IDH1-mutant vaccine in newly diagnosed astrocytoma: final analysis of the multicenter, single-arm, open-label, first-in-human phase 1 NOA16 trial.

The clonal glioma driver mutation IDH1R132H gives rise to a major histocompatibility class II-restricted neoepitope. A multicenter, first-in-human phase 1 trial met its prespecified primary endpoints by demonstrating safety and immunogenicity of an IDH1-R132H peptide vaccine (IDH1-vac) integrated into standard of care in 33 participants with newly diagnosed grade III and IV (World Health Organization classification 2007) IDH1-R132H+ astrocytomas (NOA16). Here we report on the clinical and immunological long-term follow-up of this trial as secondary and translational endpoints. The 8-year progr

Advancing poultry health: A meta-analysis of epitope-based and peptide-based vaccines against Avian Pathogenic E. coli with machine learning insights.

INTRODUCTION: Avian Pathogenic Escherichia coli (APEC) causes colibacillosis in poultry, which leads to tremendous economic losses. Traditional control methods, including antibiotics and conventional vaccines, are less effective due to the genetic diversity of APEC and developing antimicrobial resistance (AMR). Novel epitope- and peptide-based vaccines, supported by machine learning (ML), hold high promise. OBJECTIVES: This meta-analysis and systematic review evaluated the effectiveness of epitope- and peptide-vaccine-based candidates against APEC-related morbidity and mortality, production f

Genomic insights and vaccine evaluation of a virulent MLST ST234 Bacillus cereus infecting Asian sea bass (Lates calcarifer).

An outbreak of disease in farmed Asian sea bass (Lates calcarifer) was investigated. A Bacillus cereus group strain, SB01, was consistently isolated from internal organs and identified by groEL PCR. Transmission electron microscopy (TEM) revealed rod-shaped cells with prominent flagella. Experimental infection demonstrated dose-dependent mortality with an LD50 of 5.6 × 106 CFU/fish and pathological signs including ascites, splenomegaly, and multi-organ necrosis. Whole-genome sequencing revealed a 5.26 Mb chromosome and a 240 kb plasmid (pSB01) encoding anthrax toxin-related genes, including pr

Identification and protective efficacy characterization of novel immunogenic antigens for Salmonella Enteritidis vaccines.

The global poultry industry incurs substantial economic losses due to Salmonella enterica serovar Enteritidis (S. Enteritidis), underscoring the urgent need for cross-protective vaccines capable of addressing evolving pathogen diversity. Using an integrative vaccinology approach, we systematically screened immunoreactive proteins to identify novel subunit vaccine candidates. Eight candidate proteins, including type III secretion system components (SipA, SopE2, SipB, SipC), membrane-associated proteins (TolA, TolC, LamB), and the virulence factor MipA, demonstrated pronounced antigenic and immu

Alphaherpesvirus Vaccines.

Prophylactic and therapeutic vaccines for the alphaherpesviruses including varicella zoster virus (VZV) and herpes simplex virus types 1 and 2 have been the focus of enormous preclinical and clinical research. A live viral vaccine for prevention of chickenpox and a subunit therapeutic vaccine to prevent zoster are highly successful. In contrast, progress towards the development of effective prophylactic or therapeutic vaccines against HSV-1 and HSV-2 has met with limited success. This review provides an overview of the successes and failures, the different types of immune responses elicited by

DENGUE VACCINES.

The uniqueness of the dengue viruses (DENVs) and the spectrum of disease resulting from infection have made dengue vaccine development difficult. Several vaccine candidates are currently being evaluated in clinical studies. The candidate currently at the most advanced clinical development stage, a live-attenuated tetravalent vaccine based on the chimeric yellow fever-dengue virus (CYD-TDV), has progressed to Phase 3 efficacy studies. Several other live-attenuated vaccines, as well as subunit, DNA, and purified inactivated vaccine candidates are at earlier stages of clinical development. Additi

A novel TB vaccine; towards a strategy based on our understanding of BCG failure.

The protection afforded by the currently available tuberculosis vaccine, bacillus Calmette-Guérin (BCG) is insufficient and new vaccine strategies are urgently needed. Progress in our understanding of the immunological deficits of BCG combined with novel knowledge on genetics of mycobacteria has paved the way for promising new vaccine strategies. These include recombinant modified BCG vaccines, attenuated strains of Mycobacterium tuberculosis, and various non-live candidates such as DNA and subunit vaccines. Decisive for transforming technical progress into a novel tuberculosis (TB) vaccine st

Two decades of plant-based candidate vaccines: a review of the chimeric protein approaches.

Genetic engineering revolutionized the concept of traditional vaccines since subunit vaccines became reality. Additionally, over the past two decades plant-derived antigens have been studied as potential vaccines with several advantages, including low cost and convenient administration. More specifically, genetic fusions allowed the expression of fusion proteins carrying two or more components with the aim to elicit immune responses against different targets, including antigens from distinct pathogens or strains. This review aims to provide an update in the field of the production of plant-bas

Expression of a synthetic pertussis toxin operon in Escherichia coli.

Bordetella pertussis is the causative agent of whooping cough, a severe disease of infants characterised by repeated of paroxysmal coughing. Pertussis toxin (PT) is a major virulence factor of B. pertussis and is a typical A/B bacterial toxin consisting of five subunits S1-S5 in a ratio of 1:1:1:2:1. The PT subunit genes are organized into an operon which is not expressed in Escherichia coli, thus hampering the use of this organism for vaccine production. We have expressed the five PT subunits individually in E. coli by replacing the wild-type transcriptional and translational signals, and in

A BcfA-containing intranasal vaccine generated TH17 immunity and reduced B. bronchiseptica colonization and disease in mice.

Bordetella bronchiseptica (Bb) is a significant etiologic agent of respiratory disease in farm and companion animals. Current vaccines provide short-lived protection against disease, but do not elicit mucosal immunity that is critical for sustained protection. Here we tested a trivalent protein subunit vaccine composed of Bordetella antigens filamentous hemagglutinin (FHA) pertactin (Prn), and the dual antigen-adjuvant Bordetella colonization factor A (BcfA). Intranasal immunization of mice generated TH17 polarized systemic and tissue-resident CD4 + T cells and systemic and mucosal IgG and IgA

Vaccine-induced antibodies target sequestered viral antigens to prevent ocular HSV-1 pathogenesis, preserve vision, and preempt productive neuronal infection.

The cornea is essential for vision yet highly sensitive to immune-mediated damage following infection. Generating vaccines that provide sterile immunity against ocular surface pathogens without evoking vision loss is therefore clinically challenging. Here, we tested a prophylactic live-attenuated vaccine against herpes simplex virus type 1 (HSV-1), a widespread human pathogen that can cause corneal blindness. Parenteral vaccination of mice resulted in sterile immunity to subsequent HSV-1 challenge in the cornea and suppressed productive infection of the nervous system. This protection was unma

Mucosal delivery switches the response to an adjuvanted tuberculosis vaccine from systemic TH1 to tissue-resident TH17 responses without impacting the protective efficacy.

Pulmonary tuberculosis (TB) remains one of the leading causes of infectious disease death despite widespread usage of the BCG vaccine. A number of new TB vaccines have moved into clinical evaluation to replace or boost the BCG vaccine including ID93+GLA-SE, an adjuvanted subunit vaccine. The vast majority of new TB vaccines in trials are delivered parenterally even though intranasal delivery can augment lung-resident immunity and protective efficacy in small animal models. Parenteral immunization with the adjuvanted subunit vaccine ID93+GLA-SE elicits robust TH1 immunity and protection against

Heterogeneity in the A33 protein impacts the cross-protective efficacy of a candidate smallpox DNA vaccine.

We previously developed a gene-based vaccine, termed 4pox, which targets four orthopoxvirus proteins (A33, L1, B5, and A27). Because any subunit orthopoxvirus vaccine must protect against multiple species of orthopoxviruses, we are interested in understanding the cross-protective potential of our 4pox vaccine target immunogens. In our current studies, we focused on the A33 immunogen. We found one monoclonal antibody against A33, MAb-1G10, which could not bind the monkeypox virus A33 ortholog, A35. MAb-1G10 binding could be rescued if A35 amino acids 118 and 120 were substituted with those from

Homologous prime boosting based on intranasal delivery of non-pathogenic invasive Escherichia coli expressing MPT64, decreases Mycobacterium tuberculosis dissemination.

Protein-subunit vaccines as boosting strategies against tuberculosis (TB) infection are currently in the pipeline of TB vaccine research. Their main limitation is represented by their poor immunogenicity, which makes it necessary to couple protein-subunits with adjuvant molecules. In this study, we employed replication-deficient invasive Escherichia coli strains to deliver Mycobacterium tuberculosis proteins to the cytoplasm of non-phagocytic eukaryotic cells using various priming and prime-boosting vaccination protocols. Our results demonstrate that intranasal administration of invasive E. co

Heterologously type IV pilus expressed protein of Burkholderia pseudomallei is immunogenic but fails to induce protective immunity in mice.

Burkholderia pseudomallei is the causative agent of melioidosis, a severe disease of humans and animals. At present, no effective vaccine against melioidosis exists. Bacterial type IV pilin proteins have been used successfully as subunit vaccines. In this study, we evaluated a heterologously expressed and purified type IV pilus protein (PilV) of B. pseudomallei strain K96243 as a candidate subunit vaccine. PilV protein was assessed for its ability to protect BALB/c mice against B. pseudomallei strain G207 infection. Mice subcutaneously immunized with purified PilV protein produced high titers

Targeted vaccine selection in influenza vaccination.

BACKGROUND: The main target groups for influenza vaccination are the elderly, the chronically ill, infants, and toddlers. Influenza vaccines are needed that suit the immunological particularities of each of these age and risk groups. Recent years have seen the approval of influenza vaccines that are more immunogenic than before, but whose use in Germany is limited by the restriction of reimbursement to a small number of vaccines. METHODS: The Medline database was selectively searched for pertinent literature. RESULTS: The suboptimal immunogenicity of conventional influenza vaccines that cont

Recent advance in the development of tuberculosis vaccines in clinical trials and virus-like particle-based vaccine candidates.

Tuberculosis (TB) remains a serious public health threat around the world. An effective vaccine is urgently required for cost-effective, long-term control of TB. However, the only licensed vaccine Bacillus Calmette-Guerin (BCG) is limited to prevent TB for its highly variable efficacy. Substantial progress has been made in research and development (R&D) of TB vaccines in the past decades, and a dozen vaccine candidates, including live attenuated mycobacterial vaccines, killed mycobacterial vaccines, adjuvanted subunit vaccines, viral vector vaccines, and messenger RNA (mRNA) vaccines were deve

Crystal structure of the prefusion surface glycoprotein of the prototypic arenavirus LCMV.

Arenaviruses exist worldwide and can cause hemorrhagic fever and neurologic disease. A single glycoprotein expressed on the viral surface mediates entry into target cells. This glycoprotein, termed GPC, contains a membrane-associated signal peptide, a receptor-binding subunit termed GP1 and a fusion-mediating subunit termed GP2. Although GPC is a critical target of antibodies and vaccines, the structure of the metastable GP1-GP2 prefusion complex has remained elusive for all arenaviruses. Here we describe the crystal structure of the fully glycosylated prefusion GP1-GP2 complex of the prototyp

Broad immunity to SARS-CoV-2 variants of concern mediated by a SARS-CoV-2 receptor-binding domain protein vaccine.

BACKGROUND: The SARS-CoV-2 global pandemic has fuelled the generation of vaccines at an unprecedented pace and scale. However, many challenges remain, including: the emergence of vaccine-resistant mutant viruses, vaccine stability during storage and transport, waning vaccine-induced immunity, and concerns about infrequent adverse events associated with existing vaccines. METHODS: We report on a protein subunit vaccine comprising the receptor-binding domain (RBD) of the ancestral SARS-CoV-2 spike protein, dimerised with an immunoglobulin IgG1 Fc domain. These were tested in conjunction with th
