Reciprocal inhibition in patients with hand cramps.

We studied inhibition of median nerve H-reflex produced by radial nerve stimulation in both arms of 14 patients with hand cramps and 15 normal volunteers. Median nerve stimulation was delivered 1 and 0.5 msec before and 0, 0.5, 1, 2, 10, 20, 30, 50, 75, 100, 200, 500, and 1,000 msec after radial nerve stimulation. Recordings were from surface electrodes positioned over flexor carpi radialis. We identified three periods of attenuation of median nerve H-reflex amplitude in the control group. When radial nerve conditioning stimuli were delivered simultaneously, 10 and 75 msec before median nerve

Sympathoinhibitory responses to digitalis glycosides in heart failure patients. Direct evidence from sympathetic neural recordings.

Digitalis glycosides exert both excitatory and inhibitory autonomic actions in animals and produce vasoconstriction in normal humans but produce vasodilation in heart failure patients. To determine whether or not these contrasting vascular responses are due to differing autonomic actions of the drug, we compared the responses to intravenous administration of Cedilanid-D (0.02 mg/kg) in eight normal subjects (mean age, 23 +/- 1 years) and eight patients with moderate-to-severe heart failure (mean age, 52 +/- 5 years, NYHA Class III-IV). Hemodynamics and efferent sympathetic nerve activity to mu

Pertussis toxin blocks the effects of alpha 2-agonists and antagonists on locus coeruleus activity in vivo.

This study assessed the effects of pertussis toxin, which is known to inactivate G proteins and therefore to block receptors linked to G proteins, on electrophysiological activity of the locus coeruleus in vivo. Pertussis toxin was injected into the lateral cerebral ventricle of rats, and locus coeruleus activity was then recorded. Compared to vehicle-injected control animals, pretreatment with pertussis toxin markedly increased the spontaneous firing rate of locus coeruleus neurons. In addition, the alpha 2-antagonist idazoxan was no longer able to augment either spontaneous or evoked locus c

Electrophysiological characteristics of lumbar spinal cord neurons backfired from lateral reticular nucleus in the rat.

Spinal neurons antidromically activated from either the lateral reticular nucleus (LRN) or immediately adjacent areas were identified in the rat lumbar spinal cord. In agreement with previous anatomical work (60), these neurons were widely distributed in both the dorsal and ventral horns of the spinal cord and could be subdivided into three main groups according to their location: a) deep ventromedial (DVM) cells, which project more substantially to the LRN than to other supraspinal targets; b) cells of the median portion of the neck of the dorsal horn (mNDH), which project exclusively to the

Sustained upgaze in coma.

Sustained upgaze is a rare phenomenon that is observed primarily in those patients with coma secondary to cardiac arrest or prolonged hypotension. Because of "selective vulnerability" of the brain, cardiac arrest or prolonged hypotension may produce both cerebral and cerebellar cortical damage sparing the brain stem vestibulo-ocular reflex (VOR) pathways. This selective damage may be responsible for the generation of sustained upgaze. The VORs receive their selective inhibition from the cerebellar flocculus. This inhibition is exerted on only one of the two VOR pathways converging on each extr

An inhibitory role for morphine on the release of dopamine into hypophysial portal blood and on the synthesis of dopamine in tuberoinfundibular neurons.

The intracerebroventricular administration of morphine to ovariectomized rats resulted in a marked decrease in the concentration of dopamine in plasma of hypophysial portal blood. A 90% reduction in the rate of release of hypothalamic dopamine into hypophysial portal blood occurred during the 60 min following the intraventricular administration of 60 ng of morphine sulfate. A dose-related decrease in the rate of release of dopamine into the portal vasculature was observed between 7.5 ng and 60 ng of morphine sulfate. Regardless of the quantity of morphine sulfate (1-500 ng) given to the animal

Influence of heart rate and inhibition of autonomic tone on the QT interval.

To evaluate whether heart-rate-induced changes of the QT interval are dependent on autonomic tone, we studied 13 healthy subjects, mean age 67.5 years. The maximal uncorrected QT from leads I, II, V1 and V6 was determined during atrial pacing at 90 beats/min and 130 beats/min before and after i.v. administration of propranolol, 0.1 mg/kg, and atropine, 0.02 mg/kg. Significant reductions (p less than 0.01) of QT were induced by the paced increases in heart rate before drugs (10%), after propranolol (10%) and after the combination of atropine and propranolol (9%). Propranolol caused no significa

[Detrusor inactivity in central spinal cord lesions. Hypothesis of inhibitory hyperreflexia of sympathetic origin].

In connection with two observations of suprasacral complete spinal cord patients showing a clear sublesional spasticity and paradoxically and inactive and unreactive detrusor, the authors discuss the mechanisms that are usually considered: vesical distension, sacral peripheric neurogenic lesion, reflex inhibition of the detrusor of somatic origin. They express the hypothesis of an inhibition hyper-reflex of sympathetic origin, reversible under phenoxybenzamine treatment, consider the urodynamic evaluation and the therapeutic treatment and compare this type of neurogenic vesical dysfunction to

Dickinson W. Richards Lecture: Circulatory adjustments to hypoxia.

Circulatory adjustments during hypoxia act to redistribute blood flow and maintain arterial pressure. Redistribution of blood flow is accomplished by a local effect of hypoxia, which produces dilatation in coronary and cerebral vessels, and the chemoreceptor reflex, which produces vasoconstriction in skeletal muscle and the splanchnic bed and dilatation in coronary vessels. Arterial pressure is maintained primarily by the chemoreceptor reflex. If the chemoreceptor reflex fails to maintain arterial pressure, hypoxia and hypotension together activate the central pressor response. Compensatory me

The influence of naloxone on the C fiber response of dorsal horn neurons and their inhibitory control by raphe magnus stimulation.

In intact rats anesthetized with chloralose, the effects of naloxone were studied on the responses of spinal cord dorsal horn neurons to C fiber stimulation and upon the inhibition induced on these responses by stimulation of the nucleus raphé magnus (NRM). (1) A mean 44% facilitatory effect on responses to C fibers was observed for 12/19 units. (2) A mean 30% reduction of the inhibitory effects of NRM was found for 14/29 units. (3) However there is no clear relationship between these facilitatory effects and the diminution of the efficiency of NRM stimulation. These results demonstrate a faci

Increase by visual stimuli in turnover of 5-hydroxytryptamine in the superior colliculi of the rabbit.

1. Irregular light falshes were played on to one eye of dark adapted rabbits for periods of 20-80 min. The concentration of 5-hydroxyindol-3-ylacetic acid (5-HIAA) and of 5-hydroxytryptamine (5-HT) were estimated in left and right superior colliculi, thalami and hippocampi. 2. In rabbits exposed to such visual stimuli for 30-60 min, there was an increase in the 5-HIAA content of the colliculus contralateral to the stimulated retina which aberaged 17% (P = 0-02), but no rise was seen if the exposure was shortened to 20 or prolonged to 80 min. At no time was there a difference in 5-HIAA content

Does the engram of kindling model the engram of normal long term memory?

The kindling effect is a relatively permanent alteration in brain function which results from repeated electrical or chemical stimulation and culminates in the appearance of electrographic and behavioral convulsions whenever the original stimulus is reapplied. The effect results from tetanic activation in the anterior cortex, limbic system or associated areas of the adult mammalian brain, and the lasting alterations are transynaptic and quite widespread. They are based in part on synaptic facilitation, and they are accompanied by specific alterations in normal behavior. In these and other resp

Generation of Blood Vascular Endothelial-Neural 3D Organoids by Serial Induction of Differentiation on Human iPSC-Derived Embryoid Bodies.

The 3D brain organoids have been widely used as a tool to study human brain development and disorders. Although angiogenesis and blood vascular endothelial cells play important roles in brain development and pathogenesis in neurological disorders, most 3D brain organoids lack inherent endothelial cells and need either the addition of differentiated endothelial cells or to be transplanted to animals to reconstitute such vascular structures. However, these approaches could miss the developmental interactions between angiogenesis and neurogenesis in the human brain. To reconstitute a 3D organoid

HD-tACS over the left frontal aslant tract entrains theta activity associated with speech motor control.

Transient disruption or permanent damage to the left Frontal Aslant Tract (FAT) is associated with deficits in speech production. The present study examined the application of theta (4 Hz) high-definition transcranial alternating current stimulation (HD-tACS) over the left SMA and IFG -as a part of FAT- as a potential multisite protocol to modulate neural and behavioral correlates of speech motor control. Twenty-one young adults participated in three counterbalanced sessions in which they received in-phase, anti-phase, and sham theta HD-tACS. In each session, 4 Hz stimulation was applied over

Sodium valproate augments spontaneous neural tube defects and axial skeletal malformations in TO mouse fetuses [corrected].

The TO mouse exhibits a low incidence (3.65%) of spontaneous exencephaly at birth. The objectives of this study were to determine if sodium valproate (VPA) would augment this background frequency of exencephaly and to characterize its gross and histologic bases. Single doses of 200, 400, or 600 mg/kg of VPA were administered on one of gestation days (GD) 7 to 10 and fetuses were collected on GD 18. Significant augmentation of the background incidence of exencephaly was observed in the GD 7 and 8 treatment groups. Absence of the skull vault, hemorrhage, and degeneration of the exposed brain, po

A Preparatory Cranial Potential for Saccadic Eye Movements in Macaque Monkeys.

Response preparation is accomplished by gradual accumulation in neural activity until a threshold is reached. In humans, such a preparatory signal, referred to as the lateralized readiness potential (LRP), can be observed in the EEG over sensorimotor cortical areas before execution of a voluntary movement. Although well described for manual movements, less is known about preparatory EEG potentials for saccadic eye movements in humans and nonhuman primates. Hence, we describe a LRP over the frontolateral cortex in macaque monkeys. Homologous to humans, we observed lateralized electrical potenti

Dynamic acetylation profile during mammalian neurulation.

BACKGROUND: Neural tube defects (NTDs) result from failure of neural tube closure during embryogenesis. These severe birth defects of the central nervous system include anencephaly and spina bifida, and affect 0.5-2 per 1,000 pregnancies worldwide in humans. It has been demonstrated that acetylation plays a pivotal role during neural tube closure, as animal models for defective histone acetyltransferase proteins display NTDs. Acetylation represents an important component of the complex network of posttranslational regulatory interactions, suggesting a possible fundamental role during primary n

Defective cholesterol traffic and neuronal differentiation in neural stem cells of Niemann-Pick type C disease improved by valproic acid, a histone deacetylase inhibitor.

Niemann-Pick type C disease (NPC) is a neurodegenerative and lipid storage disorder for which no effective treatment is known. We previously reported that neural stem cells derived from NPC1 mice showed impaired self-renewal and differentiation. We examined whether valproic acid (VPA), a histone deacetylase inhibitor, could enhance neuronal differentiation and recover defective cholesterol metabolism in neural stem cells (NSCs) from NPC1-deficient mice (NPC1(-/-)). VPA could induce neuronal differentiation and restore impaired astrocytes in NSCs from NPC1(-/-) mice. Importantly, an increasing
