Multiantibiotic residues in commercial fish from Argentina. The presence of mixtures of antibiotics in edible fish, a challenge to health risk assessment.

The occurrence of 46 antibiotics (amphenicols, cephalosporins, dihydrofolate reductase inhibitors, fluroquinolones, macrolides, nitrofurans, penicillins, quinolones, sulfamides and tetracyclines) in Argentinean market fish were investigated by UPLC-MS/MS. Veterinary and human antimicrobials enrofloxacin, clarithromycin, roxithromycin, doxycycline and oxytetracycline were detected in 100% of the samples, being to our knowledge the first report of clarithromycin in edible fish muscle. Maximum Residual Limits were exceeded for at least one antibiotic in 82% of pacú, 57% of shad, 57% of trout and …

Quinolones and tetracyclines in aquaculture fish by a simple and rapid LC-MS/MS method.

The determination of antimicrobials in aquaculture fish is important to ensure food safety. Therefore, simple and fast multiresidue methods … nd the coefficients of determination were ≥0.98. The method was applied to Nile tilapia and rainbow trout (n = 29) and 14% of them contained enrofloxacin at levels above the limit of quantification (12.53-19.01 µg.kg-1) but below the maximum residue limit (100 µg.kg-1). Even though prohibited in Brazil and other countries, this antimicrobial reached fis …

The environmental consequences of alien species in the Swedish lakes Mälaren, Hjälmaren, Vänern and Vättern.

Twenty alien species have become established in the lakes Mälaren, Vänern, Vättern and Hjälmaren. Intentional introductions include fish and … fects and has contributed to the decimation of the indigenous noble crayfish. The stocking of brown trout and salmon with origins from different biogeographical regions has contributed to the extinction of relict indigenous fish species in L. Vänern. Although major ecosystem damage caused by the introduction of alien species, with the exception of the c …

Neurotoxic effect on two fish species and a PC12 cell line of the supernate of Vibrio alginolyticus and Vibrio anguillarum.

The biological effects of supernates obtained from different strains of Vibrio alginolyticus and Vibrio anguillarum isolated from diseased fish have been studied by inoculation on two fish species, eel and rainbow trout, and two fish cell lines. These supernates possess neuroexcitatory properties, and so, when they are injected into both fish species, they trigger convulsions, wriggling, contortive swimming and respiratory arrest coupled with increased respiratory reflex. Furthermore, after the application of the supernates on cultures of noradrenergic pheochromocytoma PC12 cells, an increase …

A model of salmon louse production in Norway: effects of increasing salmon production and public management measures.

Salmon lice Lepeophtheirus salmonis Krøyer have caused disease problems in farmed Atlantic salmon Salmo salar L. since the mid-1970s in Norway. High infection intensities and premature return of wild sea trout Salmo trutta L. were first reported in 1992. Later emaciated wild Atlantic salmon smolts carrying large amounts of lice have been observed both in fjords and offshore. The Norwegian Animal Health Authority regulations to control the problem, which came into operation in 1998, included compulsory louse level monitoring in farms and maximum legal numbers of lice per fish. Here, we present …

Rapid detection of infectious pancreatic necrosis virus (IPNV) by the enzyme-linked immunosorbent assay (ELISA).

The enzyme-linked immunosorbent assay (ELISA) was used to demonstrate the presence of infectious pancreatic necrosis virus (IPNV) antigen in cell cultures and in fish. Virus antigen could be detected in infected cell cultures before visible cytopathic effect (c.p.e.) was evident and cell cultures showing complete viral c.p.e. produced intense colour reactions. Virus antigen was detected in infected fry during and immediately after an epizootic, but although IPNV-carrier fish could be detected by the ELISA technique, the sensitivity of detection was not as great as that of isolation of the infe

Chaperone-mediated autophagy protects against hyperglycemic stress.

Chaperone-mediated autophagy (CMA) is a major pathway of lysosomal proteolysis critical for cellular homeostasis and metabolism, and whose defects have been associated with several human pathologies. While CMA has been well described in mammals, functional evidence has only recently been documented in fish, opening up new perspectives to tackle this function under a novel angle. Now we propose to explore CMA functions in the rainbow trout (RT, Oncorhynchus mykiss), a fish species recognized as a model organism of glucose intolerance and characterized by the presence of two paralogs of the CMA-…

Intracellular staining of physiologically identified photoreceptor cells and hyperpolarizing interneurons in the teleost pineal organ.

The directly photosensory pineal organ of the rainbow trout functions primarily as a luminance detector. Its neutral output reflects the level of ambient illumination in an almost linear fashion over several orders of magnitude. It may thus transmit information about the daily light-dark cycle to central projection targets in the brain, and exert an important control over putative central oscillators. We have studied single neural elements in the explanted pineal organ of the rainbow trout by combining intracellular recording with intracellular injections of either the fluorescent dye Lucifer …

Influences on bioaccumulation and biota-sediment accumulation factors for methyl-mercury in High Arctic lake food webs.

Mercury (Hg) concentrations in the fillets of Arctic char (Salvelinus alpinus) differ widely among neighboring oligotrophic lakes on Cornwallis Island (Nunavut, Canada), but the reasons for these differences are poorly understood. Climate change is likely affecting Hg cycling in lakes, although studies report conflicting or mixed responses. We quantified Hg bioaccumulation (BAFs), biota-sediment accumulation (BSAFs), and trophic transfer factors (TTFs) for chironomids and Arctic char in Char, Small, Resolute, and Meretta Lakes to explain lake-to-lake differences. Simultaneously, we leveraged t

Mercury and methylmercury concentrations in high altitude lakes and fish (Arctic charr) from the French Alps related to watershed characteristics.

Total mercury (THg) and methylmercury (MeHg) concentrations were measured in the muscle of Arctic charr (Salvelinus alpinus) and in the water column of 4 lakes that are located in the French Alps. Watershed characteristics were determined (6 coverage classes) for each lake in order to evaluate the influence of watershed composition on mercury and methylmercury concentrations in fish muscle and in the water column. THg and MeHg concentrations in surface water were relatively low and similar among lakes and watershed characteristics play a major role in determining water column Hg and MeHg level

Diet affects the cardiorespiratory response to hypoxia and autonomic control of the heart in brook char (Salvelinus fontinalis).

Taurine is the most abundant free amino acid in heart and is important in tissue volume regulation. β-alanine, a common dietary amino acid, decreases cardiac taurine flux in fish and negatively impacts their responses to stress. Brook char (Salvelinus fontinalis) fed a β-alanine enriched diet to induce taurine deficiency (TD) have lower heart rates and less pronounced decreases in heart rate (bradycardia) under hypoxia, but impacts on blood flow in vivo have not been examined. We aimed to determine if changes in taurine flux constrain cardiac output under hypoxia and to characterize the role o

Solid-phase microextraction coupled to LC-ESI-MS/MS: evaluation and correction for matrix-induced ionization suppression/enhancement for pharmaceutical analysis in biological and environmental samples.

Solid-phase microextraction (SPME) coupled to liquid chromatography with electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) has been widely used to analyze biological fluids, tissues, and environmental matrixes for a variety of organic compounds including pharmaceuticals. However, effects of the sample matrix coextracted by SPME on tandem mass spectrometry analysis have not been systematically investigated. In this study, we characterized the complexity of matrix effects (ME) by analyzing SPME extracts of fish muscle and brain tissue, blood, and bile, as well as tap water, surface

Molecular tracing confirms that infection with infectious pancreatic necrosis virus follows the smolt from hatchery to grow-out farm.

Infectious pancreatic necrosis (IPN) is an important restraint to production of salmonids in aquaculture globally. In order to implement efficacious mitigation strategies for control of this disease, it is important to understand infection routes under current production systems. IPN virus has been shown to be transmitted vertically in Rainbow trout, from broodstock to fingerlings in hatcheries, and there is circumstantial evidence suggesting that vertical transmission can also occur in Atlantic salmon, in addition to horizontal transmission between grow-out fish in farms. In this study, we sh…

[Peter-the-Great Gulf fishes are a source of human invasion of the nematode Anisakis simplex (the family Anisakidae)].

The paper presents the results of the rate of infection with the nematode Anisakis simplex in Peter-the-Great gulf fishes that are a source of human invasion. Five of 19 fish types were found to be nematode-free, and the extensiveness and intensity of invasion were observed in Siberian salmons (Oncorhynchus keta) (61% and 2-700 worms, respectively), Alaska pollacks (Theragra chalcogramma) (75.7% and 1-31 worms), and brown rock trouts (Hexagrammos octogrammus) (100% and 1-60 worms). Autopsy showed that the distribution of larvae was ambiguous in different fish species. Data on the survival of A…
