology. METHODS: This review was conducted searching libraries such as Web of Science, Scopus, MEDLINE, and Google Scholar. The aim was to facilitate the ana

A drug-inducible sex-separation technique for insects.

Here, we describe a drug-inducible genetic system for insect sex-separation that demonstrates proof-of-principle for positive sex selection in D. melanogaster. The system exploits the toxicity of commonly used broad-spectrum antibiotics geneticin and puromycin to kill the non-rescued sex. Sex-specific rescue is achieved by inserting sex-specific introns into the coding sequences of antibiotic-resistance genes. When raised on geneticin-supplemented food, the sex-sorter line establishes 100% positive selection for female progeny, while the food supplemented with puromycin positively selects 100%
