Selective anorexigenic effects of corticotropin releasing hormone in the rhesus monkey.

1. Rhesus monkeys were equipped with a novel intracerebroventricular (i.c.v.) cannula system and trained to respond under operant schedules of food presentation or termination of stimuli associated with the delivery of shock (escape). 2. CRH decreased food-maintained behavior in a dose-related manner over the range of (0.3-10 micrograms/kg) but did not affect escape responding, demonstrating a selective effect on food-maintained responding. 3. This selective effect was related to the tendency for responding to stop after delivery of a food pellet when higher doses of CRH were given, consistent

Natriuresis, kaliuresis and antidiuresis induced by central carbachol injection are mediated by muscarinic M1 receptors.

The present study investigated the effect of selective muscarinic antagonists on natriuresis, kaliuresis and antidiuresis induced by intracerebroventricular (i.c.v.) injection of carbachol in the rat. The muscarinic antagonists were given by i.c.v. injection 1 min before carbachol (1 microgram/rat). 4-Diphenylacetoxy-N-methyl-piperidine methiodide (4-DAMP), a rather selective M1 and M3 receptor antagonist, was the most potent inhibitor of carbachol-induced natriuresis, kaliuresis and antidiuresis, its ID50 being respectively 0.12, 0.04 and 0.56 nmol/rat. Pirenzepine, a selective M1 antagonist,

Relationship between regulation of morphine-induced EEG effects and changes in naloxone sensitivity.

The present data indicate that pretreatment with i.c.v. injection of dynorphin, morphine and dynorphin/morphine resulted in quantitative and qualitative changes in EEG power spectra in rats given i.c.v. morphine 24 h later. Correlated changes in sensitivity to antagonism of these EEG effects by naloxone were also found. Rats were implanted with cortical EEG electrodes and i.c.v. and i.v. cannulas. I.c.v. injections of morphine (20 micrograms/rat) produced high-voltage, slow-wave EEG bursts (1-10 Hz) associated with behavioral stupor which lasted about 2 h. Injections of i.c.v. morphine in rats

Opposite effects of central immunoneutralization of angiotensin II or atrial natriuretic peptide on luteinizing hormone release in ovariectomized rats.

Studies both in vivo and in vitro have provided evidence that LH secretion can be altered by angiotensin II (AII) or atrial natriuretic peptide (ANP); however, the physiological significance of these endogenous neuropeptides in the control of gonadotropin release is still unclear. We studied the effects of central immunoneutralization of these neuropeptides on LH and FSH release in ovariectomized rats. Antisera directed against AII or ANP were microinjected into the third cerebral ventricle (3V) of conscious, unrestrained rats with indwelling external jugular catheters and blood samples were w

Centrally administered ANF promotes appearance of a circulating sodium pump inhibitor.

We have previously reported that release of an endogenous Na pump inhibitor, a putative natriuretic hormone associated with many hypertensive states, may be regulated by the central nervous system (CNS). The atrial natriuretic factor (ANF), which causes diuresis, vasorelaxation, and inhibition of aldosterone and vasopressin release, has been documented in the CNS as well as the atria and peripheral blood. Our experiments investigate the possible interaction of ANF with a circulating specific inhibitor of the Na pump in normal rats. The Na pump inhibitory capacity of plasma from rats previously

Effect of atrial natriuretic peptide on ischaemic brain oedema.

We investigated the effect of intraventricularly administered atrial natriuretic peptide (ANP) on the brain water, sodium and potassium contents in ischaemic brain oedema. Global ischaemia (15 min) followed by recirculation (30 min) produced by a three vessel occlusion in the rat served as a model for induction of ischaemic cerebral oedema. Water content was measured by a drying-weighing method. Sodium and potassium contents were measured by means of flame photometry. The effect of ANP was evaluated by comparing these parameters of the ANP-treated group (5 micrograms/kg and 10 micrograms/kg at

Central inhibition by gamma-aminobutyric acid and muscimol of the release of vasopressin and oxytocin by an osmotic stimulus in the rat.

1. In water-loaded rats under ethanol anaesthesia, the injection of 2-4 microliters 1.54M NaCl solution (hypertonic saline:HS) into a lateral cerebral ventricle (i.c.v.) produced an antidiuretic and a pressor response, together with increased urinary excretion of vasopressin and 'oxytocin-like radioimmunoreactivity' (OLRI). In lactating rats HS also produced a milk-ejection response which was shown to be due to the release of oxytocin. 2. The injection of 20-40 micrograms gamma-aminobutyric acid (GABA) or 40-80 ng muscimol i.c.v. 2 min before HS inhibited the antidiuretic, pressor and milk-eje

Cardiovascular effects of central administration of beta-endorphin in rats receiving neonatal injection of monosodium glutamate.

1. The effects of intracerebroventricular (i.c.v.) and intracisternal (i.c.) injection of beta-endorphin on arterial blood pressure (BP) in rats that received five intraperitoneal injections of monosodium glutamate (MSG) on alternate days in the first 10 days of life were studied. 2. beta-endorphin administered into the lateral ventricles caused a prolonged elevation in BP, whereas i.c. injection of the peptide resulted in an even longer lasting reduction in BP. In the MSG-treated rat, the prolonged hypertensive effect of i.c.v. injection of beta-endorphin was completely abolished, but the eff

Evidence for a role of delta sleep-inducing peptide in slow-wave sleep and sleep-related growth hormone release in the rat.

To examine the role of delta sleep-inducing peptide (DSIP) in sleep-related growth hormone (GH) release, male rats were deprived of sleep for 4 hr by placing them on a slowly rotating wheel. Sleep deprivation by this method caused a significant increase in GH release, as indicated by the increase in plasma GH concentrations (P less than 0.01), and also in the amount of slow-wave sleep (SWS) (P less than 0.001) above initial values after removal of the animals from the rotating wheel. These increases were blocked by microinjection into the third cerebral ventricle of highly specific antiserum t

Effects of adrenalectomy and aldosterone on the action of centrally administered rat atrial natriuretic peptide-(99-126).

Intracerebroventricular (i.v.t.) administration of rat atrial natriuretic peptide-(99-126) (rANP) increases urinary volume and sodium excretion, but the mechanism is undefined. A diminished mineralocorticoid effect on the kidneys may explain the natriuretic phenomenon. This hypothesis was tested by i.v.t. rANP injection (1.25 micrograms/5 microliters) in conscious, hydrated rats pretreated beforehand with d-aldosterone (20 micrograms/kg, i.p.). Although the absolute amount of sodium excreted was reduced, aldosterone did not affect rANP-induced sodium output at 1 and 3 h. Rats which were sham-o

Anticonvulsive activity of several excitatory amino acid antagonists against barbital withdrawal-induced spontaneous convulsions.

Several excitatory amino acid antagonists were tested for an ability to prevent spontaneous convulsions seen during the barbital abstinence syndrome in rats. Barbital-dependent animals were continuously infused intracerebroventricularly (i.c.v.) for the first 48 h following barbital withdrawal with either saline, 2-amino-7-phosphonoheptanoic acid (APH), magnesium sulfate, glutamyldiethyl ester (GDEE) or cis-2,3-piperidine dicarboxylic acid (PDA) using the highest dosages which did not affect normal behavior of the rats. All animals were observed continuously from 12 to 48 h postwithdrawal and

The lack of involvement of central nervous system in the antiarrhythmic effects of prostaglandins E2, F2 alpha, and I2 in cat.

The role of the central nervous system (CNS) in the antiarrhythmic effects of prostaglandins (PGs) E2, F2 alpha, and I2 was studied by administering each agent into the left lateral cerebral ventricle (i.c.v. administration) of chloralose-anaesthetized cats. The cardiac arrhythmias were produced by intravenous (i.v.) infusion of ouabain (1 microgram/kg/min). The PGs E2, F2 alpha and I2 on i.c.v. administration in the dose range of 1 ng to 10 micrograms failed to inhibit ouabain-induced cardiac arrhythmias. However, when infused i.v., PGE2 (1 microgram/kg/min), PGF2 alpha (5 micrograms/kg/min),

Changes in central serotoninergic transmission affect clonidine analgesia in monkeys.

The effects of changes in central serotoninergic transmission on clonidine analgesia were assessed in monkeys. The minimum electrical current required for producing jaw opening is referred to as the pain threshold. Pain was induced by electrical stimulation of tooth pulp afferents. In the first series of studies, intracerebroventricular administration of clonidine (5-30 micrograms) produced dose-dependent analgesia in monkeys. The clonidine-induced analgesia was abolished or attenuated by prior injection of the animals with p-chlorophenylalanine or 5,7-dihydroxytryptamine into the third cerebr

Central nervous system actions of beta-endorphin on gastric acid secretion.

We assessed the central nervous system (CNS) actions of beta-endorphin on gastric acid secretion in awake dogs. Synthetic beta-endorphin (0.2-2.0 nmol X kg-1), but not Leu- or Met-enkephalin, microinjected into the third cerebral ventricle, significantly (P less than 0.01) decreased gastric acid secretion stimulated by pentagastrin. beta-Endorphin given intracerebroventricularly inhibited gastric acid secretion following 2-deoxy-D-glucose (P less than 0.01), but not after stimulation with histamine. Intravenous administration of beta-endorphin did not inhibit gastric acid secretion. beta-Endor

Pressor, tachycardic and feeding responses in conscious rats following i.c.v. administration of dynorphin. Central blockade by opiate and alpha 1-receptor antagonists.

Experiments were designed to determine the hemodynamic responses of conscious, unrestrained rats given intracerebroventricular (i.c.v.) injections of dynorphin A-(1-13) and the possible central receptor mechanisms mediating those changes. Male Sprague-Dawley rats (300 gb. wt.) received i.c.v. injections (by gravity flow in a total volume of 3 or 5 microliter) of control solutions of sterile saline (SS) or dimethylsulfoxide (DMSO) or 1.5, 3.0 or 6.1 nmol of dynorphin A-(1-13). Studies on thyrotropin-releasing hormone-induced micturition in cats.

In unanaesthetized cats micturition produced by thyrotropin-releasing hormone (TRH) was investigated after its injection into the cerebral ventricles through chronically implanted cannulae. TRH in doses from 0.1 to 1.0 mg evoked dose-dependent micturition. In cats treated with intracerebroventricular (i.c.v.) reserpine and 6-hydroxydopamine, but not with i.c.v. 5,6-dihydroxytryptamine and hemicholinium, the micturition caused by i.c.v. TRH was abolished. Chlorpromazine and antazoline injected into the cerebral ventricles prevented the micturition induced by i.c.v. TRH. On the other hand, mecam

Clonidine suppression and its adrenoreceptor mediation in schedule-induced polydipsia.

Rats were trained in a fixed-interval, one-minute (FI 1 min) food reinforcement schedule for 1 hour daily at reduced body weight until their lever presses, licks and water intake all became stabilized for 6 days. Two experiments were performed to examine the function of sympathetic activity in schedule-induced polydipsia. In experiment 1, intracerebroventricular injection of clonidine (0.75-37.5 nmol) produced a dose-related suppression of schedule-induced drinking and licking and schedule-dependent lever pressing; these effects were later attenuated by yohimbine (5 nmol) pretreatment. Intracerebroventricular infusions of rat satietin into rats does not produce conditioned taste aversion.

In a previous study we found that while human satietin (h-SAT) suppressed the food intake of rats it was also aversive to them. In the present study rat satietin (r-SAT) was tested for aversiveness in rats fitted with chronic third ventricle (ICV) cannulas. The rats were then given access to water for 1-hr/day and food ad lib for ten days. Fluid intake, food intake during fluid access and 24-hr total food consumption were recorded. The rats were then ICV infused with saline and 30 min later half of the animals given access to banana flavored water (Group 1) while the remainder were presented w

Thermoregulatory response in rats following administration of histamine in different CSF compartments.

Recently histamine is being considered as an important neurotransmitter/neuromodulator in the central nervous system. This has been supported from the present observations with regard to thermoregulatory responses elicited following histamine administration into different CSF compartments. Administration of histamine into the right lateral cerebral ventricle of rats showed a dose dependent fall in rectal temperature. This hypothermic response was evident only at moderately low or at thermoneutral ambient temperature. Administration of histamine into fourth ventricle produced hypothermic respon

Passive avoidance deficit following intracerebroventricular administration of cholecystokinin tetrapeptide amide in rats.

The effect of cholecystokinin tetrapeptide amide (CCK-4) injected into the lateral cerebral ventricle on memory processes was examined by a one-trial passive avoidance test in the rat. CCK-4 injection 30 and 60 min before the first retention test caused a shortened latency to response, and its chronic infusion into the lateral ventricle at a rate of 2 micrograms/day shortened the latency of the response to the level of almost complete amnesia. CCK-4 also reduced arginine-vasopressin effect on memory processes when administered simultaneously 30 min before the first retention test, but its amne
