Cornering HIV: taking advantage of interactions between selective pressures.

Adaptive immune responses, cellular restrictive factors and antiretroviral drugs, target multiple regions in the Human Immunodeficiency Virus (HIV) proteome, imposing diverse pressures to viral adaptation. However, the virus is remarkably able to escape from these pressures as mutations are selected. In many cases these mutants have diminished viral fitness. We propose that the concerted action of strategically placed agents and pressures in a host can limit HIV variation capacity while inhibiting its replication. These mechanisms would corner HIV by selecting conflicting adaptive mutations, e

Ischaemic preconditioning protects against ischaemia/reperfusion injury: emerging concepts.

INTRODUCTION: Ischaemic preconditioning (IP) has emerged as a powerful method of ameliorating ischaemia/reperfusion (I/R) injury to the myocardium. This review investigates whether this phenomenon is universally applicable in modulating I/R injury to other tissues. METHODS: A Medline search was conducted to identify both animal and human studies that described IP-induced protection from I/R injury in a variety of non-cardiac organ systems. Particular emphasis was placed on elucidation of underlying physiological concepts. RESULTS AND CONCLUSIONS: IP utilises endogenous mechanisms in skeletal

The many and varied niches occupied by Yersinia pestis as an arthropod-vectored zoonotic pathogen.

Yersinia pestis, the causative agent of bubonic and pneumonic plague, has a complex lifestyle, cycling between both arthropod and mammalian hosts. This pathogen has previously been shown to survive intracellularly within macrophages and to be capable of biofilm formation within the flea, suggesting the development of a range of strategies to ensure survival throughout its life cycle, including expression of virulence factors and tight regulation of its genes.

Adaptive and maladaptive motor axonal sprouting in aging and motoneuron disease.

Motor unit (MU) enlargement by sprouting is an important compensatory mechanism for loss of functional MUs during normal aging and neuromuscular disease. Perisynaptic Schwann cells at neuromuscular junctions extend processes that bridge between denervated and reinnervated endplates, and guide axonal sprouts to reinnervate the denervated endplates. In a rat model of partial denervation, high levels of daily neuromuscular activity have been shown to inhibit the outgrowth of sprouts by preventing Schwann cell bridging. In this review, we consider (1) the relative roles of increasing levels of oxi

Fossil Cercopithecidae from the Hadar Formation and surrounding areas of the Afar Depression, Ethiopia.

Hadar is well known as one of the most productive early hominin sites in the world. Between 1972 and 1994 a large sample of fossil cercopithecid specimens was collected from Hadar and the nearby sites of Geraru, Ahmado, and Leadu. At least five, and possibly six, species are present in the sample, including two chronological subspecies of Theropithecus oswaldi. T. o. cf. darti is known from the Middle Pliocene deposits in the Hadar area, along with Parapapio cf. jonesi, cf. Rhinocolobus turkanaensis, and a new species of Cercopithecoides, C. meaveae. There are also isolated molars from the Mid

Adaptive and phylogenetic significance of ontogenetic sequences in Archaeolemur, subfossil lemur from Madagascar.

Among the best known of recently extinct Malagasy lemurs is Archaeolemur, which is represented by many hundreds of specimens. The phylogenetic affinities of this taxon are unclear, especially in light of recent preliminary analysis of ancient DNA which does not support its previously accepted close relationship with the living Indridae. We examined the nearly complete skeletons of two adults and one juvenile and other less complete specimens to reconstruct aspects of the ontogeny of Archaeolemur. To compare the development of Archaeolemur to that of living strepsirrhines we collected data on P

Reflexes from airway rapidly adapting receptors.

Rapidly adapting receptors (RARs) occur throughout the respiratory tract from the nose to the bronchi. They have thin myelinated nerve fibres, an irregular discharge and adapt rapidly to a maintained volume stimulus, but often slowly to a chemical stimulus. They are polymodal, responding to mechanical and chemical irritant stimuli, and to many inflammatory and immunological mediators. RARs show very varied sensitivities to different stimuli, and diverse reflex responses. Those in the larynx are usually called 'irritant' receptors. They probably cause cough, the expiration reflex and other lary

Effect of acoustic clutter on prey detection by bats.

Bats that capture animal prey from substrates often emit characteristic echolocation calls that are short-duration, frequency-modulated (FM) and broadband. Such calls seem to be suited to locating prey in uncluttered habitats, including flying prey, but may be less effective for finding prey among cluttered backgrounds because echoes reflecting from the substrate mask the acoustic signature of prey. Perhaps these call designs serve primarily for spatial orientation. Furthermore, it has been unclear whether the acoustic image conveyed by FM echoes enables fine texture discrimination, or whether

Genotype effects on neurodegeneration and neuroadaptation in monoaminergic neurotransmitter systems.

Neuroadaptation and neurodegeneration in central dopaminergic and serotonergic systems are central to vulnerability, process and consequences of addictive behavior. Serotonergic dysfunction has been associated with behavior disinhibition and negative mood states that may predispose to excessive alcohol intake, while alcohol-induced stimulation of dopaminergic neurotransmission may encode the reinforcing properties of alcohol consumption. Chronic alcohol intake induces neuroadaptive reductions in striatal dopamine transporter (DAT) and D2 receptor availability, which were reversible during earl

The evolution of human pathogens: examples and clinical implications.

Recent advances in sequencing of complete bacterial genomes, molecular typing of micro-organisms, and research on microbial pathogenicity factors changed our view on the evolution of human bacterial pathogens. We review current evolutionary concepts on plague and meningococcal disease to illustrate the interplay of molecular phylogeny, epidemiology, and pathogenicity research. Furthermore, examples of the tremendous velocity of bacterial evolution under changing environmental conditions will be discussed.

Serotonin neural adaptations to ontogenetic loss of dopamine neurons in rat brain.

In rat, the neonatal destruction of nigrostriatal dopamine (DA) neurons by intracerebral administration of 6-hydroxydopamine entails dramatic changes in serotonin (5-hydroxytryptamine, 5-HT) as well as DA function. Most striking is the 5-HT hyperinnervation of the adult neostriatum, associated with increases in density of various 5-HT receptor subtypes and enhanced neuronal responsiveness to the iontophoretic application of 5-HT and its 5-HT(1B/2C) and 5-HT(2A/2C) receptor agonists, m-chlorophenylpiperazine and iododimethoxyphenylaminopropane. The topographical distribution of these changes is

Neonatal insulin secretion: implications for the programming of metabolic homeostasis.

Patterns of metabolic adaptation are described in the neonate, which generate two fundamental concepts. First, that early nutritional experiences may have long-term effects on the control of metabolic homeostasis, and second, that insulin has a fundamental role in this process. The endocrine pancreas in the neonate is unable to regulate insulin secretion in relation to blood glucose concentration with the same level of tight control seen in the older child and adult. Moreover, the pattern of metabolic adaptation in the fullterm infant in the first postnatal week is different to that of the pre

[The individual manifestations of respiratory rhythms].

The information about the individual peculiarities of the respiratory rhythmics of health people is produced in this article. It is shown, that the functional tests: voluntary hyperventilation, hypoxic hypoxia, their combined influence and natural sleep in normal and hypoxic conditions may be the convenient models for researching this problem. The dependence of the respiratory responses on the individual type of respiration in normal conditions is revealed. The authors concluded that not only the character of normal breathing but also the reactions on functional tests should be taken into acco

Adaptation and cross-resistance: an explanation for the existence of different radiosensitivities among malignant melanoma cells.

A hypothesis is presented to explain the apparent difference in the radioresponsiveness of melanoma lesions whether they are located on the skin or in other parts of the body. The hypothesis states that the radiosensitivity of a cell may change when the cell adapts to live and grow in a different environment. The most important environmental factor that affects the radiosensitivity of cutaneous melanoma cells appears to be the partial pressure of oxygen in their immediate environment. By virtue of adapting to grow in an environment having a high partial pressure of oxygen, the melanoma cells l

Correlation between proto-oncogene, fibroblast growth factor and adaptive response in brain infarct.

Ischemia/hypoxia rapidly induce ischemic changes in vulnerable neurons: cortical neurons in layers II-III and V, hippocampal neurons, cerebellar Purkinje cells and certain basal ganglia and brainstem neurons. The ischemic changes are manifested histologically by nuclear pyknosis, cytoplasmic shrinkage and basophilia. These neurons exhibit strong and persistent expression of immediate early genes (IEGs): c-fos and c-jun. The onset of IEG expression is followed within a day by enhanced bFGF expression in non-ischemic neurons in the same general regions. The appearance of bFGF is followed within
