Quantitative phase imaging (QPI) is an emerging modality, which enables the identification of abnormalities in tissue based on optical properties. QPI can be applied to any biological specimen due to its label-free imaging capability, but its use in stained tissue is unclear. Here, we study the variability of QPI with the staining dye. Several tissues such as brain, heart and lung were stained with hematoxylin and eosin, and their optical properties compared at 550 and 730 nm. We present a new concept, termed tissue lithography (TL), and its implementation which enables retrospective studies on formalin-fixed paraffin-embedded tissue sections. Tissue lithography uses a microfluidic probe to remove microscale areas of the paraffin layer on formalin-fixed paraffin-embedded biopsy samples. To revisit many theories on fetal development of the manubrium of the sternum, we examined 25 mid-term fetuses at 6-9 weeks of gestation. The initial developmental stage of the manubrium was characterized by a distinct interclavicular mesenchyme that was continuous with the developing clavicles. Because parts of the clavicle in which endochondral ossification occurs originate from the neural crest, the interclavicular mesenchyme seems to be of the same origin. The sternal bands, possibly of the lateral plate origin, were restricted at the anterior ends of the ribs in the paired thoracic walls.

We describe the case of a 37-year-old caucasian man with a history of keratoconus and long-term hard contact lens wear. Histological analysis of the corneal button after penetrating keratoplasty revealed staining with Congo red and apple green birefringence detected using polarized light. In the absence of other systemic or ocular pathology, the authors infer this amyloid deposition to be secondary to keratoconus and endeavor to discuss the possible etiopathogenesis of the disease.
