The mesencephalic neural crest cells of Hamburger-Hamilton Stage 9-Stage 11 chick embryos were surgically extirpated unilaterally in 148 embryos and bilaterally in 8 embryos. Sham operations were performed unilaterally on 16 control embryos and bilaterally on one control embryo. Embryos were fixed at various time intervals after operation, studied macroscopically, and by light and scanning electron microscopy, and their development compared with that of 47 normal embryos. The extirpated mesencephalic region was repopulated by crest cells within 6-8 hours after operation. These 'new' crest cell

Thirty patients with an acute midbrain syndrome were treated by high dose barbiturate therapy. Of these patients 19 had a severe head injury. In 8 patients the symptoms of acute midbrain syndrome developed after subarachnoid haemorrhage. In three patients these symptoms were caused by postoperative swelling or ischaemia. The results of those patients, who were treated with barbiturates after head injury were much better than in 16 other patients, who had no barbiturates. The indications for high dose barbiturate therapy in neurosurgery are discussed with reference to other publications and to

Uptake of tritiated serotonin by synaptosomes prepared from rat lateral hypothalamus was examined. Uptake of serotonin into lateral hypothalamic synaptosomes occurred by both saturable and non-saturable processes. The saturable process was a high affinity transport with kinetic parameters that agree closely with those previously reported for serotonin uptake into synaptosomes prepared from other brain regions. Fluoxetine, a selective inhibitor of uptake into serotonergic neurons, was a potent inhibitor of serotonin uptake into lateral hypothalamic synaptosomes. Desipramine and benztropine, nor

A 64-year-old woman developed complex visual hallucinations consisting of snakes crawling out of people's head and on her body. She had become blind 14 years before the onset of the hallucinations. Neurosurgical removal of a large suprasellar meningioma was followed by disappearance of her hallucinations. This case suggests that the complex hallucinations often encountered in blind subjects ("release hallucinations"), a phenomenon originally described by Charles Bonnet in 1769, are related to both sensory deprivation and an impairment in CNS functioning.

Benzodiazepine receptor binding studies have been reported suggesting the existence of at least two different benzodiazepine receptors. One distinguishing feature of these two sites is that one has a high affinity for triazolopyridazines, whereas the other has a low affinity. In this study, the regional localization of the two different receptors was examined by light microscopic radiohistochemical methods. [3H]Flunitrazepam was used to label all types of benzodiazepine receptors in slide-mounted tissue sections. CL218,872, a typical triazolopyridazine, was used to preferentially displace [3H]

Acute experiments on cats were carried out during traumatic shock to study evoked potentials in the sensorimotor cortex and reticular formation (RF) of the midbrain in response to electrodermal irritation of the contralateral paw. The cortical function was judged by thresholds of convulsant reaction while function of RF by thresholds of activation reaction in the cortex of the course of high-frequency electrical stimulation of the structures under study. It was shown that in traumatic shock, initial and pronounced depression of the cortical function paralleled long enough stability of RF funct

In the Chiari II deformity, the following defects can be visualized by computed tomography: partial or complete fusion of the corpora quadrigemina into a tectal beak or spur and invagination of the midline cerebellum to receive this spur; marked upward, transincisural growth of the cerebellum to produce a supratentorial, "extra-axial" mass effect (especially after shunting); enlargement of the pericerebellar cistern about the "towering" cerebellum, and infolding of the anterior border of the towering cerebellum to overlap the vermis and the more caudal portions of the cerebellum. Seven cases of the most severe form of tuberculous meningitis, in which a midbrain syndrome developed, are reported. Three different types of progress were observed. Exudative inflammation and cerebral edema dominated in the first group, causing the rapid development of the acute midbrain syndrome, which may turn into a bulbar syndrome. In the second group the development of the midbrain was delayed and an apallic syndrome followed. The morphological examination disclosed local diencephalic and midbrain lesions caused by herniation and specific vasculitis and vascular compression. The third gr

Stereotaxic injections of small quantities of horseradish peroxidase (HRP) are made in the vestibular complex in the hen and the labelled cells due to retrograde transport of the tracer enzyme are studied, especially in the vestibular complex but also in other parts of the brain stem. The superior vestibular nucleus sends commissural fibres to the superior, medial, and descending nuclei and to Deiters' complex. The cell group A projects to the contralateral superior nucleus and to the Deiter's complex. The medial nucleus projects to the contralateral superior and descending nuclei, Deiters' co

Interocular transfer of two figural aftereffects was examined in orthotropes and strabismic subjects. Within both groups there were persons with an appreciable degree of stereoacuity and others who had little or none. Experimental evidence from a variety of sources has suggested that stereopsis depends upon binocularity of units in the geniculostriate system. For the tilt aftereffect, interocular transfer correlated with stereoacuity among both orthotropes and strabismics. For the spiral motion aftereffect, interocular transfer did not correlate with stereoacuity; it was present among orthotro

From the study of 65 severe traumatic comas, the authors describe the syndrome of mesodiencephalic dysfunction, because of its frequency and its particular clinical signs. This peculiar level of loss of consciousness is characterized by a coma without mimic and without awakening after painful stimulation, by a stereotyped motor pattern in extension, by a perseverance of the photomotor reflex, while fronto-orbicular and vertical oculo-vestibular reflexes are lacking. This level of deterioration appears to be a critical point in the natural history of comatose patients. If this level is not surp

The spontaneous firing activity of substantia nigra (SN) dopaminergic (DA) neurons is finely tuned by the autocrine inhibition mediated by D2 DA autoreceptors (D2-ARs) that activate GIRK2 channels. Despite this regulatory mechanism, the vulnerability of SN DA neurons may nevertheless increase due to an altered spontaneous firing activity of DA neurons, Ca2+ dishomeostasis, mitochondrial stress, high dendritic arborization, aggregation of α-synuclein (α-syn), α-syn mutations, reduced levels of calbindin protein, etc. Although the intraneuronal accumulation and the spreading of misfolded α-syn i

Neuronopathic Gaucher disease (nGD) is a lysosomal storage disorder caused by GBA1 mutations, leading to defective acid β-glucosidase (GCase) and accumulation of glycosphingolipid substrates, causing inflammation and neurodegeneration. Patients with nGD manifest severe neurological symptoms, but current animal models fail to fully recapitulate the human condition, posing a major barrier to the development of effective therapies targeting the brain. To bridge this gap, we have developed midbrain-like organoids (MLOs) from human induced pluripotent stem cells of nGD patients with GBA1L444P/P415R

Fibrillary aggregation of α-synuclein in Lewy body inclusions and nigrostriatal dopaminergic neuron degeneration define Parkinson's disease neuropathology. Mutations in GBA1, encoding glucocerebrosidase, are the most frequent genetic risk factor for Parkinson's disease. However, the lack of reliable experimental models able to reproduce key neuropathological signatures has hampered clarification of the link between mutant glucocerebrosidase and Parkinson's disease pathology. Here, we describe an innovative protocol for the generation of human induced pluripotent stem cell-derived midbrain orga

The ability to study mature neuronal cells ex vivo is complicated by their non-dividing nature and difficulty in obtaining large numbers of primary cells from organisms. Thus, numerous transformed progenitor models have been developed that can be routinely cultured, then scaled, and differentiated to mature neurons. In this paper, we present a new method for differentiating one such model, the Lund human mesencephalic (LUHMES) dopaminergic neurons. This method is two days faster than some established protocols, results in nearly five times greater numbers of mature neurons, and involves fewer
