Evolution of compound eye cell types shapes visual behaviors across Heliconius butterflies.

The evolution of visual systems is tightly linked to the diversification of visually guided behaviors. Using Heliconius butterflies, renowned for their rapid diversification of wing color patterns and corresponding visual mate preferences, we investigate the evolution of compound eye cell types across a complete speciation continuum. By integrating population genomics, chromatin conformation capture, and single-nucleus multi-omics, we uncover multi-modal mechanisms driving visual system evolution. Within a population polymorphic for mate preference, we identify senseless-2 as a glia-expressing

Long-term daily light exposure boosts human retinal organoid differentiation.

Induced pluripotent stem cell-derived retinal organoids (ROs) have become promising personalized models to study inherited retinal diseases (IRDs) and develop new therapies. Although ROs mimic key retinal features and show some light responsiveness, their differentiation and maturation remain limited and lengthy. We hypothesize that standard dark culture conditions limit the expression of genes and proteins related to retinal function, delaying differentiation, and hindering disease modeling. Here, we investigate whether daily light exposure promotes photoreceptor maturation by exposing ROs to

Ocular Ciliopathies: Genetic and Mechanistic Insights into Developing Therapies.

Developing suitable medicines for genetic diseases requires a detailed understanding of not only the pathways that cause the disease, but also the identification of the genetic components involved in disease manifestation. This article focuses on the complexities associated with ocular ciliopathies - a class of debilitating disorders of the eye caused by ciliary dysfunction. Ciliated cell types have been identified in both the anterior and posterior segments of the eye. Photoreceptors (rods and cones) are the most studied ciliated neurons in the retina, which is located in the posterior eye. T

Restoration of patterned vision with an engineered photoactivatable G protein-coupled receptor.

Retinitis pigmentosa results in blindness due to degeneration of photoreceptors, but spares other retinal cells, leading to the hope that expression of light-activated signaling proteins in the surviving cells could restore vision. We used a retinal G protein-coupled receptor, mGluR2, which we chemically engineered to respond to light. In retinal ganglion cells (RGCs) of blind rd1 mice, photoswitch-charged mGluR2 ("SNAG-mGluR2") evoked robust OFF responses to light, but not in wild-type retinas, revealing selectivity for RGCs that have lost photoreceptor input. SNAG-mGluR2 enabled animals to d

The microglia in healthy and diseased retina.

The microglia are the immune cells of the central nervous system and, also the retina. They fulfil several tasks of surveillance in the healthy retina. In case of an injury or disease, microglia become activated and tries to repair the damage. However, in a lot of cases it does not work, and microglia deteriorate the situation by releasing toxic and pro-inflammatory compounds. Moreover, they further promote degenerative processes by attacking and phagocytosing damaged neurones and photoreceptors that otherwise would possibly have the chance to survive. Such deleterious action of the microglia

Effects of nitric oxide on horizontal cells in the rabbit retina.

Retinal horizontal cells display large receptive fields as a result of extensive electrical coupling via gap junctions. There is abundant evidence that these gap junctions are dynamically regulated by changes in the adaptational state of the retina. The neuromodulator dopamine appears to play a major role in regulating gap junctional conductances of horizontal cells. Emerging evidence indicates that nitric oxide (NO) also acts as a neuromodulator in the retina and, more specifically, regulates the coupling between horizontal cells. In the present study, we examined the effects of a nitric oxid

Semiconducting Polymer Blend Films for Photosensitive Subretinal Prosthesis Design-Properties, Biocompatibility, and Impact of Light on Polymer-Cell Interactions.

This study examines polymer-based semiconducting materials as functional candidates for subretinal prosthetic interfaces designed to restore visual function in individuals affected by photoreceptor degeneration. Polymer blend thin films designed to couple high photosensitivity with efficient charge transport and favourable biological interactions were characterized in terms of their physicochemical and optoelectronic properties, stability under physiological conditions, and cytocompatibility with retinal pigment epithelium (RPE) cells. Because retinal cells operate under variable illumination,

The ultrastructural pathology of congenital murine toxoplasmic retinochoroiditis. Part II: The morphology of the inflammatory changes.

A congenital murine model of toxoplasmic retinochoroiditis was employed to study the ultrastructural pathology of retinal parasitization by Toxoplasma gondii. Forty-two eyes from infected mice (18-22-weeks-old) and 24 eyes from control animals were studied by light microscopy (semithin sections). Twenty-six of the eyes from infected animals and six from the control group were subsequently selected for transmission electron microscopy. Control tissues showed no significant abnormality. The pathological changes in diseased tissues ranged in severity from low-grade mononuclear cell infiltration i

Light-modulated stem cells in the camera-type eye of an annelid model for adult brain plasticity.

Camera-type eyes in vertebrates and cephalopods are striking examples of parallel evolution of a complex structure. While comparisons have focused on these two groups, camera-type eyes with likely high functionality are also found in other invertebrate phyla with simpler brains. Employing single-cell RNA sequencing, we identify neurogenic cells in the adult eyes and brain of the marine annelid worm Platynereis dumerilii. Distinct neural stem cells in the camera-type adult eyes, located at the edge of the cup-shaped retina, and adjacent to the glass body/lens, produce radial lines of cells, rem

Optogenetic targeting of AII amacrine cells restores retinal computations performed by the inner retina.

Most inherited retinal dystrophies display progressive photoreceptor cell degeneration leading to severe visual impairment. Optogenetic reactivation of inner retinal neurons is a promising avenue to restore vision in retinas having lost their photoreceptors. Expression of optogenetic proteins in surviving ganglion cells, the retinal output, allows them to take on the lost photoreceptive function. Nonetheless, this creates an exclusively ON retina by expression of depolarizing optogenetic proteins in all classes of ganglion cells, whereas a normal retina extracts several features from the visua

Pyrroloquinoline Quinone Protects Against Light-Induced Retinal Damage in Association with the Suppression of c-Fos Signalling.

Age-related macular degeneration (AMD) is a progressive retinal disorder characterised by oxidative stress and inflammation. Although pyrroloquinoline quinone (PQQ) has been reported to exert neuroprotective effects, its specific efficacy in in vivo models of AMD pathophysiology has not yet been elucidated. In this study, we evaluated the protective effects of PQQ against all-trans-retinal (ATR)-induced cytotoxicity in ARPE-19 cells and light-induced photoreceptor degeneration in rats. Pretreatment of ARPE-19 cells with PQQ dose-dependently mitigated ATR-induced cytotoxicity. In the in vivo mo

Optogenetic vision restoration in the face of secondary and tertiary remodeling in the rd1 mouse retina.

Photoreceptor loss in retinal degeneration is followed by progressive remodeling of the surviving retina, which may present a barrier to vision restoration. To determine the impact of remodeling on the retina's suitability for therapeutic interventions, we track changes in visual code produced by the optogenetic actuator ReaChR expressed in ON-bipolar cells of the rd1 mouse at 3, 5, 9, and 12 months. Anatomical analyses confirm that these ages encompass phase II (photoreceptor degeneration) to phase III (inner retinal thinning, dysmorphia, pigment epithelium infiltration) remodeling. Multi-ele

Molecular profiling of resident and infiltrating mononuclear phagocytes during rapid adult retinal degeneration using single-cell RNA sequencing.

Neuroinflammation commonly accompanies neurodegeneration, but the specific roles of resident and infiltrating immune cells during degeneration remains controversial. Much of the difficulty in assessing myeloid cell-specific functions during disease progression arises from the inability to clearly distinguish between activated microglia and bone marrow-derived monocytes and macrophages in various stages of differentiation and activation within the central nervous system. Using an inducible model of photoreceptor cell death, we investigated the prevalence of infiltrating monocytes and macrophage

Ush1c gene expression levels in the ear and eye suggest different roles for Ush1c in neurosensory organs in a new Ush1c knockout mouse.

Usher syndrome (USH) is the most common form of deaf-blindness in humans. Molecular characterization revealed that the USH gene products form a macromolecular protein network in hair cells of the inner ear and in photoreceptor cells of the retina via binding to PDZ domains in the scaffold protein harmonin encoded by the Ush1c gene in mice and humans. Although several mouse mutants for the Ush1c gene have been described, we generated a targeted null mutation Ush1c mouse model in which the first four exons of the Ush1c gene were replaced with a reporter gene. Here, we assessed the expression pat…

Ultrastructure and organization of circumventricular organs and endolymphatic tubules in the treefrog, Hyla versicolor.

The morphology and anatomical relationship of the paraphysis, anterior choroid plexus, and endolymphatic tubules of Hyla versicolor suggest that these structures may act as a functional unit to regulate composition of the cerebrospinal fluid (CSF). Ependymal cells of both the paraphysis and the choroid plexus exhibit ultrastructural features (an extensive microvillar border, large intercellular spaces containing an amorphous material, and apically located mitochondria) which indicate active exchange between the blood and CSF. Mast cells (which may play a role in regulating vascular permeabilit

Signal processing in a simple vertebrate photoreceptor system: the teleost pineal organ.

The integrating circuitry and efferent pathways for neural signals evoked in the photosensory pineal organ by changes in ambient illumination have been investigated by a multidisciplinary approach. Intrapineal efferent neurons were identified by means of retrograde filling with horseradish peroxidase (HRP). In addition to several types of neurons, photoreceptor cells that emitted axons to the brain via the pineal tract were observed. The presence of several populations of local interneurons (putatively cholinergic, GABAergic and substance P-containing) and possible afferent (putatively noradre…

Thyroid hormone receptor beta-2 (TRβ2) overexpression modulates photoreceptor phenotype diversity in a ligand-dependent manner.

BACKGROUND: Vertebrate color vision results from the specification of photoreceptor subtypes expressing distinct opsins. Thyroid hormone (TH) and its receptor TRβ2 are essential regulators of long-wavelength-sensitive (LWS) cone development, but their ligand-dependent roles in regulating cone subtype fate remain unclear. METHODS: We investigated how varying TH availability and TRβ2 overexpression impact cone photoreceptor diversity and opsin expression using a gain-of-function transgenic zebrafish line (crx:trβ2), which expresses TRβ2 in all photoreceptors, and manipulated TH levels through T

Glutamine is the only amino acid that can adequately support the generation of NADPH in rod photoreceptors.

NADPH, the primary source of reducing equivalents in the cytosol, is used in vertebrate rod photoreceptor outer segments to reduce the all-trans retinal released from photoactivated visual pigment to all-trans retinol. Light activation of the visual pigment isomerizes the 11-cis retinal chromophore to all-trans, thereby destroying it and necessitating its regeneration. Release and reduction of all-trans retinal are the first steps in the series of reactions that regenerate the visual pigment. Glucose and glutamine can both support the reduction of all-trans retinal to retinol, indicating that

Early photoreceptor outer segment loss and retinoschisis in Cohen syndrome.

PURPOSE: To describe early structural and functional retinal changes in a patient with Cohen syndrome. METHODS: A 13-month-old Caucasian girl of Irish and Spanish ancestry was noted to have micrognathia and laryngomalacia at birth, which prompted a genetic evaluation that revealed biallelic deletions in COH1 (VPS13B) (a maternally inherited 60-kb deletion involving exons 26-32 and a paternally inherited 3.5-kb deletion within exon 17) consistent with Cohen syndrome. She underwent a complete ophthalmic examination, full-field flash electroretinography and retinal imaging with spectral domain o

Arap1 Deficiency Causes Photoreceptor Degeneration in Mice.

PURPOSE: Small guanosine triphosphatase (GTPase) ADP-ribosylation factors (Arfs) regulate membrane traffic and actin reorganization under the control of GTPase-activating proteins (GAPs). Arap1 is an Arf-directed GAP that inhibits the trafficking of epidermal growth factor receptor (EGFR) to the early endosome, but the diversity of its functions is incompletely understood. The aim of this study was to determine the role of Arap1 in the mammalian retina. METHODS: Genetically engineered Arap1 knockout mice were screened for ocular abnormalities in the National Institutes of Health Knockout Mous
