Acetylcholine receptor (AChR) binding and AChR modulating antibodies were found with approximately the same frequency (86%) in 349 patients with myasthenia gravis (MG). However, the total yield of positive serological results was significantly improved (90%) by assaying AChR modulating antibodies when AChR binding antibodies were not detected, because in 27 patients (8%) only one of the two tests was positive. The immunoprecipitation test for AChR blocking antibodies yielded fewer positive results (52%), but there was a significant correlation between the degree of AChR blockade and generaliza…

In the superior cervical ganglion (SCG) of rats, the interaction of sodium bromide (NaBr) with various drugs which interfere with the GABA system, such as 3-(4-chlorophenyl)-4-aminobutyrate [( + )baclofen, Bac], ( + )bicuculline (Bic), picrotoxin (Pic) and chlorpromazine (CPZ), and the effects of NaBr on the K(+)-induced release of [(3)H]acetylcholine ([(3)H]ACh) were studied in vitro. The effects on the evoked potentials induced by preganglionic stimulation were analysed in situ. The in vitro experiments revealed that 1 mM NaBr inhibits both the basal and the K(+)-induced release of [(3)H]ACh…

In this paper we describe our studies of the effects of calmodulin inhibitors on the relationship between the release of acetylcholine (ACh) and protein phosphorylation induced by tityustoxin (TsTX), K+ and ouabain. We observed that in cortical slices of brain, the TsTX induced release of ACh was inhibited in the presence of the calmodulin inhibitors trifluoperazine, W-7 and prenylamine, but not by trifluoperazine sulphoxide. The release of ACh elicited by ouabain and K+ (33 mM) was also inhibited by trifluoperazine and chlorpromazine. Studies on protein phosphorylation with crude or purified …

Continuous pressure recordings were made in guinea-pig antral pouches in vitro. The antral phasic activity was stimulated by acetylcholine and by the cholinesterase inhibitory agent physostigmine. Dopamine and isoprenaline both significantly impaired antral pressure responses to physostigmine, whereas the acetylcholine-induced response was not significantly reduced. This suggests that both dopamine and isoprenaline to some extent act by reducing the spontaneously released acetylcholine, suggesting that both drugs act on intramural cholinergic neural pathways.

In Xenopus laevis oocytes, adenosine and other purinergic agonists induce a K+-conductance increase that is fully mimicked by intracellular application of cAMP. Acetylcholine suppresses the K+-conductance increase caused by adenosine, by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine, or by intracellular injection of cAMP. This effect of acetylcholine is not mimicked by intracellular injection of Ca2+ or of the Ca-mobilizing agent inositol 1,4,5-trisphosphate. However, adenosine and cAMP responses are inhibited by 4 beta-phorbol 12,13-dibutyrate and 4 beta-phorbol 12-myristate 13-…

The involvement of Ca ions in the mediation of muscarinic Cl- current responses in Xenopus oocytes was studied using the voltage-clamp technique and direct measurements of 45Ca efflux. The injection of Ca into the oocytes produced a dose-dependent transient inward (depolarizing) current carried by Cl. This current was occasionally followed by a second, long-lasting inward current. The muscarinic response was evoked by the application of acetylcholine (ACh). It consisted of a transient inward current response, and a long-lasting inward current response, both inward currents carried by Cl ions. …

The effect of tityustoxin and ruthenium red on the release of acetylcholine from slices of cortex from the brain of the rat was studied. The results showed that ruthenium red stimulated the elicited release of acetylcholine (ACh) by tityustoxin. The effect was dependent on the incubation time and the concentration of ruthenium red. The stimulation was dependent on calcium and was blocked by the presence of tetrodotoxin and ethyleneglycol-bis(beta aminoethyl ether)N,N'-tetraacetic acid (EGTA). It is suggested that the inhibition of buffering by calcium in mitochondria, by ruthenium red increase…

The choline homologue 3-[(trimethylammonio)methyl]catechol (TMC) has been synthesized, and the controllable features of its complex oxidation have been examined spectroscopically and correlated with its toxin binding inactivating reactions with the acetylcholine receptor (AcChR) from Torpedo californica electroplax. Affinity-dependent reactions of early intermediates in the oxidation of TMC are suggested to intercede covalently in this inactivation. At pH 7.4, where the oxidative polymerization of catechols proceeds spontaneously, pyrocatechol produced no effect on the toxin binding function o…

The effects of gallamine, gallopamil and nifedipine on isotonic contractions of the isolated taenia of the guinea-pig caecum produced by acetylcholine (ACh) or carbachol (CCh) were investigated. Gallamine (0.1 to 0.3 mM) inhibited contractions produced by CCh more than those produced by ACh. The difference was still present after pretreatment of the tissue with paraoxon (10 microM for 20 min) to inhibit cholinesterases or in experiments carried out in the presence of tetrodotoxin (0.3 microM) to exclude possible ganglionic stimulation by the agonists. Gallopamil or nifedipine selectively inhib…

The effect of cholecystokinin octapeptide (CCK-8) and its analogue, ceruletide on release of acetylcholine (ACh) from the cerebral cortex was investigated in urethane-anaesthetized and in unanaesthetized rats. Cholecystokinin octapeptide and ceruletide markedly stimulated output of ACh at doses of 1.5 and 5.0 micrograms/kg (i.p.), respectively. This effect was prevented by proglumide (160 mg/kg i.p.), a specific cholecystokinin receptor antagonist. At doses of 10 micrograms/kg (i.p.) and more, both CCK-8 and ceruletide decreased output of ACh from the cerebral cortex. The decrease was prevente…

The effects of trimethyl-tin (anion-hydroxyde ionophore, inhibiting oxydative phosphorylation and H(+)-ATPase) probenecid (inhibitor of anion transport in neural cells) and phenylglyoxal (arginine-specific reagent, inhibiting chloride exchanges in erythrocytes) were examined in Torpedo synaptosomes prepared from electric organ. All drugs significantly reduced the stimulated release of acetylcholine triggered by depolarization of nerve endings with high-K(+) and/or gramicidin D. In contrast, trimethyl-tin, probenecid and phenylglyoxal did not affect the ionophore A23187-induced release of acety…

The enantiomers of perhydrohistrionicotoxin were studied in their effects on endplate currents recorded at the junctional region of sartorius muscles of Rana pipiens. The two optical antipodes progressively decreased the peak amplitude of the endplate currents and were indistinguishable from each other at all times. The enantiomers shortened equally the time constants for endplate current decay, but did not alter their voltage sensitivities. Although perhydrohistrionicotoxin contains 4 chiral centers, complete steric inversion does not alter its effects on the acetylcholine receptor-ion channe…

Studies were made of the effects of the benzodiazepines clonazepam (Cz), flunitrazepam (Fz) and Diazepam (Dz) on the spontaneous release of acetylcholine (ACh) in the anterior horn of the lateral cerebral ventricle of cats superficially anaesthetized with urethane. The amount of released ACh was determined in 15-min samples by a bioassay on a segment from isolated guinea-pig ileum. It was found that benzodiazepines applied i.p. in doses of: 5 mg/kg (Dz), 10 mg/kg (Cz) and 0.5 mg/kg (Fz) decreased the ACh release in the effluent flowing out through the cannulated aqueduct. If the released ACh i…

Tityustoxin (TsTX) increased the release and synthesis of acetylcholine (ACh) in slices of rat brain hippocampus, hypothalamus, thalamus, cortex and striatum. The effect was highest in slices of hippocampus and cortex and smallest in hypothalamus. These effects of TsTX were dependent on the presence of Na+ and Ca2+ in the incubation medium. EGTA, 0.1 mM, blocked the effect of TsTX in all areas, except for the hippocampus and frontal striatum, where a concentration of 10 mM was required. Tetrodotoxin blocked the increase in the release and synthesis of ACh induced by tityustoxin.

Adult ewes were studied to determine the effects of barbiturate anesthesia on renal function and the natriuretic response to renal vasodilation by papaverine and acetylcholine. During a period of consciousness, renal blood flow, glomerular filtration rate, urinary Na excretion (UNaV) and arterial blood pressure were monitored. Renal blood flow was maximally increased by renal arterial infusion of papaverine or acetylcholine and changes in renal function were observed. The sheep were subsequently anesthetized and the experimental protocol was repeated. Barbiturate anesthesia resulted in statist…

Peroxidase-conjugated alpha-bungarotoxin (P-BGT) was used for the ultrastructural localization of the acetylcholine receptor in motor endplates. Brachial biceps muscle specimens were obtained from six patients with myasthenia gravis (MG) (two ocular and four generalized), five other patients with neuromuscular diseases (limb-girdle dystrophy, polymyositis, and amyotrophic lateral sclerosis) and two controls. In all patients with generalized MG, most of the endplates showed a marked decrease in P-BGT binding. In one of two patients with ocular MG, the amount and distribution of P-BGT binding ap…

Acetylcholine (ACh) sensitivity and extrajunctional receptor distribution of the rat diaphragm were closely monitored during the early period following denervation. Both contracture in response to 10 microgram/ml of ACh and extrajunctional binding of [125I]alpha-bungarotoxin ([125I]alpha-BTX)) were first detectable 30 h after cutting the phrenic nerve in the thorax. If the nerve were cut more proximally, leaving a 3.5 cm distal nerve stump, the same level of ACh contracture and [125I]alpha-BTX binding did not appear until 40 h after operation. This 10-h delay was far longer than the 3-h delay …
