Integrated 2D-3D Proteomic Profiling Identifies MLK4 as a Microenvironment-Responsive Regulator of Chemotherapeutic Resistance in Human Glioblastoma Cells.

BACKGROUND: Therapeutic resistance is a major cause of treatment failure in glioblastoma (GBM), highlighting the need for physiologically re … ive RT-PCR, and functional studies were conducted using genetic depletion followed by assessment of temozolomide (TMZ) sensitivity. RESULTS: Comparative analysis identified 13 proteins consistently differentially expressed between 2D and 3D cultures: NDUFB5, RNGTT, MLK4, SYN1, DDX5, EIF2AK2, ITGA1, ZNF33B, ZNF343, WDR19, JPH3, CCT8L2, and FNDC3A. The cytoskeletal regulator Coronin-1A plays a multidirectional role in glioblastoma stemness.

Glioblastoma (GBM) recurrence is driven by therapy-resistant cells that escape surgical detection and withstand subsequent chemoradiotherapy … d intraoperative fluorescence detectability. Furthermore, Coro1A depletion increased sensitivity to temozolomide (TMZ) and X-ray irradiation, revealing its critical contribution to therapy resistance. Collectively, these findings establish Coro1A as a central cytoskeletal regulator that unifies invasion, diagnostic escape, and therapeutic resilience-highlighti …

Title: Role of O6-methylguanine DNA methyltransferase status as predictive factor for alkylatin-based chemotherapy in advanced neuroendocrine tumors: State of the art.

BACKGROUND: Temozolomide (TMZ) and other alkylating agents (ALK) yield high response rates when used in metastatic neuroendocrine tumors (NETs). Several series reported higher response rates when used in tumors with low expression of O6-Methylguanine DNA Methyltransferase (MGMT) with immunohistochemistry or promoter methylation. However, evidence is most consistent in pancreatic neuroendocrine tumors. Variations in MGMT assessment further complicate direct comparisons and clinical application. Thus, the predictive role of MGMT in patients with NETs treated with ALK is still debated. METHODS: …

The Toxic Effect and Mechanism of TMZ Combined with siHOXB9 on Glioblastoma Cells.

Glioblastoma (GBM) represents a highly invasive primary malignant tumor within the central nervous system (CNS). Temozolomide (TMZ), a first-line chemotherapy agent for GBM treatment, has significant limitations, including drug resistance, poor water solubility, a short half-life, and notable toxic side effects. The innovation of the TMZ dosage form is pivotal for enhancing its therapeutic efficacy. In this study, solid lipid nanoparticles (SLN) loaded with Angiopep-2 (A2) and TMZ (TMZ-A2SLN), a nanopolymer featuring a solid spherical morphology and a particle size of approximately 100 nm, wer…

Macrophage membrane-camouflaged pure-drug nanomedicine for synergistic chemo- and interstitial photodynamic therapy against glioblastoma.

Glioblastoma (GBM) persists as a highly fatal malignancy, with current clinical treatments showing minimal progress over years. Interstitial photodynamic therapy (iPDT) holds promise due to its minimally invasive nature and low toxicity but is impeded by poor photosensitizer penetration and inadequate GBM targeting. Here, we developed a biomimetic pure-drug nanomedicine (MM@CT), which co-assembles the photosensitizer chlorin e6 (Ce6) and the first-line chemotherapeutic drug (temozolomide, TMZ) for GBM, then camouflaged with macrophage membranes. This design eliminates the need for traditional …

Low YTHDC1 Expression Upregulates FSCN1 to Promote Nuclear F-Actin Formation and Facilitate Double-strand DNA Breaks Repair in TMZ-Resistant Glioblastoma.

Glioblastoma (GBM) is an aggressive and recurrent malignancy with a poor prognosis. Although temozolomide (TMZ) is a cornerstone of GBM treatment, its efficacy is often compromised by inherent or acquired resistance, underscoring the urgent need to uncover molecular mechanisms, discover new therapeutic targets, and develop innovative treatment strategies. In this study, we found an increased formation of filamentous actin (F-actin) within the nuclei of TMZ-resistant GBM cells. We also showed that overexpression of FSCN1 in TMZ-resistant GBM cells promotes F-actin formation and facilitates the …

[Immunotherapy in the treatment of aggressive pituitary tumors and carcinomas].

Since the therapeutic failure of temozolomide (TMZ), the first-line chemotherapy recommended against aggressive pituitary tumors (APTs) and pituitary carcinomas (PCs), there have been no solid evidence-based treatment recommendations. However, immune checkpoint inhibitors (ICIs) have emerged as a promising and recent therapeutic option, generating hope after the first successful case was published in 2018. APTs and PCs are rare tumors with poor prognosis due to their invasive behavior and high recurrence rate despite conventional treatments. These neoplasms pose a therapeutic challenge in endo…

Valproic Acid Reduces Invasiveness and Cellular Growth in 2D and 3D Glioblastoma Cell Lines.

Glioblastoma (GBM) is the most common malignant brain tumor, with a poor prognosis and low survival. Its treatment includes complete surgical resection followed by radiotherapy combined with temozolomide (TMZ). GBM contains glial stem cells (GSCs), which contribute to tumor progression, invasiveness, and drug resistance. The histone deacetylase (HDAC) inhibitor valproic acid (VA) has been shown to be a potent antitumor and cytostatic agent. In this study, we tested the effects of VA on glioma cell proliferation, migration, and apoptosis using T98G monolayer and spheroid cells. T98G and U-87MG …
