A longitudinal investigation of an outbreak of toe tip necrosis syndrome in western Canadian feedlot cattle.

This investigation followed a cohort (n = 21) of single-sourced Angus cross yearling heifers diagnosed with toe tip necrosis syndrome (TTNS). The animals were intensively followed for ~7 months, over which time 5 animals were euthanized because of intractable lameness or life-threatening sequelae such as cellulitis and myositis. Treatment consisted of systemic antimicrobial and nonsteroidal anti-inflammatory therapy, removing the necrotic horn tissue to facilitate drainage, and relocating the cattle to an earthen-floor pen. Sixteen of the 21 heifers recovered uneventfully. Upon sectioning the

Serological survey for antibody to Encephalitozoon cuniculi in horses in the USA.

Encephalitozoon cuniculi is an obligate intracellular microsporidian parasite that can result in clinical and subclinical infection in many species. In the present study, a serological survey was conducted using samples from 105 horses in the state of New Jersey; 49 of the samples were obtained from clinically abnormal animals. Five or 4.8% of 105 serum samples were found to demonstrate reactivity by ELISA with titers of 1:64 to 1:1,024. One of the samples was obtained from a clinically normal horse. Clinical signs and diagnoses from the other animals included lameness, colic, osteochondritis

Neuroendocrine factors influencing polydipsia in psychiatric patients: an hypothesis.

Polydipsia and water intoxication cause considerable morbidity and mortality in chronic psychiatric patients. The pathophysiology of the disorder is unknown, and there is no effective treatment. Angiotensin II is an important dipsogen in animals; in humans, some conditions with abnormal thirst are associated with increased angiotensin function. Chronic D2 dopamine receptor blockade increases angiotensin II-induced thirst in animals; in humans, increased peripheral response to angiotensin II is documented. Chronic D2 blockade with typical neuroleptics may increase sensitivity to angiotensin II

Adenoviral Gene Transfer of Gremlin Modulates Vascular Endothelial Growth Factor-A-Induced Angiogenesis in Porcine Myocardium.

Coronary artery disease is a major cause of death and disability worldwide. New therapies are needed for patients who do not benefit or are not suitable for current treatments. Angiogenic gene therapy using vascular endothelial growth factors (VEGFs) has shown potential in preclinical trials. However, undesired side effects, such as increased permeability, limit their therapeutic potential. The aim of this study was to investigate if adenoviral gene transfer of a VEGF receptor 2 (VEGFR-2) ligand Gremlin, given simultaneously with VEGF-A, could modulate VEGFR-2-mediated increase in permeability

Intravenous glutamine administration reduces lung and distal organ injury in malnourished rats with sepsis.

Malnutrition is a risk factor for infection, compromising immune response. Glutamine (Gln) protects the lungs and distal organs in well-nourished septic and nonseptic conditions; however, no study to date has analyzed the effects of Gln in the presence of sepsis and malnutrition. In the present work, we tested the hypothesis that early therapy with intravenous Gln prevents lung and distal organ damage in septic malnourished rats. Protein-energy malnutrition was induced in male Wistar rats for 4 weeks. At the end of 4 weeks, malnourished animals were assigned to a sepsis-inducing cecal ligation

Treatment with MG53 ameliorates traumatic brain injury-associated acute kidney injury.

INTRODUCTION: Multiorgan dysfunction (MOD) after traumatic brain injury (TBI) results in increased morbidity and mortality. There is emerging evidence demonstrating TBI-induced inflammatory responses; however, the mechanisms driving TBI-induced organ injury remains unknown and understudied. MG53, a cell membrane repair protein, has been shown to reduce brain lesion size following TBI. In this study, we aimed to establish a large animal model of post-TBI MOD, determine MG53's role in renal protection following TBI, and explore a mechanistic link between endothelial cell dysfunction and post-TBI

Reduction of Alcohol-Dependent Lung Pathological Features in Rats Treated with Fenofibrate.

Alcohol use disorder (AUD) is a public health problem characterized by a marked increment in systemic inflammation. In the last few years, it has been described as the role of alcohol in neuroinflammation affecting some aspects of neuronal function. Interestingly, inflammation is reduced with fenofibrate treatment, a PPARα agonist used to treat dyslipidemia. On the other hand, alcohol has been associated with chronic inflammation and fibrosis in the lungs, affecting their normal function and increasing respiratory infections. However, a deep characterization of the role of alcohol in the worse

Isolated lower extremity chemotherapeutic infusion for treatment of osteosarcoma: experimental study and preliminary clinical report.

PURPOSE: To increase the dose of drug delivered to a tumor while maintaining tolerable systemic side effects, an interventional technique of isolated lower extremity infusion was investigated. MATERIALS AND METHODS: Experiments were performed in eight dogs. Four dogs were treated by a combination of intraarterial (IA) femoral cisplatin infusion at a dose of 2.0 mg/kg with drug removal from the ipsilateral extremity venous blood by a dialyzer. The other four dogs comprised the control group. In these animals, left femoral arterial IA cisplatin infusion was performed without dialysis. Leukocyte

Veterinary use of bacteriophage therapy in intensively-reared livestock.

Zoonoses are infectious diseases transmitted directly or indirectly between animals and humans. Several important zoonotic pathogens colonize farm animals asymptomatically, which may lead to contamination of the food chain and public health hazards. Moreover, routine sampling of carcasses at retail by government authorities over the past 20 years suggests the prevalence of antibiotic resistance in foodborne pathogens has increased. If this continues, antibiotics may be ineffective against such pathogens in the future and alternative approaches, such as phage therapy, may be necessary. Intensiv

Proteasome inhibitor PS-341, a potential therapeutic agent for adult T-cell leukemia.

Nuclear factor kappaB (NF-kappaB) plays a major role in the pathogenesis of human T-cell lymphotrophic virus I-associated malignancy. Proteasome inhibitors provide a rational approach to control constitutively activated NF-kappaB in human T-cell lymphotrophic virus I-infected T cells. We report that the proteasome inhibitor PS-341 decreased NF-kappaB DNA binding activity by preventing degradation of IkappaB(alpha). In our murine model of adult T-cell leukemia, PS-341 used alone did not yield prolongation of the survival of tumor-bearing mice. However, when combined with the current clinically

Toxoplasmosis in Atlantic bottle-nosed dolphins (Tursiops truncatus).

A female Atlantic bottle-nosed dolphin (Tursiops truncatus) and her calf were found beached on Picnic Island in Tampa Bay, Florida, USA. Despite therapy the animals died. Necropsy revealed severe pneumonia and lymphadenopathy in the mother and the calf, gastric ulcers and infection with the stomach digenean Braunina cordiformis in the mother, and a large, pale liver in the calf. Toxoplasma gondii was identified by light and electron microscopy and by immunohistochemistry in tissues of both animals. Toxoplasma gondii was associated with interstitial pneumonia, necrotizing adrenalitis, and cardi

Effect of non-steroidal anti-inflammatory drugs on fertility of male rats.

In an attempt to study the influence of seminal prostaglandin reduction on male fertility, the effect of prolonged treatment with 4 non-steroidal anti-inflammatory drugs (acetylsalicylic acid, indomethacin, naproxen and phenylbutazone) on fertility was determined in male rats. Before the fertility experiments, the pharmacokinetics of the drugs were determined to find dosage regimens by which drug concentrations known as active from human anti-inflammatory therapy could be reached and maintained in the animals. Except for phenylbutazone, all drugs decreased prostaglandin E-2 level in seminal fl

[Modification of antibody formation in animal experiments by glucocorticoids and aristolochic acid].

In the bone marrow of guinea pigs a decrease of lymphocytes resulted after immunisation against Q-fever antigen, compensating within 4 weeks. Under treatment with prednisolone normalisation occurred within 3, with aristolochic acid as well as with prednisolone and aristolochic acid already up to the 2nd week. After immunisation plasma cell increase was lasting longer in the aristolochic acid group than in other groups. The antibody determinations resulted in different values. After immunisation with Q-fever antigen the antibody levels in rabbits increased without significant differences betwee

Gentamicin gradient patterns and morphological changes in human kidneys.

The gentamicin gradient pattern was studied in ten autopsied patients who expired during gentamicin therapy. Although two of the patients had acute renal failure, none was thought to have gentamicin-related nephrotoxicity. Gentamicin concentrations in cortex significantly exceeded those in medulla or serum (p less than 0.05 and p less than 0.01, respectively). No correlation was found between the cortical concentrations and the total drug dose, the time interval between the last injection and death, or the plasma creatinine concentrations in this heterogeneous group of patients. In addition, t

Progress in the understanding and treatment of Fabry disease.

BACKGROUND: Fabry disease is caused by α-galactosidase A deficiency. Substrates of this lysosomal enzyme accumulate, resulting in cellular dysfunction. Patients experience neuropathic pain, kidney failure, heart disease, and strokes. SCOPE OF REVIEW: The clinical picture and molecular features of Fabry disease are described, along with updates on disease mechanisms, animal models, and therapies. MAJOR CONCLUSIONS: How the accumulation of α-galactosidase A substrates, mainly glycosphingolipids, leads to organ damage is incompletely understood. Enzyme replacement and chaperone therapies are cl

Animal models of idiopathic membranous nephropathy: Recent advances and future perspectives.

Idiopathic membranous nephropathy (IMN) is one of the main causes of adult nephrotic syndrome. A subset of untreated or inadequately treated patients eventually progress to end-stage renal disease (ESRD), posing a significant clinical challenge. Although the discovery of novel podocyte target antigens has deepened our understanding of IMN pathogenesis, the precise molecular mechanisms remain incompletely elucidated, and effective targeted therapies are still lacking. Animal models play an irreplaceable role in uncovering IMN pathogenesis and developing effective therapies. In recent years, wit

Pharmacological Treatment of Degenerative Cervical Myelopathy: A Critical Review of Current Evidence.

Degenerative cervical myelopathy (DCM) is the leading cause of spinal cord dysfunction in adults, representing substantial morbidity and significant financial and resource burdens. Typically, patients with progressive DCM will eventually receive surgical treatment. Nonetheless, despite advancements in pharmacotherapeutics, evidence for pharmacological therapy remains limited. Health professionals from various fields would find interest in pharmacological agents that could benefit patients with mild DCM or enhance surgical outcomes. This review aims to consolidate all clinical and experimental

The antidepressant fluoxetine protects the hippocampus from brain damage in experimental pneumococcal meningitis.

BACKGROUND: High mortality and morbidity rates are observed in patients with bacterial meningitis (BM) and urge for new adjuvant treatments in addition to standard antibiotic therapies. In BM the hippocampal dentate gyrus is injured by apoptosis while in cortical areas ischemic necrosis occurs. Experimental therapies aimed at reducing the inflammatory response and brain damage have successfully been evaluated in animal models of BM. Fluoxetine (FLX) is an anti-depressant of the selective serotonin reuptake inhibitors (SSRI) and was previously shown to be neuroprotective in vitro and in vivo. W

The Role of Animal Research in Pandemic Responses.

The significant advances made by the global scientific community during the COVID-19 pandemic, exemplified by the development of multiple SARS-CoV-2 vaccines in less than 1 y, were made possible in part because of animal research. Historically, animals have been used to study the characterization, treatment, and prevention of most of the major infectious disease outbreaks that humans have faced. From the advent of modern 'germ theory' prior to the 1918 Spanish Flu pandemic through the more recent Ebola and Zika virus outbreaks, research that uses animals has revealed or supported key discoveri

A New Era for Rare Genetic Diseases: Messenger RNA Therapy.

Exogenous delivery of messenger RNA (mRNA) is emerging as a new class of medicine with broad applicability including the potential to treat rare monogenic disorders. Recent advances in mRNA technology, including modifications to the mRNA itself along with improvements to the delivery vehicle, have transformed the utility of mRNA as a potential therapy to restore or replace different types of therapeutic proteins. Preclinical proof-of-concept has been demonstrated for mRNA therapy for three different rare metabolic disorders: methylmalonic acidemia, acute intermittent porphyria, and Fabry disea
