Mitochondrial neurogastrointestinal encephalomyopathy in china: a novel TYMP variant and comprehensive clinical-genetic insights.

BACKGROUND: Mitochondrial neurogastrointestinal encephalopathy (MNGIE) is a rare autosomal recessive disorder caused by variants in the TYMP gene, which encodes thymidine phosphorylase (TP). It is characterized by multisystem involvement, with prominent gastrointestinal, neurological, and systemic manifestations that typically exhibit progressive worsening over time. METHODS: We characterized a multigenerational MNGIE family through comprehensive proband analysis, identifying compound heterozygous TYMP variants (c.131G > C, p.Arg44Pro and c.1268T>G, p.Leu423Arg in trans) as the molecular basi

Exploiting Natural Language Processing to Unveil Topics and Trends of Traumatic Brain Injury Research.

Traumatic brain injury (TBI) has evolved from a topic of relative obscurity to one of widespread scientific and lay interest. The scope and focus of TBI research have shifted, and research trends have changed in response to public and scientific interest. This study has two primary goals: first, to identify the predominant themes in TBI research; and second, to delineate "hot" and "cold" areas of interest by evaluating the current popularity or decline of these topics. Hot topics may be dwarfed in absolute numbers by other, larger TBI research areas but are rapidly gaining interest. Likewise,

Inferring relationships among major psychiatric disorders in a resting-state functional connectivity-informed embedding space.

Major neuropsychiatric disorders such as major depressive disorder (MDD) and schizophrenia (SCZ), as well as the neurodevelopmental disorder autism spectrum disorder (ASD), are traditionally treated as distinct clinical entities. However, genome-wide association studies indicate shared genetic risks, motivating a transdiagnostic view. Resting-state functional connectivity (rsFC) is a promising biomarker for these disorders, but its high dimensionality complicates inference of inter-disorder relationships in the native feature space. Here, we develop an rsFC-based embedding-relation workflow th

Shared genetic underpinnings of gray matter volume alterations and metabolic traits in major depressive disorder.

BACKGROUND: Major depressive disorder (MDD) is linked to extensive gray matter volume (GMV) reductions and frequently co-occurs with metabolic dysfunction. However, the shared genetic basis linking neurostructural abnormalities and metabolic traits remains poorly understood. METHODS: Using a coordinate-based meta-analytic framework, we synthesized findings from 57 voxel-based morphometry (VBM) studies to characterize GMV alterations in MDD. Spatial transcriptomic correlation analysis was performed using the Allen Human Brain Atlas to identify genes associated with these alterations. In parall

The Causal Relationship Between Asthma and Hippocampal Volume: A Study Based on Bidirectional Mendelian Randomization Analysis.

BACKGROUND: Asthma is a common chronic respiratory disease, and its potential association with the central nervous system has garnered increasing attention in recent years. While observational studies suggest that asthma may affect hippocampal structure and function through mechanisms such as chronic inflammation and hypoxia, its causal relationship remains unclear. METHODS: In this study, we employed a two-sample Mendelian randomization (MR) analysis, utilizing large-scale genome-wide association study (GWAS) data to systematically investigate the potential causal relationship between asthma

Identifying Four Obesity Axes Through Integrative Multiomics and Imaging Analysis.

UNLABELLED: We aimed to identify distinct axes of obesity using advanced magnetic resonance imaging (MRI)-derived phenotypes. We used 24 MRI-derived fat distribution and muscle volume measures (UK Biobank; N = 33,122) to construct obesity axes through principal component analysis. Genome-wide association studies were performed for each axis to uncover genetic factors, followed by pathway enrichment, genetic correlation, and Mendelian randomization analyses to investigate disease associations. Four primary obesity axes were identified: 1) general obesity, reflecting higher fat accumulation in a

Brain-based gene expression and corresponding behavioural relevance of risk genes for broad antisocial behaviour.

Antisocial behaviour (ASB) involves persistent irresponsible, delinquent activities violating rights and safety of others. A meta-analysis of genome-wide association studies revealed significant genetic associations with ASB, yet their brain expression patterns and behavioural relevance remain unclear. Our investigation of fifteen genes associated with ASB examined their biological role and distribution across tissues, integrating post-mortem brain sample data from the Allen-Human-Brain Atlas and the Genotype-Tissue Expression project. We found that these genes were differentially expressed in

The Causal Relationships Between Inflammatory Proteins, Brain Structure, and Psychiatric Disorders: A Two-Step Mendelian Randomization Analysis.

BACKGROUND AND HYPOTHESIS: Inflammatory proteins are implicated in psychiatric disorders, but the causality and underlying mechanisms remain unclear. STUDY DESIGN: We conducted bidirectional Mendelian randomization (MR) using genetic variants from genome-wide association studies (GWAS) for 91 inflammatory proteins (N = 14 824) and 11 psychiatric disorders (N = 9725 to 1 035 760). The primary analysis used the inverse variance weighted (IVW) method, with additional sensitivity analyses to confirm robustness. A two-step MR approach assessed whether brain imaging-derived phenotypes (IDPs) mediat

Cerebral cortex changes in FD, IBS, and GERD: A Mendelian randomization study.

BACKGROUND: Prospective and cross-sectional studies have reported an association between functional gastrointestinal disorders and anxiety and depression. However, the causal relationship remains uncertain. To clarify this, we utilized Mendelian randomization (MR) to assess the causal effects of common gastrointestinal disorders on cortical structures. METHODS: Genome-wide association study (GWAS) data was gathered for functional dyspepsia (FD), irritable bowel syndrome (IBS), and gastroesophageal reflux disease (GERD) from European populations numbering 329,262, 16,792, and 602,604, respecti

Changes in resting-state functional connectivity of large-scale brain networks in bulimia nervosa: evidence from causal analysis.

Bulimia nervosa (BN) has been observationally linked to the functional connectivity (FC) of large-scale brain networks, but the biological mechanisms remain unclear. This study used two-sample Mendelian randomization (MR) with genetic variations as instrumental variables (IVs) to explore potential causal relationships between FC and BN. Summary data from genome-wide association studies (GWAS) involving 2,564 individuals were analyzed to identify genetically predicted BN. Functional magnetic resonance imaging parameters and materials were sourced from the UK Biobank. The variables underwent ind

[Risk factors and prevention strategies in eating disorders].

Scientific evidence supports an interactive model of biopsychosocial influences on the etiology of eating disorders. New genome-wide association studies suggest underlying metabolic dysregulation in anorexia nervosa. Among the psychological factors, body dissatisfaction (IC) is the most consistent predictor, mainly in females. Low self-esteem, negative affect and internalization of the ideal of thinness predict IC. Certain personality traits and other individual factors such as a high body mass index, emotional problems and some metabolic and digestive diseases, social factors such as some spo

Analysis of MRI-derived spleen iron in the UK Biobank identifies genetic variation linked to iron homeostasis and hemolysis.

The spleen plays a key role in iron homeostasis. It is the largest filter of the blood and performs iron reuptake from old or damaged erythrocytes. Despite this role, spleen iron concentration has not been measured in a large, population-based cohort. In this study, we quantify spleen iron in 41,764 participants of the UK Biobank by using magnetic resonance imaging and provide a reference range for spleen iron in an unselected population. Through genome-wide association study, we identify associations between spleen iron and regulatory variation at two hereditary spherocytosis genes, ANK1 and

A rare homozygous MFSD8 single-base-pair deletion and frameshift in the whole genome sequence of a Chinese Crested dog with neuronal ceroid lipofuscinosis.

BACKGROUND: The neuronal ceroid lipofuscinoses are heritable lysosomal storage diseases characterized by progressive neurological impairment and the accumulation of autofluorescent storage granules in neurons and other cell types. Various forms of human neuronal ceroid lipofuscinosis have been attributed to mutations in at least 13 different genes. So far, mutations in the canine orthologs of 7 of these genes have been identified in DNA from dogs with neuronal ceroid lipofuscinosis. The identification of new causal mutations could lead to the establishment of canine models to investigate the p

Interstitial deletions of the short arm of chromosome 4 in a patient with mental retardation and focal seizure.

Interstitial deletion of the proximal short arm of chromosome 4 has rarely been described. This defect is associated with variable clinical manifestations, including mental retardation, unusual facial appearance, and minor limb abnormalities. We describe a girl diagnosed with moderate mental retardation and seizures with an interstitial deletion of the short arm of chromosome 4 [46, XX, del(4)(p12p15.2)].

Development of a blood test for uterine sarcoma-Diagnosis and monitoring (DOORS-D and DOORS-M) studies.

Uterine sarcomas can be difficult to differentiate from uterine fibroids due to many shared symptoms and imaging features, which can result in delayed or missed diagnosis, or over treatment. The 'Development of a blood test for uterine Sarcoma - Diagnosis' (DOORS-D) (ISRCTN14800787) and 'Development of a blood test for uterine Sarcoma - Monitoring' (DOORS-M) (ISRCTN14174468) studies aim to explore the role for circulating tumour DNA (ctDNA) to diagnose and to monitor uterine sarcomas. DOORS-D will recruit patients who have a suspected uterine sarcoma or large fibroid and are due to undergo sur

DeepPathway: predicting pathway expression from histopathology images.

MOTIVATION: Spatial transcriptomics (ST) technologies provide spatially resolved gene expression along with image data, allowing the integrative analysis of complex tissue microenvironment. Despite their potential, the widespread adoption of ST remains limited due to high costs, and methodological challenges in data acquisition. Thus, there have been recent efforts to develop deep learning methods for inferring spatial gene expression from much cheaper and easily available haematoxylin and eosin (H&E) images. These methods demonstrate promising results in reconstructing transcriptomic landscap

Artificial intelligence: a new era in prostate cancer diagnosis and treatment.

Prostate cancer (PCa) represents one of the most prevalent cancers among men, with substantial challenges in timely and accurate diagnosis and subsequent treatment. Traditional diagnosis and treatment methods for PCa, such as prostate-specific antigen (PSA) biomarker detection, digital rectal examination, imaging (CT/MRI) analysis, and biopsy histopathological examination, suffer from limitations such as a lack of specificity, generation of false positives or negatives, and difficulty in handling large data, leading to overdiagnosis and overtreatment. The integration of artificial intelligence
