Pulmonary sarcomatoid carcinoma is rare, with limited treatment options and poor prognosis. In contrast to other non small cell lung carcinomas, not much is known about its molecular biology. In an endemic country like India, lung cancer is often masked by tuberculosis and presents in advanced stages. We report here an unusual case of pulmonary sarcomatoid carcinoma, in a young non-smoker male, who had co-existent tuberculosis masking and delaying the diagnosis of malignancy. On molecular study, the tumor showed ALK gene rearrangement, both by immunohistochemistry and fluorescence in-situ hybr

Hereditary hemorrhagic telangiectasia (HHT) is a rare autosomal dominant disease. The diagnostic criteria of HHT, or Curaçao criteria, include the following: recurrent epistaxis or nighttime nose bleeding, mucocutaneous telangiectases, visceral arteriovenous malformation, or an appropriate family history. The diagnosis is classified as definite if three criteria are present, possible if two criteria are present, and unlikely if only one is present. Nowadays, the confirmation of HHT diagnosis is based on molecular genetic studies. It has been showed that only mutations of genes encoding protein

AIMS: Hereditary hemorrhagic telangiectasia is an autosomal dominant disorder characterized by telangiectasia, epistaxis, and vascular malformations. Pathogenic mutations were found in ENG, AVCRL1, SMAD4, and GDF genes. In this study, we present our database of patients with hereditary hemorrhagic telangiectasia regarding the phenotype-genotype relations and discuss two novel ENG gene pathogenic variations in two unrelated families.
METHODS: Next Generation Sequencing analysis was performed on the peripheral blood of nine patients with hereditary hemorrhagic telangiectasia in four unrelated f

Canid reproduction is unique among other mammals in that females experience long and variable periods of ovarian inactivity. While the domestic dog exhibits a non-seasonal, largely sporadic monoestrus occurring once or twice a year, most wild canids, such as the gray wolf (Canis lupus) and red wolf (Canis rufus), are seasonal breeders with onset apparently dependent on species, latitudinal location and/or variety of environment factors. Neuroendocrine controls of ovarian functions have been mostly studied in the dog, but less so in their wild counterparts, due to difficulties in regular blood

Nemaline myopathies (NMs) are a group of congenital muscle diseases caused by mutations in at least 10 genes and associated with a range of clinical symptoms. NM is defined on muscle biopsy by the presence of cytoplasmic rod-like structures (nemaline rods) composed of cytoskeletal material. Myofiber smallness is also found in many cases of NM and may represent a cause of weakness that can be counteracted by treatment. We have used i.p. injection of activin type IIB receptor (ActRIIB)-mFc (an inhibitor of myostatin signaling) to promote hypertrophy and increase strength in our prior murine work

Trophoblast stem cells (TSCs) serve as a valuable system for investigating the signaling pathways that regulate extraembryonic lineage commitment. Although trophoblast specification has been extensively characterized in humans and rodents, comparable tools and molecular frameworks remain underdeveloped in pigs, a species of significant agricultural and biomedical relevance. This study identifies defined biochemical conditions that enable efficient conversion of porcine (p) expanded pluripotent stem cells (EPSCs) into induced TSCs (iTSCs). The results demonstrate that combined inhibition of TGF

Glucose is traditionally viewed as a substrate for ATP production and glycogen storage in skeletal muscle. Here, we review evidence that glucose also serves as a building block for biomass synthesis in proliferating muscle satellite (stem) cells and hypertrophying skeletal muscle fibers, drawing parallels to anabolic metabolic reprogramming in cancer cells. In cancer and other growing cells, increased glucose uptake, aerobic glycolysis (Warburg effect), and the TCA cycle provide substrates for anabolic pathways that generate macromolecules required for growth and proliferation. Skeletal muscle is increasingly recognized as a dynamic endocrine and paracrine organ that communicates with distal tissues through a diverse secretome of peptides, proteins, metabolites, and extracellular vesicles (EVs), collectively referred to as myokines and exerkines. Beyond cataloging individual factors, emerging evidence suggests that muscle-derived signals can convey information through an integrated, context-dependent "endocrine code"-a pattern defined by secretion kinetics, co-released signal combinations, delivery modalities, and target-tissue receptor landscapes. This review synthe

Systemic delivery of mRNA therapeutics to lung cancer tumors remains challenging despite recent advances in organ-targeting lipid nanoparticles (LNPs). This study introduces novel LNPs, comprising commercially available DC-cholesterol (a cationic derivative of cholesterol) and 113-O12B ionizable lipid (DC_113), which provide selective mRNA translation in healthy lungs while achieving preferential protein expression in pulmonary tumors compared to adjacent lung tissue in an orthotopic cancer model. Analysis of LNPs with diverse ionizable lipids revealed that DC_113 has an apparent pKa above 7.9

Sotatercept, a first-in-class activin signaling inhibitor, has demonstrated disease-modifying efficacy in adults with pulmonary arterial hypertension (PAH). Pediatric data remain limited to ongoing clinical trials. We conducted an IRB-approved, single-center retrospective analysis of pediatric patients with WHO Group 1 or Group 3 PAH treated with off-label sotatercept at Rady Children's Hospital / UC San Diego. All patients had received maximal background triple therapy yet demonstrated persistent symptoms or adverse hemodynamics. Right heart catheterization was performed before sotatercept in

Current weight-loss drugs, such as GLP-1 receptor agonists, are effective at reducing body weight but are often associated with concomitant loss of muscle mass. Thus, there is growing interest in developing next-generation anti-obesity agents that preserve muscle tissue. ACVR2A has emerged as a promising target in this context, as its inhibition may uncouple fat loss from muscle wasting. While several anti-ACVR2A monoclonal antibodies have advanced into clinical trials, small-molecule inhibitors remain scarcely reported. In this study, virtual screening followed by experimental validation was

Sarcopenia, defined as the progressive decline of muscle mass, strength, and function, severely compromises autonomy and quality of life in older adults. This systematic review evaluated synergistic effects of protein supplementation combined with resistance exercise on biochemical and functional biomarkers of sarcopenia. The search for scientific evidence was conducted in PubMed, Scopus, ScienceDirect, and Cochrane databases (2019-2025), applying explicit inclusion and exclusion criteria, like only randomized controlled trials in humans, published in English, Spanish, or French, were included

PURPOSE OF REVIEW: Systemic sclerosis (SSc) is a chronic, severe autoimmune disease characterized by progressive fibrosis of the skin and internal organs. Pulmonary (arterial) hypertension (P(A)H) is a common complication and leading cause of death. This review highlights preclinical and clinical advances of recent years that contribute to our understanding of pathomechanisms, risk stratification, treatment and potential targets in SSc-P(A)H.
RECENT FINDINGS: As SSc-PH emerges from a complex interplay of immune dysregulation, endothelial injury, metabolic reprogramming and fibroproliferative

Early-onset preeclampsia (EOPE) remains a leading cause of maternal and perinatal morbidity worldwide. This review synthesizes evidence that premature placental senescence, centered on syncytiotrophoblast stress and the senescence-associated secretory phenotype (SASP), provides a mechanistic framework connecting placental injury to the maternal syndrome. In EOPE, defective spiral artery remodeling (45) and repeated ischemia-reperfusion can promote oxidative damage, persistent DNA-damage responses, impaired autophagy, and durable cell-cycle arrest in trophoblast lineages. Convergent data show e

Diabetes mellitus (DM) is characterized by the loss or dysfunction pancreatic β-cells. Human amniotic epithelial cells (hAEC), which retain pluripotency markers and are readily obtainable from term placentas, represent a promising alternative source of stem cells. We investigated whether hAECs can be guided through pancreatic ontogeny to generate insulin-producing β-like cells. hAEC from uncomplicated term deliveries were expanded to passage 1 and exposed to a four-stage differentiation sequence that sequentially modulated Activin/WNT, KGF/TGF-β, retinoic-acid/hedgehog, and EGF/Noggin signalin

The transforming growth factor-β (TGF-β) pathway typically inhibits tumorigenesis but can promote metastasis during cancer progression. Activin receptor-like kinase 4 (ALK4), a type I TGF-β family receptor, is frequently downregulated or mutated in cancers, and reduced ALK4 expression correlates with poorer outcomes. However, its role and mechanism of action in cancer progression remains unclear. We demonstrate that ALK4 loss enhances anchorage-independent growth, migration, invasion, and epithelial-mesenchymal transition in vitro, as well as cancer progression in breast and pancreatic cancer

BACKGROUND: Granulosa-like cells (GLCs) were differentiated from human endometrium-derived induced pluripotent stem cells (heiPSCs). This study aimed to establish a GLC differentiation protocol as a defined means to produce autologous estradiol (in vitro), highlighting its therapeutic potential as an alternative to conventional menopausal hormone therapy.
METHODS: Endometrial cells from hysterectomy specimens were reprogrammed into iPSCs using episomal vectors encoding SOX2, OCT4, c-MYC, and KLF4. Differentiation was induced using Activin A and CHIR99021 for mesoderm induction, followed by BM

The advent of Janus kinase inhibitors (JAKis) inaugurated a novel era in the treatment of myelofibrosis (MF), a myeloproliferative neoplasm with heterogeneous clinical manifestations. Four JAKis have been approved for intermediate or high-risk MF, in the United States. Regulatory approval of the first JAK1/2 inhibitor, ruxolitinib, in 2011, transformed the landscape of MF by markedly controlling splenomegaly and constitutional symptoms, improving patients' quality of life, and prolonging survival. Fedratinib, the second approved JAKi, is preferred in the second-line setting. Ruxolitinib and fe

» Fibrodysplasia ossificans progressiva (FOP), the most severe form of heterotopic ossification (HO) in humans, may lead to severe thoracic deformity (TD) and thoracic insufficiency syndrome (TIS), the most common cause of mortality in individuals with FOP.» In this study, we examine the multiple causes of TD in FOP and propose a dynamic model for the development and evolution of TD that can be used to guide clinical care. This model posits that multiple factors, all originating from the causative gain-of-function mutation in activin receptor A, type 1, lead to TD in FOP.» Factors that lead to

Sporadic inclusion body myositis (IBM) is a highly debilitating muscle degenerative and rare disease of the middle aged and elderly. Because immunosuppressants fail to prevent muscle wasting in IBM patients and can even exacerbate it, drugs like AVGN7.2 are being developed to halt degeneration and to enhance muscle mass and function. AVGN7.2 is a novel gene therapeutic that attenuates activin receptors through muscle-specific human (h) SMAD7 expression and as part of its preclinical development, we performed a 91-day single-dose toxicology assessment of systemic safety, biodistribution and imm
