Probabilistic tractography of the extracranial branches of the trigeminal nerve using diffusion tensor imaging.

PURPOSE: The peripheral course of the trigeminal nerves is complex and spans multiple bony foramen and tissue compartments throughout the face. Diffusion tensor imaging of these nerves is difficult due to the complex tissue interfaces and relatively low MR signal. The purpose of this work is to develop a method for reliable diffusion tensor imaging-based fiber tracking of the peripheral branches of the trigeminal nerve. METHODS: We prospectively acquired imaging data from six healthy adult participants with a 3.0-Tesla system, including T2-weighted short tau inversion recovery with variable f

Innervation of the rat pineal gland by pituitary adenylate cyclase-activating polypeptide (PACAP)-immunoreactive nerve fibres.

Pituitary adenylate cyclase-activating polypeptide (PACAP)-immunoreactive nerve fibres were demonstrated in the rat pineal gland. These fibres entered the pineal gland through the conarian nerve at the distal tip of the gland. A high density of the fibres was observed in the capsule of the gland, from where the immunoreactive elements penetrated into the pineal perivascular spaces and parenchyma. The majority of PACAP-immunoreactive nerve fibres also contained calcitonin gene-related peptide (CGRP). Some PACAP-immunoreactive nerve fibres contained neuropeptide Y (NPY), but only occasionally wa

[Sciatica and herniated disk. Current aspects of pathophysiology and pain mechanisms].

Pathoanatomical and pathophysiological aspects of spinal nerve roots involved in disk herniation are discussed, and recent advances in our understanding of nerve root physiology and pathophysiology summarised. Evidence is presented suggesting that nerve roots may be affected not only by mechanical compression but also by nucleus pulposus tissue via biochemical mechanisms. Clinical symptoms are also discussed, as are recent findings regarding the diagnosis and treatment of disk herniation and sciatica.

Diagnostic dilemmas in unusual presentations of gout.

BACKGROUND: Gout is known to mimic conditions as diverse as joint and soft tissue infections, skin malignancies, nerve compression syndromes and soft tissue tumours. Upper limb involvement is unusual. OBJECTIVE: We discuss four cases of gout in the hand and wrist masquerading as a soft tissue tumour, nodular extensor tenosynovitis, septic arthritis of the wrist and acute and chronic median nerve compression. These cases illustrate that gout in the hand and wrist can resemble more sinister conditions, often posing a diagnostic challenge even to the experienced clinician. DISCUSSION: In the ma

Suppurative tenosynovitis with chronic carpal tunnel syndrome due to corticosteroid injections: A case report.

INTRODUCTION: Steroid hormone blocking is a common treatment for tenosynovitis. However, local steroid hormone blocking requires careful attention to the local inflammatory response, as infection can cause severe local soft tissue inflammation and damage. PRESENTATION OF CASE: A 46-year-old female patient received local steroid hormone blocking treatment for tenosynovitis of the right thumb at another hospital 3 months earlier. Five days later, the patient gradually experienced redness, swelling, and pain in her right hand. By day 7, the symptoms worsened with increased swelling, wrist pain,

High-voltage electrical stimulation improves nerve regeneration after sciatic crush injury.

BACKGROUND: Peripheral nerve injury causes prolonged functional limitation being a clinical challenge to identify resources that accelerates its recovery. OBJECTIVES: To investigate the effect of high-voltage electrical stimulation (HVES) on the morphometric and functional characteristics of the regenerated nerve after crush injury in rats. METHODS: Twenty Wistar rats were randomly allocated into 4 groups: Control (CON) - without injury and without HVES; Denervated (D) - sciatic nerve crush only; Denervated + HVES - sciatic nerve crush and HVES; SHAM - without injury but HVES. The HVES and S

Transgene expression and effective gene silencing in vagal afferent neurons in vivo using recombinant adeno-associated virus vectors.

Vagal afferent fibres innervating thoracic structures such as the respiratory tract and oesophagus are diverse, comprising several subtypes of functionally distinct C-fibres and A-fibres. Both morphological and functional studies of these nerve subtypes would be advanced by selective, effective and long-term transduction of vagal afferent neurons with viral vectors. Here we addressed the hypothesis that vagal sensory neurons can be transduced with adeno-associated virus (AAV) vectors in vivo, in a manner that would be useful for morphological assessment of nerve terminals, using enhanced green

Prevalence and distribution of HSV-1, VZV, and HHV-6 in human cranial nerve nuclei III, IV, VI, VII, and XII.

The etiology of idiopathic cranial nerve palsies often remains unresolved. It has been hypothesised that viral reactivation of herpesviruses in the corresponding nuclei in the brainstem is the cause. We investigated the distribution of herpes simplex virus type 1 (HSV-1) and varicella zoster virus (VZV) in nuclei that are associated with peripheral sensory ganglia [oculomotor (nIII), facial (nVII) nuclei] and in nuclei that are not associated with peripheral sensory ganglia [trochlear (nIV), abducens (nVI), and hypoglossal (nXII) nuclei] of five human brainstems. Samples of the cranial nerve n

Sema3A chemorepellant regulates the timing and patterning of dental nerves during development of incisor tooth germ.

Semaphorin 3A (Sema3A) axon repellant serves multiple developmental functions. Sema3A mRNAs are expressed in epithelial and mesenchymal components of the developing incisor in a dynamic manner. Here, we investigate the functions of Sema3A during development of incisors using Sema3A-deficient mice. We analyze histomorphogenesis and innervation of mandibular incisors using immunohistochemistry as well as computed tomography and thick tissue confocal imaging. Whereas no apparent disturbances in histomorphogenesis or hard tissue formation of Sema3A (-/-) incisors were observed, nerve fibers were p

Tactile function in skin-equivalent grafts.

Cultured grafts are excellent wound covers; however, their somatosensory capabilities are unknown. This is a preliminary report of a study which determined whether grafts of cultured skin become innervated and also examined whether seeding grafts with target tissue improved nerve growth or functional recovery. Autologous skin for grafting was generated from adult rat biopsy tissue. Dissociated keratinocytes were seeded on top of fibroblast-contracted collagen gels (skin-equivalents). Some animals received grafts composed entirely of skin-equivalents. Others had grafts with 2-mm punch biopsies

Redefining bladder sympathetic Innervation with three-dimensional image reconstruction.

The primary goals of functional bladder reconstruction following spinal cord injury are to restore voiding power and reduce voiding resistance. The sympathetic nerve is one of the critical constraints within the reduce voiding resistance. In this paper, we sought to generate a three-dimensional (3D) reconstruction of bladder-innervating sympathetic nervous tissue, utilizing histopathological samples derived from male and female cadavers. Bladder tissue samples were collected from four formalin-fixed cadavers (two male, two female). After preparing axial cross-sections of the bladder at 4 mm in

Regenerative peripheral nerve interface viability and signal transduction with an implanted electrode.

BACKGROUND: The regenerative peripheral nerve interface is an internal interface for signal transduction with external electronics of prosthetic limbs; it consists of an electrode and a unit of free muscle that is neurotized by a transected residual peripheral nerve. Adding a conductive polymer coating on electrodes improves electrode conductivity. This study examines regenerative peripheral nerve interface tissue viability and signal fidelity in the presence of an implanted electrode coated or uncoated with a conductive polymer. METHODS: In a rat model, the extensor digitorum longus muscle w

The inflammatory response to sciatic nerve injury in a familial amyloidotic polyneuropathy mouse model.

Inflammation is a hallmark of several neurodegenerative disorders including familial amyloidotic polyneuropathy (FAP). FAP is associated wit … ted over time in V30M mice, accompanying a peak of inflammation. Strikingly, V30M TTR deposition in nerve tissue occurred, suggesting that inflammation contributes to TTR polymerization. In response to nerve injury, V30M mice display a downregulated innate immune response when compared to WT mice. More specifically, we saw decreased expression of cytokines and …

Age-related changes in androgen receptor levels in cranial nerve nuclei of male rats.

Sensory and motor events are important for mating, and several cranial nerve nuclei mediating these events contain androgen receptors (AR). Since mating behavior declines with age, we tested whether AR binding is decreased in cranial nerve nuclei of old male rats. Cytosol and cell nuclear androgen receptor (AR) binding was assessed in the cochlear, hypoglossal and facial nuclei in young (4 months) and old (20 months) male Fischer 344 rats. For comparison with other neural and non-neural tissue, AR binding in combined hypothalamus, preoptic area and amygdala (HPA), and muscle tissue from tongue

Preclinical investigation of cavernous nerve injury-induced erectile dysfunction treated with HAFSC-derived secretome.

BACKGROUND: Erectile dysfunction (ED) significantly affects both psychological and physiological well-being, with limited effective treatments available for nerve injury-induced ED. Stem cell therapy, particularly using human amniotic fluid stem cells (hAFSCs), shows promise due to the secretome (hAFSC-S), which is rich in growth factors and chemoattractants. This study investigated the paracrine effects of hAFSC-S on erectile function recovery in a rat model of bilateral cavernous nerve crush (BCNC) injury. MATERIALS AND METHODS: Thirty rats were divided into the sham and BCNC groups. The BC

Facelift Surgery Following Superficial Parotidectomy: Is it Safe?

INTRODUCTION: Patients who have previously undergone superficial parotidectomy may also seek facelift surgery for facial aging and rejuvenation. These patients present unique challenges compared to a standard facelift patient. Most concerning is the location of facial nerve branches, which may be superficial and displaced. In addition, significant contour deformities and abnormal scar patterns may be present. The purpose of the study is to review our series of patients and assess potential morbidity and safety of facelift surgery in superficial parotidectomy patients. METHODS: A retrospective

Salvage of open tibial fracture with segmental loss of tibial nerve: case report and review of the literature.

We report a case history, treatment, and follow-up of an open comminuted distal tibial fracture with significant soft tissue loss and segmental loss of the tibial nerve and posterior tibial artery. This constellation of injuries with an insensate plantar foot has often been an indication for amputation. In this instance, a functional distal extremity was salvaged with the use of Ilizarov fixation, delayed primary tibial nerve cable grafting, and staged soft tissue coverage. Clinical follow-up and review of the literature on the techniques used are offered for consideration.
