Dopamine receptor functions: lessons from knockout mice [corrected].

In the past few years, a number of laboratories have used gene targeting via homologous recombination to generate mice deficient for key molecules involved in dopaminergic (DAergic) transmission. This tremendous effort has resulted in the successful generation and characterization of mice deficient for the neurotransmitter DA, the main terminator of DAergic neurotransmission (the DA transporter), and all five subtypes of DA receptors. This review summarizes the results from studies of the various DA receptor knockout mice and of mice deficient in proteins that mediate DA receptor signaling. It

Depotentiation in the dentate gyrus of freely moving rats is modulated by D1/D5 dopamine receptors.

Hippocampal depotentiation comprises a reversal of tetanization- induced long-term potentiation (LTP) which occurs following low-frequency s … antagonist SCH 23390. Neither D1/D5 agonist nor antagonist had an effect on LTP expression or basal synaptic transmission. These results highlight differences between D1/D5 receptor-involvement in LTP and depotentiation in the CA1 region and DG, and indicate that whereas D1/D5 receptor activation may not be a critical factor in LTP induction in the DG, a differential r …

Voltage-gated calcium channels in autonomic neuroeffector transmission.

Calcium influx through voltage-gated calcium channels (VGCCs) is required for neurotransmitter release. Recent research has characterised several pharmacologically and electrophysiologically distinct VGCC subtypes, some of which are involved in neurotransmitter release. Transmitter release from autonomic neurons can be coupled to calcium entry through N-, P/Q- and/or R-type VGCCs; the precise combination of VGCC subtypes appears to vary according to the neurotransmitter, tissue and species. L-type channels rarely appear to be important in autonomic neurotransmitter release. There does not appe

Role of Ito cells in the liver function.

Ito cells (perisinusoidal fat-storing cells, stellate cells, lipocytes) of the liver are mesenchymal cells located in the space of Disse. They are the main place of vitamin A storage in characteristic lipid droplets. Moreover, they demonstrate synthetic activity of collagens and other extracellular matrix proteins involved in hepatic fibrosis. Their long, contractile cytoplasmic processes encompass the sinusoids and can affect sinusoidal blood flow. Because of their location in the space of Disse and connection with other sinusoidal and parenchymal cells, they are probably involved in local ne

Neurons of the olfactory organ projecting to the caudal telencephalon and hypothalamus: a carbocyanine-dye labelling study in the brown trout (Teleostei).

The caudal extrabulbar projections and their neurons of origin in the trout were studied after carbocyanine-dye (DiI) labelling in either the olfactory organ or the caudal telencephalon. DiI application to the caudal telencephalon labelled bipolar neurons in the olfactory epithelium, where they were sparsely distributed throughout the olfactory lamellae. Labelled fibres ran scattered in the olfactory nerve without forming bundles. DiI application to the olfactory organ labelled extrabulbar projections to the ventral telencephalon, preoptic region and tuberal hypothalamus. These results confirm

Receptors, photoreception and brain perception. New insights.

Once photons have activated photosensitive cell receptors, a biochemical process mediated by G-proteins transforms the initial signal into nerve potentials. The generated impulses transmit the information through ganglion cells, after a complex interaction with other neurons by means of different neurotransmitters. Since visual function is processed in parallel, ganglion cells are divided into M-neurons which are in charge of capturing large objects, P-neurons capable of analyzing fine details and colors, and non-M, non-P neurons which are sensitive to changes in light intensity. Electrodiagnosis of infantile botulism.

Analysis of the literature on the electrophysiologic features of infantile botulism was undertaken. Small compound muscle action potential amplitude is a very sensitive feature but lacks specificity. The decremental response to 2- to 3-Hz repetitive nerve stimulation is inconsistent and not a reliable sign. Tetanic and posttetanic facilitation are highly sensitive and highly specific. Absence of posttetanic exhaustion is also highly specific for infant botulism and shared only by hypermagnesemia. We conclude that the findings of low compound muscle action potential amplitude in combination wit

Retardation of fast axonal transport in wobbler mice.

To investigate axonal function in a model of early motor neuron disease, we examined fast and slow components of anterograde axonal transport in the less-affected hindlimb motor neurons of wobbler mice. To study the fast component (FC), we injected tritiated amino acids into the lumbar spinal cord and retrieved the sciatic nerve after 2 or 3 h. The transport distance was the extent of the plateau of labeling; regression analysis indicated that FC was 25% slower in wobbler mice than in unaffected littermates (P < 0.01). To study slow component (SC), [35S]methionine was injected. Transport dista

Central auditory metabolic activity induced by intense noise exposure.

Neural activity in the central auditory system was mapped by measuring 2-deoxyglucose (2-DG) uptake during a one hour exposure to a two-octave (1414-5656 Hz) band of noise. Gerbils were exposed to 100, 110 or 120 dB SPL, intensities which can produce only temporary (100 dB) or both temporary and permanent (120 dB) hearing loss. Exposure to 100 dB SPL evoked high levels of neural activity throughout responsive regions of auditory nuclei. At 110 dB SPL, a central region of low neural activity was surrounded by areas exhibiting increased activity. At 120 dB SPL, neural activity was low in almost

Pattern of ongoing activity in rat superior cervical ganglion neurons projecting to a specific target.

The ongoing activity of superior cervical ganglion neurons projecting to the rat submandibular gland was investigated with intracellular techniques. The relevant cells were identified by retrograde labeling with the fluorescent dye DiI placed in the submandibular gland 7-10 days earlier. The majority of labeled cells (89%) were 'silent' during a standard 5-min period of recording. In contrast, only 15% of cells projecting to targets other than the submandibular gland were silent during the standard period of observation. Evidently, ganglion cells innervating different targets have different pa

VEP projections in congenital nystagmus; VEP asymmetry in albinism: a comparison study.

Visual evoked potential (VEP) asymmetry in which a preponderance of nasal and temporal retina afferents project to the contralateral hemisphere after full-field monocular stimulation is considered specific to albinism. Some reports, however, suggest that patients with congenital nystagmus (CN) share the albino-like visual pathway anomaly. To examine the clinical specificity of albino misrouting, VEP topography was assessed in ten patients with congenital nystagmus and in ten age-matched albino patients. As an additional control, the VEP response profiles from eight albino patients with no nyst

Importance of compression onset rate for the degree of impairment of impulse propagation in experimental compression injury of the porcine cauda equina.

The effects of a rapid (0.05-0.1 seconds) and a slow (20 seconds) onset rate of cauda equina compression on impulse propagation, in terms of tail muscle electromyogram (EMG) amplitude, were analyzed in an experimental model of the pig cauda equina. Sham compression and compression at 50 mm Hg at either rapid or slow onset induced no or only minor functional impairment. Compression at 100 or 200 mm Hg induced impairment of the impulse propagation that was proportional to the applied pressure. The effects were more pronounced (P less than 0.01) for the rapid than for the slow onset of compressio

Modulation of peripheral sympathetic nerve transmission.

The past 15 years have been witness to a remarkable growth in knowledge regarding the modulation of "sympathetic traffic" to neuroeffector organs, including vascular tissue. The release of norepinephrine from peripheral sympathetic neurons is now known to be under both negative and positive feedback control. Norepinephrine, when released from peripheral neurons, acts on presynaptic alpha 2-receptors to inhibit further neurotransmission. Vascular postsynaptic alpha 2-receptors, sensitive to circulating catecholamines, subserve vasoconstriction. The antihypertensive agents clonidine, guanabenz a

Bingeing behavior and plasma amino acids: a possible involvement of brain serotonin in bulimia nervosa.

It has been suggested that bingeing and vomiting behavior may be an attempt to suppress hunger or reduce dysphoria. Theoretically, such relationships could involve a mechanism whereby bingeing and vomiting change plasma amino acids which, in turn, enhance brain serotonin-mediated satiety and/or improvement in mood. This hypothesis is based on data showing that the intake of dietary carbohydrates increases the uptake of tryptophan (TRP), the precursor of serotonin, into the brain by increasing the plasma TRP ratio (the ratio of the plasma TRP concentration to the summed concentrations of other

Full-wave rectification from a mixed electrical-chemical synapse.

Electrical and chemical synapses usually reinforce one another, but the pyloric late-to-lateral pyloric (PL-to-LP) neuronal connections in lobster stomatogastric ganglia create an inverted U-shaped transfer function between the two neurons: regardless of whether the PL membrane voltage swings positive or negative, the postsynaptic LP voltage will go negative. When the presynaptic cell voltage goes negative, the effect on the LP voltage is due to electrical coupling. During positive presynaptic voltages, the strong contribution of graded chemical inhibition from the PL to the LP neuron override

[Unilateral sensory neuropathy of the trigeminal nerve as the leading symptom of primary Sjögren syndrome ("sicca syndrome")].

We report a case who presenting with a progressive numbness and mild hyperpathia in the second cutaneous division of the left trigeminal nerve as the leading symptom of Primary Sjögren's Syndrome. Further typical features of this autoimmune disorder are keratoconjunctivitis sicca and xerostomia. A symmetrical, predominantly sensory polyneuropathy can be revealed by sensory nerve conduction studies of the median and sural nerves. Laboratory findings mostly include elevated erythrocyte sedimentation rate, hypergammaglobulinemia and hypercomplementemia. The presence of the precipitating antinucle
