In Vivo Identification of Adducts from the New Hypoxia-Activated Prodrug CP-506 Using DNA Adductomics.

Many chemotherapeutic drugs exert their cytotoxicity through the formation of DNA modifications (adducts), which interfere with DNA replication, an overactive process in rapidly dividing cancer cells. Side effects from the therapy are common, however, because these drugs also affect rapidly dividing noncancerous cells. Hypoxia-activated prodrugs (HAPs) have been developed to reduce these side effects as they preferentially activate in hypoxic environments, a hallmark of solid tumors. CP-506 is a newly developed DNA-alkylating HAP designed to exert strong activity under hypoxia. The resulting C

Atroposelective Synthesis of Biaryl N-Oxides via Cu-Catalyzed De Novo Heteroaromatic N-Oxide Ring Formation.

Heteroaromatic N-oxides, renowned for their highly polar N─O bond and robust structure, exhibit significant bioactivities and have played a pivotal role in various drug development projects since the discovery of Minoxidil. Moreover, heteroaromatic N-oxides, featuring axially chiral biaryl frameworks, are indispensable as Lewis base catalysts and ligands in organic synthesis. Despite their importance, synthesizing these chiral compounds is challenging, necessitating chiral starting materials or resolution processes. Catalytic strategies rely on the functionalization of heteroaromatic N-oxide c

Apoptotic effect of noscapine in breast cancer cell lines.

Cancer is a public health problem in the world and breast cancer is the most frequently cancer in women. Approximately 15% of the breast cancers are triple-negative. Apoptosis regulates normal growth, homeostasis, development, embryogenesis and appropriate strategy to treat cancer. Bax is a protein pro-apoptotic enhancer of apoptosis in contrast to Bcl-2 with antiapoptotic properties. Initiator caspase-9 and caspase-8 are features of intrinsic and extrinsic apoptosis pathway, respectively. NF-κB is a transcription factor known to be involved in the initiation and progression of breast cancer.

Chemotherapy-induced loss of bone and muscle mass in a mouse model of breast cancer bone metastases and cachexia.

BACKGROUND: Chemotherapy used to treat malignancy can lead to loss of skeletal muscle mass and reduced force production, and can reduce bone volume in mice. We have shown that bone-muscle crosstalk is a key nexus in skeletal muscle function and bone homeostasis in osteolytic breast cancer bone metastases. Because chemotherapy has significant negative side effects on bone mass, and because bone loss can drive skeletal muscle weakness, we have examined the effects of chemotherapy on the musculoskeletal system in mice with breast cancer bone metastases. METHODS AND RESULTS: Six-week-old Female a

A Ruthenium-BODIPY Photosensitizer for Light-Triggered Apoptosis in Triple-Negative Breast Cancer Cells.

The selective activation of therapeutic agents within the tumor microenvironment remains a cornerstone in enhancing the efficacy and safety of cancer treatments. This study introduces RuB3, an agent for photodynamic therapy (PDT), and explores its potential to selectively induce apoptosis in MDA-MB-231 breast cancer cells through controlled light exposure. Employing advanced live cell imaging techniques, fluorescence microscopy, and flow cytometry, we demonstrated that RuB3 facilitates targeted ROS generation and consequent apoptotic pathways upon light activation. Notably, RuB3 exhibited a su

A Chemoenzymatic Labeling Strategy for Site-Specific Analysis of Tumor-Associated Sialyl Thomsen-Friedenreich Antigen.

Sialyl Thomsen-Friedenreich (ST) antigen, the attachment of trisaccharide Neu5Acα2-3Galβ1-3GalNAc on a serine or threonine residue, is a well-known tumor-associated carbohydrate antigen. There is a growing body of studies indicating that aberrant expression of ST antigen has a strong relationship with tumor invasion and metastasis. However, details about the biological functions of ST antigen are still poorly understood, as there is a lack of an effective method for glycosylation site analysis of the ST antigen. In this study, we developed a chemoenzymatic labeling strategy for the efficient s

Synergistic combination of pirfenidone and paclitaxel suppresses migration and stemness in triple-negative breast cancer: implications of EMT and pluripotency pathways.

Triple-negative breast cancer (TNBC) is an aggressive type of cancer with limited treatment options and high risks of metastasis and recurrence. Growing evidence shows that paclitaxel (PTX), a first-line chemotherapeutic agent in TNBC, may paradoxically promote pro-metastatic features of epithelial-mesenchymal transition (EMT) and cancer stemness in surviving cancer cells. The present study investigated the potential of pirfenidone (PFD), an anti-fibrotic drug, in combination with PTX against cancer proliferation, migration, and stemness of MDA-MB-231 cells. Findings by CompuSyn demonstrated s…

Poly Organotin Acetates against DNA with Possible Implementation on Human Breast Cancer.

Two known tin-based polymers of formula {[R₃Sn(CH₃COO)]n} where R = n-Bu⁻ (1) and R = Ph⁻ (2),were evaluated for their in vitro biological properties. The compounds were characterized via their physical properties and FT-IR, 119Sn Mössbauer, and ¹H NMR spectroscopic data. The molecular structures were confirmed by single-crystal X-Ray diffraction crystallography. The geometry around the tin(IV) ion is trigonal bi-pyramidal. Variations in O⁻Sn⁻O···Sn' torsion angles lead to zig-zag and helical supramolecular assemblies for 1 and 2, respectively. The in vitro cell viability against human breast

Small molecule targeting of the p38/Mk2 stress signaling pathways to improve cancer treatment.

PURPOSE: Although a long-term goal of cancer therapy always has been the development of agents that selectively destroy cancer cells, more recent trends have been to seek secondary agents that sensitize cancer cells to existing treatment regimens. In this regard, the present study explored the possibility of using small molecule inhibitors of p38MAPK/MK2 stress signaling pathways as potential agents to enhance the sensitivity of cancer cells with abrogated G1 checkpoint to the DNA damaging agent etoposide by specifically targeting the DNA damage-induced G2 cell cycle checkpoint. METHODS: We h

Turning the Spotlight Away from PDT: Chemodynamic Dominance of 2-(2-Bromopyridin-4-yl)-1H-imidazo[4,5-f][1,10]phenanthroline Based Ru(II)/Ir(III)/Re(I) Complexes against MDA-MB-231.

A series of ruthenium(II), iridium(III), and rhenium(I) complexes incorporating a 2-bromopyridine imidazophenanthroline ligand were synthesized and initially poised as photodynamic therapy candidates, supported by TD-DFT and excited-state lifetime data. Interestingly, cytotoxicity studies in triple negative cancer cells (MDA-MB-231) showed only marginal light-induced effects and low phototoxicity index values; the photodynamic therapeutic potential fell short. In contrast, the ruthenium complex [RuL] delivered striking anticancer activity in the dark, marking a clear shift toward chemodynamic

Analysis of Pathological Activities of CCN Proteins in Bone Metastasis.

Bone metastasis is a common occurrence in human malignancies, including breast, prostate, and lung cancer, and is associated with a high morbidity rate because of intractable bone pain, pathological fractures, hypercalcemia, and nerve compression. Animal models of bone metastasis are important tools to investigate the pathogenesis and develop treatment strategies. However, there are few models of spontaneous bone metastasis despite the fact that animals often spontaneously develop cancer. Here, we describe methods for developing a mouse model of breast cancer bone metastasis achieved by inject

Dual-targeted pH-responsive nanoliposome (M5/NMN/DEAP/iRGD-Lip): Immunostimulatory-photodynamic synergy for breast cancer therapy.

Photodynamic therapy (PDT) holds promise for combination antitumor therapies by triggering immunogenic cell death (ICD). ICD is defined as the process by which tumor cells, upon death induced by external stimuli, convert from a non‑immunogenic to an immunogenic state, thereby mediating an anti‑tumor immune response in the host. But the poor aqueous solubility and inadequate tumor targeting of photosensitizers hinder their clinical translation. This study focuses on a novel BODIPY photosensitizer (M5) and aims to improve its antitumor efficacy via efficient tumor-targeted delivery and controlla

NOSH-aspirin (NBS-1120) inhibits estrogen receptor-negative breast cancer in vitro and in vivo by modulating redox-sensitive signaling pathways.

Estrogen receptor (ER)-negative breast cancers are known to be aggressive and unresponsive to antiestrogen therapy, and triple-negative breast cancers are associated with poor prognosis and metastasis. Thus, new targeted therapies are needed. Forkhead box M1 (FOXM1) is abundantly expressed in human cancers and implicated in protecting tumor cells from oxidative stress by reducing the levels of intracellular reactive oxygen species (ROS). Aspirin, a prototypical anticancer agent with deleterious side effects that has been modified to release nitric oxide and hydrogen sulfide is called nitric ox
