The first and second phases of the human microbiome project provided a view of mucosal surfaces and the skin of humans that mapped an abundant and complex ecosystem (microbiota) that is composed of bacteria (bacteriobiota), fungi (mycobiota), viruses (virobiota), enteric phages, archaea, protists, and helminths. Intestinal dysbiosis describes an adverse shift in microbial homeostasis in the gut that enhances intestinal epithelial permeability, translocating toxins that may lead to endotoxemia. Numerous intestinal and extra-intestinal illnesses have been linked to gut dysbiosis, including infla

Hidradenitis suppurativa (HS) is a chronic inflammatory skin condition, primarily affecting young individuals, with a significant impact on their quality of life due to recurrent, painful nodules, abscesses, and oozing sinus tracts, primarily affecting intertriginous areas. The pathogenesis of HS is multifactorial, involving a complex interplay between genetic predisposition, immune dysregulation, microbial, and environmental factors. While it is known that cutaneous and gut microbiome contribute to innate immune dysregulation in HS, their precise involvement in disease pathogenesis remains un

A comprehensive characterization of the human body resistome [sets of antibiotic resistance genes (ARGs)] is yet to be done and paramount for addressing the antibiotic microbial resistance threat. Here, we study the resistome of 771 samples from five major body parts (skin, nares, vagina, gut, and oral cavity) of healthy subjects from the Human Microbiome Project (HMP) and addressed the potential dispersion of ARGs in pristine environments. A total of 28,714 ARGs belonging to 235 different ARG types were found in the HMP proteome dataset (n = 9.1 × 107 proteins analyzed). Our study reveals a d

Atopic dermatitis (AD) is a chronic immune-mediated inflammatory skin disease. An overgrowth of Staphylococcus aureus (S. aureus) and decreased microbial diversity is apparent in 70%-90% of AD patients. SA.100 is a recombinant endolysin targeting S. aureus that might be a novel treatment for patients with mild-to-moderate AD. To test safety, pharmacodynamics and efficacy of SA.100 a double-blind, randomized, vehicle-controlled trial in 53 subjects with mild-to-moderate AD was performed. Patients were randomized equally to topical SA.100 or vehicle with stratification for S. aureus positivity.

The understanding of atopic dermatitis (AD) pathogenesis is evolving beyond the barrier-immune dichotomy, with neuropsychological factors gaining prominence as a central component. This review systematically decodes the multi-level crosslink within the "brain-skin axis" in AD, encompassing recent advances in peripheral sensory neuron sensitization, central neural remodeling, and neuro-immune interactions. We elaborate on the roles of neuropeptides, cytokines, the autonomic nervous system, and the hypothalamic-pituitary-adrenal axis in the itch-scratch cycle, inflammation amplification, and moo

Epidermolysis bullosa (EB) is a rare inherited genodermatosis that affects both the skin and gastrointestinal tract, potentially leading to altered gut microbiota. This study aimed to assess the diversity and composition of the gut microbiome in EB patients compared to healthy controls. Twelve EB patients-six with dystrophic epidermolysis bullosa patients (pDEB) and six with nondystrophic epidermolysis bullosa patients (pNDEB)-and 12 age- and gender-matched healthy controls were included. Fecal samples were collected, and 16S rRNA gene sequencing (V3-V4 region) was performed. Alpha diversity i

E-cigarettes are widely perceived as a safer alternative to tobacco, and that perception often extends to their use around infants and children. Yet exhaled aerosol deposits onto indoor surfaces, leaving a residue, known as third-hand e-cigarette aerosol, that can be re-emitted, ingested, or absorbed through the skin. The developmental consequences of such exposure have been almost entirely unexplored. In this issue of Pediatric Research, Zaman et al. begin to address that gap, exposing neonatal mice to third-hand e-cigarette aerosol-contaminated terrycloth and carpet in early-life. The author

Erythrodermic psoriasis(EP) is a rare, life-threatening variant affecting 75%-90% of the body surface area. Characterized by widespread erythema and potential systemic symptoms like fever and lymphadenopathy, it severely impairs patient quality of life. The pathogenesis of erythrodermic psoriasis is not fully understood. It is a multifactorial, multistep process suspected to result from an abnormal immune response induced by both genetic and environmental factors. Key contributors to erythrodermic psoriasis onset include specific gene polymorphisms, altered expression of adhesion molecules, dy

The compounds that make up human body odor have been catalogued by researchers in many fields. Yet few are aware of exactly where these molecules come from. Volatile body-odor compounds are often cited as being produced primarily via microbial activity from precursors in sweat. While this is a source of many human volatiles, here we synthesize data showing that some of the most distinctive and abundant components of human odor instead originate from precursors in sebum, via reactions that do not involve the skin microbiome. We also review the unique biochemistry of human sebaceous glands and d…

Lichen planus is a chronic disease affecting the skin, appendages, and mucous membranes. A cutaneous lichen planus is a rare disease occurring in less than 1% of the general population, while oral illness is up to five times more prevalent; still, both forms equally impair the patient's quality of life. The etiology of lichen planus is not entirely understood. Yet, immune-mediated mechanisms have been recognized since environmental factors such as hepatitis virus infection, mechanical trauma, psychological stress, or microbiome changes can trigger the disease in genetically susceptible individ

"Maskne" has been one of the negative spinoffs of the universally acceptable and effective role of face mask in containing the ongoing pandemic of SARS-CoV-2 virus or COVID 19. Several factors contribute to its aetiopathogenesis including heat, humidity, mechanical friction and microbiome dysbiosis in a complex interplay under the occlusive area of the mask. Clinically, the morphology is not very different from acne vulgaris with comedones and inflammatory acne, but in a characteristic distribution over a roughly circular area of face covered by mask. Since the use of face masks is likely to c

Technologies allowing genetic sequencing of the human microbiome are opening new realms to discovery. The host microbiota substantially impacts immune responses both in immune-mediated inflammatory diseases (IMIDs) and in tumors affecting tissues beyond skin and mucosae. However, a mechanistic link between host microbiota and cancer or IMIDs has not been well established. Here, we propose T helper 17 (TH17) lymphocytes as the connecting factor between host microbiota and rheumatoid or psoriatic arthritides, multiple sclerosis, breast or ovarian cancer, and multiple myeloma. We theorize that si

AIM: A novel electrospun biocompatible nanofibrous material loaded with commensal bacteria for potential preventive treatment of the diabetic foot was developed.
MATERIALS & METHODS: Two biocompatible polymers (carboxymethylcellulose and polyethylene oxide) were combined with a bacterium isolate from the skin located between the toes of a healthy adult (identified using a matrix-assisted laser desorption/ionization mass spectrometry-based method as a strain of Staphylococcus epidermidis). Higher bacteria loads in the material were assured through their encapsulation in polyethylenimine. The n

The microbiome of the shoulder demonstrates distinctive differences to other orthopaedic surgical sites. Recent studies have demonstrated that the most common organisms found in deep shoulder infections are coagulase-negative staph lococcal species and Propionibacterium acnes . Many studies support diligent hand washing, decreasing operative time, routine glove changing, minimizing operating room traffic, and covering instruments as means for decreasing the risk of deep infection. On the other hand, hair clipping and the use of adhesive drapes may have little effect on decreasing the incidence

Inflammatory dermatological diseases represent a significant global health burden, with emerging evidence suggesting that modulation of the gut-skin axis microbial interventions may offer therapeutic benefits. However, current evidence is fragmented, with considerable heterogeneity limiting definitive conclusions. A systematic review and a meta-analysis were conducted following PRISMA guidelines, registered in PROSPERO (CRD42024629809). Seven databases were searched for randomized controlled trials evaluating probiotics, synbiotics, or postbiotics in inflammatory skin conditions. Primary outco

UNLABELLED: Vulvar lichen sclerosus (VLS) is a chronic skin condition affecting the vulva, causing significant discomfort, but its etiology in prepubertal girls remains poorly understood. VLS presents with itching, irritation, and pain. Recent studies suggest that cutaneous dysbiosis might play a role in VLS. Our study aims to investigate differences in the vulvar skin microbiota among prepubertal girls with VLS, those with labial adhesions, and healthy controls, and to explore potential microbial links to VLS. We performed a comparative analysis of 16S ribosomal RNA (rRNA) sequences from vulv

We aimed to determine the relationships between intestinal mucosal injury (based on diamine oxidase [DAO] concentrations), physiological modulators of intercellular tight junctions, and leaky gut biomarkers (based on zonulin concentrations) in dogs with atopic dermatitis. We previously demonstrated that gut restoration alleviates canine atopic dermatitis (CaD) based on the interaction between the gut microbiome and dermatological diseases, which is referred to as the "gut-skin axis." However, we observed the reverse in this study. This short-term, open-label, non-repeated study involved 30 own

In traditional Chinese medicine (TCM), aging is viewed as "deficiency" complicated by blood stasis and qi stagnation. "Kang Shuai Lao Pian" (KSLP) is a Ming Dynasty herbal formula widely used to repenish qi, nourish yin, calm the heart, and tranquilize the mind.
AIM OF THE STUDY: We aimed to evaluate the systemic anti-aging activity of KSLP and to determine its mode of action in naturally senescent mice.
MATERIALS AND METHODS: Male 16-18-month-old mice received daily intr

Type 1 diabetes is a condition that can lead to serious long-term complications and can have significant psychological and quality of life implications. Its incidence is increasing in all parts of the world, but the reasons for this are incompletely understood. Genetic factors alone cannot explain such a rapid increase in incidence; therefore, environmental factors must be implicated. Lifestyle factors have been classically associated with type 2 diabetes. However, there are data implicating obesity and insulin resistance to type 1 diabetes as well (accelerator hypothesis). Cholesterol has als
