One-Step Assembly of Homogeneous DAR4 ADCs via a Traceless Fc Ligand-Directed Acylation Strategy.

Site-specific antibody-drug conjugates (ADCs) represent a promising class of biotherapeutics with enhanced pharmacological profiles. we herein report a novel one-step strategy for preparing homogeneous ADCs with a drug-to-antibody ratio (DAR) of 4. This approach leverages a Fc ligand-directed thioester-based acylating reagent combined with a β-glutamic acid-based branching linker to enable precise conjugation of four MMAE molecules per antibody. Through systematic optimization of buffer composition and pH, we successfully mitigated the hydrophobicity-driven aggregation typically associated wit

β-glucuronidase-triggered ortho-hydroxyl-mediated cyclization strategy for the efficient delivery of phenolic payloads in antibody-drug conjugates.

While the payload landscape of antibody-drug conjugates (ADCs) has diversified beyond amine-containing compounds, the application of potent cytotoxic agents possessing phenolic hydroxyl groups remains restricted. This is primarily due to the scarcity of linker technologies capable of balancing systemic stability with efficient release kinetics, as conventional strategies for phenolic conjugation often suffer from premature hydrolysis or sluggish activation. Herein, we report the design and evaluation of a novel self-immolative linker strategy based on an ortho-hydroxyaniline scaffold, which fa

Emerging treatment strategies for newly diagnosed diffuse large B-cell lymphoma.

Despite recent progress in improving patient survival, the outcomes in approximately 40% of patients with diffuse large B-cell lymphoma (DLBCL) remain poor. For many years, R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone) has been the standard first-line treatment for DLBCL; however, pola-R-CHP (polatuzumab vedotin, rituximab, cyclophosphamide, doxorubicin, and prednisone) has recently emerged as a new treatment option. In parallel, novel molecularly targeted therapies that target signaling pathways, cellular antigens, and epigenetic regulators associated with hig

[ 68 Ga]Ga-NECT-224 PET/CT Shows Increasing Nectin-4 Expression in Progressive Urothelial Carcinoma Under ADC-Therapy With Enfortumab-Vedotin.

A 64-year-old man with metachronous metastatic urothelial carcinoma originating from the right renal pelvis underwent nectin-4-targeted [ 68 Ga]Ga-NECT-224 PET/CT prior to initiation of nectin-4-targeted antibody-drug conjugate (ADC) therapy with enfortumab-vedotin (PADCEV), in combination with pembrolizumab. A CT follow-up after 3 months of ADC therapy showed a progressive adrenal metastasis. Subsequent [ 68 Ga]Ga-NECT-224 PET/CT restaging revealed multiple new lesions with intense nectin-4 expression. This case illustrates the potential of [ 68 Ga]Ga-NECT-224 PET/CT to visualize heterogeneou

The human trophoblast cell-surface antigen 2 (TROP-2) expression on metastasized breast cancer.

PURPOSE: The antibody-drug conjugates (ADCs) sacituzumab govitecan (SG) and datopotamab deruxtecan target trophoblast cell surface antigen 2 (TROP-2) and have shown significant efficacy in HER2-negative metastatic breast cancer (mBC). As TROP-2 may serve as a target across multiple treatment lines, detailed information on TROP-2 expression over time is of great interest. METHODS: TROP-2 expression was analyzed in breast cancer (BC) samples from patients treated at the University Hospital Würzburg between 2004 and 2025 at clinically indicated biopsy time points (TP), and, in a subset, before a

An antibody-drug conjugate active against antibiotic-resistant Neisseria gonorrhoeae.

Neisseria gonorrhoeae is the causative agent of gonorrhea, a sexually transmitted infection which is rising in incidence, with increasing drug-resistant strains posing a significant public health threat. To address the urgent need for novel therapies, we developed an antibody-drug conjugate (ADC) that targets this important human pathogen. We utilized Tridecaptin A1, a potent antimicrobial peptide (AMP) against Gram-negative bacteria that exhibits significant toxicity against human cells, limiting its development for clinical use. By conjugating the Tridecaptin A1 analogue, Oct-TriA1 to a mono

Caged Glycan-Antibody Conjugates for Tumor-Selective Activation of Lysosome-Targeting Chimeras.

Lysosome-targeting chimeras (LYTACs) induce lysosomal degradation of extracellular and membrane proteins by bridging target proteins with lysosomal trafficking receptors. However, conventional LYTACs often suffer from off-tissue effects, whereas reported tissue-specific LYTACs typically display limited degradation efficiency. To address these challenges, we report a chemically activatable LYTAC platform that leverages tumor microenvironment-specific cues to achieve precise and safe protein degradation in vivo. We designed a glutathione (GSH)-responsive caged mannose-6-phosphate glycan, GSH-pM6

Mesothelin-directed protein-drug conjugates for mesothelin-low solid tumor therapy.

Mesothelin (MSLN) is an attractive therapeutic target for precision cancer treatments. However, MSLN can be cleaved and shed from tumor cells, resulting in the presence of soluble MSLN (sMSLN), which significantly hinders the efficacy of MSLN-targeted therapies. Here, we identify a MSLN-targeting designed ankyrin repeat protein (DARPin) M7, which specifically binds to the protease-sensitive C-terminal region of MSLN. Furthermore, we develop two auristatin-based DARPin-drug conjugates (DARPin-DCs), M7A-DC and M7GA-DC. We show that M7A-DC and M7GA-DC are effectively internalized by MSLN-positive

Precemtabart tocentecan, an anti-CEACAM5 antibody-drug conjugate, in metastatic colorectal cancer: a phase 1 trial.

CEACAM5, a cell surface protein, is overexpressed in colorectal cancer (CRC). Precemtabart tocentecan (Precem-TcT, previously M9140) is an anti-CEACAM5 antibody-drug conjugate with the topoisomerase 1 inhibitor exatecan as payload. Precem-TcT demonstrated strong antitumor activity and potent bystander activity in preclinical models. Its toxicity profile in cynomolgus monkeys was consistent with that of exatecan. In the dose-escalation stage of the phase 1 trial of Precem-TcT (PROCEADE-CRC-01), 40 heavily pretreated patients with irinotecan-refractory metastatic CRC received Precem-TcT every 3

The Antibody-Drug Conjugate Sacituzumab Govitecan (IMMU-132) Represents a Potential Novel Therapeutic Strategy in Cholangiocarcinoma.

Cholangiocarcinoma is a rare and aggressive cancer type with limited therapeutic options. Several novel antibody-drug conjugates (ADC) have demonstrated promising antitumor activity in solid tumors. This study aimed to investigate the expression and the potential prognostic role of the protein targets of the recently developed ADCs in cholangiocarcinoma. Moreover, the study aimed to establish patient-derived tumor organoids (PDO) and to employ them for in vitro ADC testing. We evaluated the expression of TROP2, NECTIN4, folate receptor 1, HER2, and HER3 via IHC in a cholangiocarcinoma tissue m

[Polatuzumab vedotin in the treatment of relapsed or refractory DLBCL: a case report].

Diffuse large B-cell lymphomas (DLBCLs) represent one of the most common and aggressive forms of non-Hodgkin lymphoma (NHL), characterized by rapid B-cell growth and a significantly high risk of relapse or refractoriness. The clinical course for these patients is long and complicated with a sometimes dismal prognosis, and the therapeutic approach must include effective and innovative chemotherapeutic treatments. The therapeutic strategies for DLBCLs in recent years has seen considerable pharmacological enrichment, including drug-conjugated antibodies such as polatuzumab vedotin. Indeed, this d

Efficacy and safety of trastuzumab deruxtecan in HER2-positive breast cancer patients with brain metastases after failure of pyrotinib-based therapy.

Tyrosine kinase inhibitors (TKIs) and trastuzumab deruxtecan (T-DXd) have shown efficacy in HER2-positive patients with brain metastases (BMs). This paper analyzed the efficacy and safety of T-DXd in HER2-positive breast cancer patients with BMs who progressed after pyrotinib treatment. We conducted a single-center, retrospective cohort study. HER2-positive patients with BMs who received T-DXd treatment after disease progression following pyrotinib therapy were identified from electronic medical records. The primary endpoint of this study was central nervous system progression-free survival (C

Potential Mechanisms of Interstitial Lung Disease Induced by Antibody-Drug Conjugates Based on Quantitative Analysis of Drug Distribution.

Antibody-drug conjugates (ADC) are a rapidly advancing category of therapeutic agents with notable anticancer efficacy. However, the emergence of interstitial lung disease as a severe ADC-associated adverse event highlights the need to better understand the underlying mechanisms. In this study, xenograft model mice with tumors expressing different levels of the trophoblast antigen 2 (TROP2) were generated by subcutaneously transplanting the various TROP2-expressing cancer lines. The mice received different doses of TROP2-eribulin, a novel TROP2-targeting ADC, composed of an anti-TROP2 antibody
