Exposure to alcohol during gestation can have profound consequences, but not all cells within the embryo are affected equally. Recent advances in molecular embryology have allowed an exploration of this variation. Much of this research has focused on the embryo's vulnerability to the facial malformations characteristic of fetal alcohol syndrome. Studies using mice and chicks show that alcohol exposure at specific stages of early embryo development results in significant death among the cells destined to give rise to facial structures (i.e., cranial neural crest cells). This type of cell death …

A melanotic paraganglioma occurred in a 57-year-old woman, located in the left paravertebral space of the upper mediastinum. It was totally … mour associated melanophages. The simultaneous expression of functional properties of two different neural crest derived cells in one tumour stresses the close relationship between all neural crest elements and is in accordance with the observation of other melanotic, non-melanomatous tumours.

The neural crest is involved in the differentiation of the cardiac outflow tract and branchial arches. Branchial arches one and two give rise to the eustachian tubes. Anatomically different eustachian tubes (e.g., short length) are found in persons prone to otitis media. We hypothesized a relationship between outflow tract anomalies and eustachian tube anomalies. The medical records of 238 children with a catheterization diagnosis of congenital heart disease were reviewed for otitis diagnoses. Children with syndromes known to be associated with recurring otitis media were excluded. Recurrent o…

An infant with oculocutaneous albinism and congenital glaucoma is described. The association of these two uncommon congenital disorders might suggest a common defective factor. Theoretically this patient broadens the spectrum of disorders of neural crest cell migration associated with albinism.

Neuroblastoma (NBL) is a neuroectodermal tumor derived from neural crest cells. The biological and clinical behavior of NB is extremely heterogenous. We here report a newborn who presented as 4S NBL with a massive hepatomegaly resulting in IVC syndrome.

Gastrointestinal neuroectodermal tumors (GNETs) are extremely rare and intriguing malignancies originating from neural crest cells in the digestive tract. The digestive tract's neural crest cells can give rise to incredibly unusual and interesting gastrointestinal neuroectodermal tumors (GNETs). GNETs present considerable hurdles in diagnosis and management because of their rarity and varied expression. In this case report, a 45-year-old male patient is described who had signs of GNET, such as exhaustion, weight loss, and abdominal pain. A 7-cm jejunum tumor and related thickening of the gut w…

Congenital central hypoventilation syndrome (CCHS) and Hirschsprung's disease (HSCR) are often classified as neurocristopathies and are thought to share a common molecular pathogenesis related to the genes that control the development of neural crest cells. We examined whether endothelin-3 (ET-3), one of the developmental regulators of neural crest cells and of which null mutation results in aganglionic megacolon in mice, fulfills the requirements for such a common molecule. To investigate the possible involvement of ET-3 in central ventilatory control, we measured ventilation in mutant mice d…

OBJECTIVE: The objective of this study was to demonstrate that the branchiooculo-facial (BOF) syndrome is a cervicocephalic neural crest maldevelopment.
RESULTS: Using an embryologic study, we linked the clinical features and the level of the neural crest deficiency. We report here two cases of BOF syndrome with a particular branchial cleft presenting as bilateral supernumerary thymus glands on the surface of the skin; one of the cases was associated with tetralogy of Fallot. One patient underwent lip reconstruction at 4 months, combined with excision of bilateral auricular pits and superior …

BACKGROUND: Mesenchymal stem/stromal cells (MSCs) are of great therapeutic value due to their role in maintaining the function of hematopoietic stem/progenitor cells (HSPCs). MSCs derived from human pluripotent stem cells represent an ideal alternative because of their unlimited supply. However, the role of MSCs with neural crest origin derived from HPSCs on the maintenance of HSPCs has not been reported.
METHODS: Flow cytometric analysis, RNA sequencing and differentiation ability were applied to detect the characteristics of stromal cells from 3D human brain organoids. Human umbilical cord …

Enteric glia are neural crest derivatives. Kabouridis et al. (2015) now show in adult animals that new glia arise within the ganglia of enteric plexuses and then migrate centripetally to colonize the mucosa. Remarkably, enteric microbiota regulate this critical migration.

Carney complex is a multiple endocrine neoplasia (MEN) syndrome that affects the adrenal cortex, the pituitary and thyroid glands, and the gonads. The complex is also associated with skin and mucosa pigmentation abnormalities and myxoid and other neoplasms of mesenchymal and neural crest origin. Thus, this syndrome also belongs to another group of genetic disorders, the lentiginoses (or lentigenoses), which include the Peutz-Jeghers, LEOPARD, arterial dissections and lentiginosis, and Laugier-Hunziker syndromes, Cowden disease and Ruvalcaba-Myhre-Smith (Bannayan-Zonana) syndrome and the centro…

Axolotl embryos have previously been used to study neural crest cell migration. In embryos of the normal wild type, neural crest cells migrate subepidermally to form pigment cells. In the trunk of the white mutant embryo, these cells are unable to migrate, possibly due to an inherited delay in the maturation of the local extracellular matrix. The present investigation reveals a reduced incorporation of [35S]sulfate into PG-M/versican-like proteoglycans synthesized in epidermal explants from the dorsal trunk of white mutant embryos during stages pertinent to migration. This is the major form of…

Gross phenotypic observations, histology, and tissue culture showed that the gene for sex-linked imperfect albinism that occurred at the University of Saskatchewan (Sal-s), allows a small amount of melanin pigment to be deposited in eyes and feathers. Melanin pigment accumulates in retinal pigment epithelial and cultured neural crest cells, but neural crest cells pigmenting the feathers transfer their pigment as it is produced, and this is seen as a constant amount of color in successive generations of feathers. Despite differences from early reports, it would appear that the phenotype produce…

A case of Shah-Waardenburg syndrome, a rare variant of Waardenburg syndrome, is presented. Inherited as an autosomal recessive or dominant trait, the disorder presumably results from defective migration of neural crest cells. It clinically manifests with pigmentary anomalies and congenital megacolon.

Neuroblastoma, ganglioneuroblastoma and ganglioneuroma are neuroblastic tumours derived from primordial neural crest tissue. The authors report a rare presentation of a ganglioneuroma of the mandible arising from a metastasis of an adrenal neuroblastoma. The pathogenesis and behaviour of ganglioneuromas is discussed, together with recommendations for their management.

BACKGROUND: The etiology of the anatomic and functional abnormalities of the esophagus in infants surviving congenital diaphragmatic hernia (CDH) remains unclear. We showed previously that fetal rats with CDH have malformations of neural crest-derived structures. The aim of this study was to examine the anatomy of the vagus and the recurrent laryngeal nerves, both of neural crest origin, in rats with CDH.
METHODS: We used the nitrofen-induced CDH fetal rat model. Nine control fetuses from four dams and nine fetuses with CDH from seven dams were included in this study. Embryos were fixed in fo…

Neurovegetative system consist of preganglionar fibre from neural tube and postganglionar from neural crest. We examined 13 patients with Fuchs' syndrome. The first sign was heterochromia, followed by keratic precipitates, pathological cataract and finally secondary glaucoma. The cause of Fuchs' syndrome was the damage of superior cervical ganglion. We studied 4 cases of Claude Bernard Horner's syndrome through damages of preganglionar sympathetic system, in 3 cases because of laterocervical neoplasia and in the last case through brachial plexus damages.
