Dysregulation of DNA methylation and expression of imprinted genes in mouse placentas of fetal growth restriction induced by maternal cadmium exposure.

Cadmium (Cd) is one of the most toxic environmental pollutants that cause fetal malformation and growth restriction. However, the molecular mechanisms underlying maternal Cd toxicity on fetal growth remain largely unknown. Specifically, the expression profiles and the regulation mechanisms of the imprinted genes, have been poorly characterized in the etiology of Cd-induced fetal growth restriction (FGR). In the present study, 13 imprinted genes associated with the fetal growth and placenta development were selected and their expression patterns were examined in the Cd-exposed placentas. Quanti

Reproductive Outcomes Following Maternal Exposure to the Events of September 11, 2001, at the World Trade Center, in New York City.

OBJECTIVES: To estimate associations between exposure to the events of September 11, 2001, (9/11) and low birth weight (LBW), preterm delivery (PD), and small size for gestational age (SGA). METHODS: We matched birth certificates filed in New York City for singleton births between 9/11 and the end of 2010 to 9/11-related exposure data provided by mothers who were World Trade Center Health Registry enrollees. Generalized estimating equations estimated associations between exposures and LBW, PD, and SGA. RESULTS: Among 3360 births, 5.8% were LBW, 6.5% were PD, and 9% were SGA. Having incurred

Effects of maternal inflammation and exposure to cigarette smoke on birth weight and delivery of preterm babies in a cohort of indigenous Australian women.

Sudden infant death syndrome (SIDS), neonatal deaths, and deaths from infection are higher among Indigenous Australians. This study aimed to determine the effects of inflammatory responses and exposure to cigarette smoke, two important factors associated with sudden death in infancy, on preterm birth, and birth weight in a cohort of Indigenous mothers. Indigenous Australian women (n = 131) were recruited as part of a longitudinal study while attending antenatal care clinics during pregnancy; blood samples were collected up to three times in pregnancy. Serum cotinine, indicating exposure to cig

Transplacental carcinogenesis with dibenzo[def,p]chrysene (DBC): timing of maternal exposures determines target tissue response in offspring.

Dibenzo[def,p]chrysene (DBC) is a transplacental carcinogen in mice (15mg/kg; gestation day (GD) 17). To mimic residual exposure throughout pregnancy, dams received four smaller doses of DBC (3.75mg/kg) on GD 5, 9, 13 and 17. This regimen alleviated the previously established carcinogenic responses in the thymus, lung, and liver. However, there was a marked increase in ovarian tumors (females) and hyperplastic testes (males). [(14)C]-DBC (GD 17) dosing revealed transplacental distribution to fetal tissues at 10-fold lower concentrations than in paired maternal tissue and residual [(14)C] 3week

Evidence of gestational heroin exposure by comparative analysis of fetal and maternal body fluids, tissues, and hair in a heroin-related death.

In order to determine the fetal-maternal distribution of heroin and its main metabolites (6-monoacetylmorphine and morphine) the drug concentrations were measured in autopsy material. The heroin-related death of a pregnant drug abuser (approximately the 32nd week of gestation) provided the fetal and maternal material. Fetal and maternal hair was analyzed in order to obtain long-term information on the transplacental opiate transfer. Morphine and 6-monoacetylmorphine were detected in toxic concentrations in maternal as well as in fetal tissues and body fluids. The drug concentrations in the fet

Gestational exposure to per- and polyfluoroalkyl substances drives maternal liver dysfunction through lipid dysregulation and NF-κB/EGFR-STAT3 inflammatory activation.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous persistent pollutants in pregnant women, yet mechanisms of PFAS-triggered gestational liver injury remain poorly understood. This work quantified 18 serum PFAS in 221 pregnant participants and explored PFAS-liver function associations via epidemiological analyses, network toxicology and supportive experiments in HepG2 cells. Ten PFAS exhibited detection rates over 80%, with Perfluorooctane sulfonate (PFOS, 6.14 μg L-1), perfluorohexane sulfonate (PFHxS, 5.54 μg L-1) and perfluorooctanoic acid (PFOA, 4.79 μg L-1) as dominant congeners. Maternal Exposure to Microplastics and High-Fructose Diet Induces Offspring Hypertension via Disruption of H2S Signaling, Gut Microbiota, and Metabolic Networks.

Maternal consumption of a high-fructose (HF) diet or exposure to microplastics (MPs) can each independently affect kidney development and increase the risk of hypertension in adult offspring, yet their combined impact remains poorly understood. Dysregulation of hydrogen sulfide (H2S) signaling and alterations in gut microbiota are potential mediators of this programming. Pregnant rats received either standard chow or a 60% HF diet, with half of each group additionally exposed to sulfate-modified MPs (1 mg/L) with a 5 μm diameter throughout pregnancy and lactation. Male offspring were divided i

Exposure-associated health implications of potentially toxic elements in maternal and umbilical cord blood at Ishaka adventist hospital, Bushenyi District, Uganda.

Prenatal exposure to potentially toxic elements (PTEs), including heavy metals and metalloids, poses a significant public health risk in low- and middle-income countries (LMICs), where environmental surveillance is limited. This study assessed maternal and foetal exposure to PTEs in maternal and umbilical cord blood samples (n = 32 each) collected at Ishaka Adventist Hospital, Uganda, an agricultural region with known environmental contamination. Concentrations of arsenic (As), lead (Pb), chromium (Cr), nickel (Ni), copper (Cu), iron (Fe), and zinc (Zn) were quantified using Microwave Plasma A

Maternal control during prenatal oxycodone exposure: differential effects of voluntary versus involuntary self-administration on offspring substance use vulnerability in rats.

Prenatal opioid exposure is a growing clinical concern, yet the impact of maternal control over drug intake on long-term offspring outcomes remains unclear. Here, we utilized a translational rat model to examine how the mode of oxycodone administration during pregnancy influences substance use vulnerability in adult offspring. Female Sprague-Dawley rats (n = 10-11/group) were assigned to one of three groups: voluntary intravenous self-administration (Oxycodone-Lead), yoked non-contingent oxycodone delivery (Oxycodone-Yoked), or yoked saline control (Saline-Yoked). The dam self-administered oxy

Maternal microRNAs linked to birth deficits due to prenatal alcohol exposure are dominant modifiers of gene expression in invasive trophoblast cells.

INTRODUCTION: Prenatal alcohol exposure (PAE) can impair placental development and result in fetal growth restriction. However, the underlying molecular mechanisms are unclear. We previously found that PAE in some, but not all pregnancies, resulted in significantly elevated circulating levels of 11 microRNAs (HEamiRNAs) in the 2nd trimester. This subgroup of PAE women experienced adverse infant birth outcomes due to PAE, Subsequently, these HEamiRNAs collectively but not individually, were found to inhibit a defined set of placental epithelial-mesenchymal transition (EMT) genes and, inhibited

Maternal exposure to polystyrene nanoplastics during gestation and lactation impaired skeletal growth in progeny mice by inhibiting neutrophil extracellular trap formation.

Microplastics and nanoplastics are widely distributed in the natural environment and shown to accumulate in living organisms. While their potential impact on human health has been investigated, significant uncertainties remain regarding their toxic effects and mechanisms of interaction with the human skeletal system. We examined the potential effects of polystyrene nanoplastics (PS-NPs, 100 nm) on skeletal health and the underlying molecular mechanisms using the human RAW264.7 and MC3T3-E1 cell lines as in-vitro models, along with a murine model. Maternal exposure to PS-NPs (10 mg/L) through d…

Transcriptional evidence of HPA axis dysregulation in adolescent females: Unique contributions of chronic early-life stressor exposure and maternal depression history.

BACKGROUND: Depression risk increases dramatically for adolescent females following the pubertal transition. Although chronic early-life stressor exposure and a maternal history of depression are established risk factors for depression onset in this population, we know little about the biological mechanisms underlying these associations. METHOD: To investigate, we examined how chronic early-life stressor exposure and maternal depression history were associated with stress-related gene expression patterns, using a high-risk family design in 48 psychiatrically healthy adolescent females, 20 of

Maternal exposure to phenanthrene induces testicular apoptosis and Sertoli cell dysfunction in F1 adult male mice: a histological and molecular study.

OBJECTIVE: Phenanthrene, a polycyclic aromatic hydrocarbon, is found in abundance in environmental pollutants, food, and drinking water. This substance can accumulate in body tissues and exert harmful effects. Moreover, phenanthrene can cross the placental barrier, potentially impacting fetal development. We aimed to explore the impacts of maternal exposure to phenanthrene on testicular tissue and Sertoli cell function in F1 mice. METHODS: Female rats with vaginal plugs were randomly assigned to one of three groups: control, sham, or phenanthrene. The control group received no intervention du…

Association between media exposure and maternal health service use in Nepal: A further analysis of Nepal Demographic and Health Survey-2022.

BACKGROUND: Nepal has made incremental progress in improving coverage of maternal health services leading to improved health outcomes. Government and other health sector stakeholders have consistently considered dissemination of educational messages on maternal health through mass media. However, in Nepal's context, the media's influence on the uptake of maternal health services is less known. This study examines the links between media exposure and maternal health service use in Nepal. METHOD: Our analysis is based on the nationally representative Nepal Demographic and Health Survey (NDHS) 2

Chronic inhibition of astrocytic aquaporin-4 induces autistic-like behavior in control rat offspring similar to maternal exposure to valproic acid.

Social communication and interaction deficits, memory impairment, and anxiety-like behavior are characterized in many people identified with autism spectrum disorder (ASD). A thorough understanding of the specific aspects that contribute to the deficiencies associated with ASD can aid research into the etiology of the disorder while also providing targets for more effective intervention. As part of the ASD pathophysiology, alterations in synaptogenesis and abnormal network connections were seen in high-order brain areas, which control social behavior and communication. The early emergence of m

Critical windows of exposure to air pollution and gestational diabetes: assessing effect modification by maternal pre-existing conditions and environmental factors.

BACKGROUND: Ambient air pollution has been associated with gestational diabetes (GD), but critical windows of exposure and whether maternal pre-existing conditions and other environmental factors modify the associations remains inconclusive. METHODS: We conducted a retrospective cohort study of all singleton live birth that occurred between April 1st 2006 and March 31st 2018 in Ontario, Canada. Ambient air pollution data (i.e., fine particulate matter with a diameter ≤ 2.5 μm (PM2.5), nitrogen dioxide (NO2) and ozone (O3)) were assigned to the study population in spatial resolution of approxi
