En Face OCT and the Phenotype Characterization of Drusen.

PURPOSE: To detect and differentiate the various subtypes of drusen and subretinal drusenoid deposits (SDDs) using single-capture en face sp … nuated signal representing soft drusen. CONCLUSIONS: En face OCT is a simple and practical tool to phenotype the various classes of drusen and SDDs. Composition differences between the various drusen and SDDs can be captured with en face OCT at varying segmentation levels due to differences in light reflectance and penetration characteristics. These OCT fe …

Skeletal muscle phenotype and blood pressure after acute exercises.

PURPOSE: The inter-individual responses in systolic blood pressure (SBP) vary from no change to marked reduction after acute exercise. We hypothesized that skeletal muscle phenotype modifies SBP responses. METHODS: Normotensive participants (12 males and 8 females, age 27 ± 6 years) performed two 30-min exercises, continuous and interval, by cycle ergometer on separate days. The area under the curve (AUC) for SBP was calculated until 60 min after exercise. Intensity of continuous exercise was 60% of peak exercise workload and interval exercise 8 × 1 min intervals (80-90% of peak exercise work…

Hypoxia-Induced Changes in Endothelial Cell Phenotype and Function.

Significance: Endothelial cells (ECs) are specialized cells lining the interior surface of blood vessels, playing a crucial role in vascular biology. They exhibit remarkable versatility, adapting to various tissue requirements. Their ability to respond to physiological and pathological stimuli ensures proper tissue function and homeostasis. Recent Advances: Hypoxia is when the oxygen level in a given organ, tissue, or cell type drops below the physiological level and is insufficient to maintain adequate homeostasis. ECs respond to hypoxia by activating various mechanisms. Hypoxia-induced chang

Phenotype response for the invasive Petaurus notatus in Tasmania.

A mainland native nocturnal arboreal glider Petaurus notatus (Krefft's Glider) was introduced to Tasmania in the 1830s as a pet and its subsequent escape and landscape spread has been associated with the predation of endemic and migratory birds. Using geometric morphometrics, we assessed whether P. notatus experienced a phenotypic response, visible in their skull, after introduction to Tasmania. We found significant skull form variation between Tasmanian (n = 57) and Victorian (n = 102) glider specimens. Specifically, there is an increase in rostrum length-particularly at the mid-cranium-a dor

Genotype-phenotype correlation in multiple endocrine neoplasia type 1.

BACKGROUNDAmong patients with multiple endocrine neoplasia type 1 (MEN1), 80% develop duodenopancreatic neuroendocrine tumors (dpNETs), of whom 15%-25% die of metastasis. There is a need to identify biomarkers to predict aggressive disease. MEN1 genotype affords an attractive possibility as a biomarker, as it remains constant during life. Currently, patients are clinically diagnosed with MEN1 by the presence of ≥2 primary endocrine tumors (pituitary, parathyroid, and pancreas) or ≥1 primary endocrine tumor with a positive family history. From 10% to 30% of patients diagnosed clinically with ME

Cannabidiol polarizes human neutrophils toward a cancer-promoting phenotype.

INTRODUCTION: Cannabidiol (CBD) is widely used as a natural alternative supplementary treatment for side effects and symptom relief in many … TS: We show that CBD induces human primary neutrophils to polarize into an N2-like cancer-promoting phenotype. CBD-exposed neutrophils exhibit reduced oxidative burst, reduce bacterial killing, and altered the production of cytokine and chemokine arrays like N2-polarized cells. CBD-treated cells also rapidly display a landscape of surface markers compatible …

C4 photosynthesis, trait spectra, and the fast-efficient phenotype.

It has been 60 years since the discovery of C4 photosynthesis, an event that rewrote our understanding of plant adaptation, ecosystem responses to global change, and global food security. Despite six decades of research, one aspect of C4 photosynthesis that remains poorly understood is how the pathway fits into the broader context of adaptive trait spectra, which form our modern view of functional trait ecology. The C4 CO2-concentrating mechanism supports a general C4 plant phenotype capable of fast growth and high resource-use efficiencies. The fast-efficient C4 phenotype has the potential to…

Epilepsy expands the phenotype of L-arginine:glycine amidinotransferase deficiency.

OBJECTIVE: L-arginine:glycine amidinotransferase (AGAT) deficiency is a rare autosomal recessive disorder affecting creatine biosynthesis, leading to developmental delay, intellectual disabilities, and myopathy. Unlike other creatine deficiency disorders, its link to epilepsy remains uncertain. This study presents the first reported epilepsy cases in AGAT deficiency, analyzing seizure patterns and response to creatine monohydrate supplementation. METHODS: We retrospectively analyzed two AGAT-deficient probands identified through a national collaboration. Biochemical assessments of creatine an

3-M syndrome: evolution of the phenotype over time.

BACKGROUND: 3-M syndrome is an autosomal recessive disease characterized by short stature, facial dysmorphism and skeletal anomalies. To date, biallelic pathogenic CUL7 variants are responsible for the majority of cases, but biallelic deleterious changes in OBSL1 and CCDC8 can also establish the diagnosis. CASES PRESENTATION: We report two unrelated newborns showing clinical signs compatible with 3-M syndrome and we describe the evolution of the phenotype of the first patient over time. Molecular analysis identified two compound heterozygous CUL7 variants in the first individual and a homozygo…

The Opposite Phenotype of Sotos Syndrome: 5q35.2q35.3 Microduplication Syndrome.

Sotos syndrome is a genetic disorder resulting from heterozygous pathogenic variants or deletions in the nuclear receptor-binding SET domain protein 1 (NSD1) gene. It is characterized by prenatal and postnatal overgrowth, macrocephaly, distinctive craniofacial features, learning disability, and advanced bone maturation. In contrast, Reverse Sotos syndrome, arises from duplications within the NSD1 gene, presenting with an opposite clinical phenotype, including microcephaly, developmental delay, short stature and delayed bone maturation. To date, the reverse clinical phenotype associated with th…

Transformation of Thyroid Eye Disease Phenotype Following Teprotumumab Treatment.

The authors report a case of a phenotypic shift of thyroid eye disease following treatment with teprotumumab. A 35-year-old female nonsmoker with Graves disease developed mild type I thyroid eye disease featuring unilateral lid retraction and was treated with teprotumumab. Following completion of treatment, a reactivation was associated with progression to severe type II phenotype, with marked extraocular muscle enlargement, bilateral proptosis, and nonsteroid-responsive compressive optic neuropathy requiring orbital decompression. This case raises important questions regarding the potential i…

Genotype-Phenotype Correlations in the Hyperparathyroidism-Jaw Tumor Syndrome.

Establishing genotype-phenotype correlations in disorders of hereditary endocrine neoplasia is important for clinical screening, genetic counseling, prognostication, surveillance, and surgical strategy, and may also provide clues about disease pathogenesis. Important genotype-phenotype correlations are recognized, for example, in pheochromocytoma/paraganglioma and multiple endocrine neoplasia type 2A. The presence of such correlations has been less clear in other familial endocrine disorders associated with primary hyperparathyroidism including multiple endocrine neoplasia type 1, and the hype…
