Primary structure of a new actin-binding protein from human seminal plasma.

Secretory actin-binding protein (SABP), a glycoprotein from human seminal plasma, was isolated according to Akiyama and Kimura [Akiyama, K. & Kimura, H. (1990) Biochim. Biophys. Acta 1040, 206-210]. The complete amino acid sequence of SABP was determined with the aid of fragments generated by trypsin, Staphylococcus aureus V8 protease and pepsin. The single polypeptide chain of SABP contains 118 amino acids with a calculated Mr of 13,506 and pyroglutamic acid as the N-terminal residue. A single N-glycosidic carbohydrate moiety is located at Asn77. The carbohydrate composition shows an unusuall

UV damage-specific DNA-binding protein in xeroderma pigmentosum complementation group E.

The gel mobility shift assay method revealed a specifically ultraviolet (UV) damage recognizing, DNA-binding protein in nuclear extracts of normal human cells. The resulted DNA/protein complexes caused the two retarded mobility shifts. Four xeroderma pigmentosum complementation group E (XPE) fibroblast strains derived from unrelated Japanese families were not deficient in such a DNA damage recognition/binding protein because of the normal complex formation and gel mobility shifts, although we confirmed the reported lack of the protein in the European XPE (XP2RO and XP3RO) cells. Thus, the abse

Liver fatty acid-binding protein in two cases of human lipid storage.

FABPs in the various tissues play an important role in the intracellular fatty acid transport and metabolism. Reye's syndrome (RS) and multisystemic lipid storage (MLS) are human disorders characterized by a disturbance of lipid metabolism of unknown etiology. We investigated for the first time L-FABP in these two conditions. Affinity purified antibodies against chicken L-FABP were raised in rabbits, and found to cross-react specifically with partially purified human L-FABP. L-FABP content in liver samples of two patients with RS and MLS was investigated by immuno-histochemistry, SDS-PAGE and

Insulin-like growth factor receptors and binding protein in rat neuroblastoma cells.

B104, an established rat neuroblastoma cell line exhibiting specific neuronal qualities, was chosen as a model to study insulin-like growth factor (IGF) binding and action in the central nervous system. Specific binding of [125I]IGF-II to B104 membranes averaged 12.2 +/- 4.0% (mean +/- SD)/100 micrograms/ml protein compared with [125I]IGF-I binding of 10.1 +/- 2.9%. In competitive binding studies employing [125I]IGF-II as the radioligand, high affinity for IGF-II was demonstrated (50% displacement at 2.7 ng/ml), with none for IGF-I or insulin. Upon cross-linking [125I]IGF-I to membranes under

Defective phosphorylation of a calmodulin-binding protein in cystic-fibrosis submandibular glands.

Calmodulin-binding proteins in fractions purified from human submandibular glands by calmodulin-Sepharose were phosphorylated with [gamma-32P]ATP, in the absence of exogenous protein kinase. The major proteins phosphorylated had molecular masses of 45, 51 and 61 kDa. Phosphorylation was increased by activators of protein kinase C and inhibited by H-7. Phosphorylation of the 61 kDa band was markedly decreased in cystic-fibrosis submandibular glands.

[Clinical significance of competitive protein binding assay of 25-hydroxyvitamin D concentration].

The level of serum 25-hydroxyvitamin D (25-OH-D3) was determined in 94 cases of healthy volunteers and 98 patients with various diseases. Th … 110.3 +/- 22.5 ng/ml, being much higher than that of the controls (P less than 0.001). Competitive protein binding assay for measuring 25-OH-D3 is a simple and economic method. It is sensitive and specific as it provides distinct discrimination between healthy controls and patients with vitamin D deficiency or overdosage.

Calcium-binding and aggregation properties of parathyroid secretory protein-I (chromogranin A).

Secretory protein-I (SP-I, also referred to as chromogranin A) is a major acidic, sulfated glycoprotein of the parathyroid gland secretory granule. It is cosecreted with parathormone under calcium regulation. SP-I is similar if not identical to chromogranin A of the adrenal medulla chromaffin cell and has been found in most endocrine cells. Its function is unknown, but it may play a role in the packaging of secreted substances. We have examined the calcium-binding properties of purified bovine SP-I and find that it binds 14 or more Ca2+ at low affinity. Measurements of sedimentation equilibriu

Protein binding studies of technetium-99m-labeled phosphine and isocyanide cationic complexes.

Most 99mTc/phosphine/isocyanide complexes synthesized to date show preferential uptake by the myocardium of many animal species but not in man. A new complex has been synthesized, [99mTc(DEPE)2(CNR)2], +(DEPIC), where R = t - butyl isocyanide, which in three animal species images the myocardium very well, but in humans it remains primarily in the blood pool. One reason for the difference in the behavior of these complexes in different species could be the characteristics of their binding to plasma proteins. The protein binding characteristics of DEPIC and two other well-known complexes have th…

A plasma steroid hormone binding protein in the viviparous water snake, Nerodia.

A plasma steroid binding protein (SHBP) with medium-high affinity and limited capacity was characterized in the viviparous water snake, Nerodia. This SHBP shows similarity to SHBPs previously described in some other nonmammalian species. A single binding component was detected by Scatchard analyses with a medium-high affinity for testosterone (T), estradiol (E), progesterone (P), and corticosterone (B). Equilibrium dissociation constants (Kd) for these steroids are as follows: T, 3.6 x 10(-8) M; E, 3.7 x 10(-8) M; P, 5.9 x 10(-8) M; B, 12.1 x 10(-8) M. In competition studies (at saturation) th

Human endometrial antioestrogen-binding protein: characterization and levels during the menstrual cycle.

Human endometrial cytosol (90,000 g, 60 mins) contains a protein that binds non-steroidal anti-oestrogens with high specificity and high affinity (Kd = 10(-10)M). It binds oestrogens and other steroid hormones poorly, and can be physically distinguished from the oestrogen receptor protein by gel filtration and ion exchange chromatography. The endometrial anti-oestrogen binding protein is larger and less negatively charged than the oestrogen receptor. In this regard, it resembles the human myometrial anti-oestrogen binding protein. Unlike the myometrial protein, however, levels of the endometri

Isolation and partial characterization of a 110-kD dimer actin-binding protein.

Two Triton-insoluble fractions were isolated from Acanthamoeba castellanii. The major non-membrane proteins in both fractions were actin (30-40%), myosin II (4-9%), myosin I (1-5%), and a 55-kD polypeptide (10%). The 55-kD polypeptide did not react with antibodies against tubulins from turkey brain, paramecium, or yeast. All of these proteins were much more concentrated in the Triton-insoluble fractions than in the whole homogenate or soluble supernatant. The 55-kD polypeptide was extracted with 0.3 M NaCl, fractionated by ammonium sulfate, and purified to near homogeneity by DEAE-cellulose an

Histochemical observation of F 1881 binding protein in human benign prostatic hypertrophy.

A 3-carboxymethyloxime conjugate of R 1881 was conjugated with bovine serum albumin and reacted with fluorescein isothiocyanate. Tissue sections from patients with benign prostatic hypertrophy were incubated with this compound. Fluorescence was observed in the cytoplasm of gland cells, although both positive and negative cells were distributed evenly in some areas. Neither the stroma nor the nuclei of epithelial cells were fluorescent. Preincubation with a 20 fold molar excess of R 1881-carboxymethyloxime-bovine serum albumin, 1 microM of R 1881, or 1 microM of triamcinolone acetonide diminish

Lymphocytes binding C-reactive protein and streptococcal membranes in acute rheumatic fever.

Peripheral blood lymphocytes binding CRP and streptococcal membrane antigens were studied in 19 patients with ARF with the use of a number of paired analyses for cell surface markers. A small fraction (6.9%) of T cells showed binding to streptococcal membrane. A large degree of overlap (38% to 46%) occurred in membrane-binding T cells and those with identifiable CRP. One third of T gamma cells showed CRP bindings, and approximately one quarter to T gamma cells in both ARF patients and normal controls showed membrane la antigen. A considerable concordance (50% to 75%) was noted between T cells

Influence of chronic renal failure and hemodialysis on diflunisal plasma protein binding.

Diflunisal protein binding was studied by equilibrium dialysis at 37 degrees in plasma from healthy, uremic, and geriatric subjects. Binding data were computer analyzed assuming 2 classes of independent binding sites (Scatchard model). K1, the primary association constant for the diflunisal-albumin interaction, was substantially lower in uremic plasma (2.39 +/- 0.29 x 10(5) M-1) than in normal plasma (6.86 +/- 0.59 x 10(5) M-1). No difference was found between the number of primary diflunisal binding sites (N1) in uremic and normal plasma. In geriatric plasma neither K1 nor N1 differed from th…

The potent anti-HIV protein cyanovirin-N contains two novel carbohydrate binding sites that selectively bind to Man(8) D1D3 and Man(9) with nanomolar affinity: implications for binding to the HIV envelope protein gp120.

Cyanovirin-N (CVN) is a monomeric 11 kDa cyanobacterial protein that potently inactivates diverse strains of human immunodeficiency virus (HIV) at the level of cell fusion by virtue of high affinity interactions with the surface envelope glycoprotein gp120. Several lines of evidence have suggested that CVN-gp120 interactions are in part mediated by N-linked complex carbohydrates present on gp120, but experimental evidence has been lacking. To this end we screened a comprehensive panel of carbohydrates which represent structurally the N-linked carbohydrates found on gp120 for their ability to i

Y-box-binding protein 1 stabilizes the capsid protein of porcine circovirus type 2 to promote viral replication.

Porcine circovirus type 2 (PCV2), a major causative agent of PCV2-associated diseases, poses a serious threat to the global swine industry. The stability of PCV2 capsid (Cap) protein is critical for viral replication; however, the underlying mechanisms regulating Cap stability during PCV2 infection remain poorly understood. In this study, co-immunoprecipitation assays were used to demonstrate that PCV2 Cap protein can maintain its own stability by binding to the cellular Y-box-binding protein 1 (YBX1) during viral infection. Furthermore, binding domain mapping experiments revealed two specific
