The human decidua establishes immune tolerance at the maternal-fetal interface and is essential for successful embryo implantation and development. Here, we conducted a spatial transcriptomic analysis of human decidua from early pregnancies in both healthy donors and patients with recurrent pregnancy loss (RPL). Our analysis revealed two distinct spatial domains, named implantation zone (IZ) and glandular-secretory zone (GZ), corresponding to the layers of decidua compacta and spongiosa, respectively. The decidual natural killer cell subset (dNK1) and the decidual macrophage subset (dM2), both

Artificial mitochondrial transplantation (AMT) holds great promise for reprogramming cellular metabolism and restoring cell function. Its clinical translation, however, relies on access to mitochondria that are both of high purity and metabolically active, requirements that current isolation techniques struggle to meet. Conventional differential centrifugation (DC) method yields heterogeneous and low-activity mitochondria, whereas magnetic bead (MB)-based immuno-isolation leaves non-biodegradable beads permanently attached. Herein, we present a Light-Activated Mitochondrial Isolation (LAMI) pl

There is broad consensus that successful repair of severe peripheral nerve injuries requires recreating key structural and cellular features of the natural regenerative process, particularly the action of Bands of Büngner (BoB), longitudinal Schwann cell (SC) structures that guide regenerating axons. Current biomaterial-based strategies have shown limited efficacy, in part because they do not sufficiently reproduce the anisotropic and cellular microenvironment established by BoB, resulting in disorganized axonal growth and reduced regenerative efficiency across long gaps. To address this limit

Psoriasis has a complex immune microenvironment yet most spatial analyses remain two dimensional with routine histopathology. We examined whether immunohistochemistry (IHC) combined with three-dimensional (3D) digital reconstruction can quantify the immune-epithelial architecture across the psoriatic spectrum. Serial sections from 15 psoriasis patients (with lesional and peri-lesional skin 1 cm away from the psoriatic plaque) and 52 healthy donors were reconstructed into 174 tissue stacks (10,700 whole-slide images). CD3+ T cells, CD68+ myeloid cells and mast cells were mapped in 3D including

Cortisol or its medicinal form hydrocortisone (HCT) in accessible biofluids is widely used as a biomarker of stress and stress-related disorders, including Cushing syndrome, chronic fatigue syndrome, post-traumatic stress disorders, and fibromyalgia. Here, we report a simple nanomaterial-free electrochemical sensing strategy for selective cortisol detection based on a molecularly imprinted polymer (MIP) formed directly on indium tin oxide (ITO). The resulting sensor exhibited a wide linear dynamic range (100 pg/mL to 160 ng/mL) in artificial sweat, covering clinically relevant cortisol concent

Human pluripotent stem cells, including embryonic stem cells and induced pluripotent stem cells, can be directed to self-organize into three-dimensional retinal organoids that recapitulate the development, cellular architecture, and key functional features of the human retina. These organoids provide a powerful in vitro platform that models key stages of human retinogenesis and offers unique opportunities to study mechanisms of retinal development and disease in a human context that is not easily accessible in vivo. Recent advances in genome editing, single-cell and multiomics technologies, an

Lysosome-targeting chimeras (LYTACs) have emerged as a strategy for eliminating secreted, extracellular, and plasma-membrane proteins by redirecting them to the endolysosomal system. By coupling target recognition to receptor-mediated uptake, LYTACs exploit endogenous trafficking pathways to access disease-associated proteins beyond the reach of conventional intracellular degradation mechanisms. Related target-intrinsic and cross-linking-driven strategies can likewise promote lysosomal target delivery without recruiting a separate clearance receptor. However, target internalization alone does

Viral infection transforms microbial cells into metabolically reprogrammed "virocells," yet the metabolic architecture underlying this transition remains poorly resolved. Here we investigate how bacteriophages reshape host metabolism in Cellulophaga baltica across time and phage types, uncovering coordinated metabolic reorganization during infection that includes pathway-level patterns consistent with membrane remodeling, amino-acid recycling, and redistribution of cellular resources. Temporal analyses further distinguished infection strategies: efficient phages produced structured, phased met

Late maturity α-amylase (LMA) poses severe quality and economic threats to cereal crops by degrading grain quality through cold-induced alpha-amylase synthesis. Despite its economic impact, the genetic mechanism of cold-induced LMA remains poorly understood, particularly in hexaploid wheat due to its large, redundant genome. To overcome these limitations, we used barley as a diploid model to dissect the genetic architecture of cold-induced LMA. A genome-wide association study (GWAS) was conducted on 352 accessions from the mini-world barley core collection grown under controlled normal and col

Multiscale tissue ecosystems are governed by coupled intracellular decision-making, cell-cell interactions, and spatially structured microenvironmental signals, yet these scales are often studied separately. Here we present MISSTE, a modular framework that integrates Boolean intracellular state logic, agent-based modeling, and partial differential equation fields within a unified spatial simulation architecture. As a proof of concept, we applied MISSTE to CAR-T therapy in a solid tumor microenvironment. The model recapitulated emergent features of CAR-T behavior, including limited tumor penetr

Erythropoiesis is a complex, spatially constrained morphogenetic process that transforms hematopoietic progenitors into specialized, enucleated red blood cells. While decades of research have defined the cell-intrinsic transcriptional and epigenetic circuits, governed by master regulators such as GATA1 and KLF1, the critical role of the microenvironmental niche remains less understood. Recent advances in single-cell omics have revealed a multi-layered landscape in which transcriptional potential is often uncoupled from proteomic and metabolic output. The transition from dissociative single-cel

Artemisinin combination therapies are central to malaria control, but their efficacy is threatened by the emergence of resistant Plasmodium falciparum strains. While the role of Pfkelch13 mutations is well established, mutations in Pfcoronin have also been implicated in treatment failure, revealing layers of resistance complexity. Here, we define the cellular mechanism of Pfcoronin‑mediated artemisinin resistance. PfCoronin interacts with PfActin and localizes to the parasite plasma membrane, the digestive vacuole membrane, and small vesicles containing host cell material. Mutations in PfCoron

Multi-contrast optical coherence tomography (MC-OCT), incorporating optical coherence tomography angiography (OCTA) and polarization-sensitive OCT (PS-OCT), offers a powerful non-invasive approach for interrogating ovarian microvascular and extracellular matrix (ECM) remodeling in vivo. However, its application to ovarian aging has remained largely unexplored. Here, we establish and validate MC-OCT as a robust imaging platform for visualizing and quantifying age-related changes in vascular architecture and collagen organization in mouse ovaries. Using in vivo OCTA, we demonstrate a progressive

Much of the regulatory information that shapes plant gene expression lies outside protein-coding regions, including many loci associated with agronomic traits. Deep learning models use DNA sequences and multi-omics data to examine components of this cis-regulatory information. This review compares convolutional, Transformer-based and graph architectures used to represent local sequence features, chromatin state and three-dimensional genome organisation. We assess their applications to transcription-factor binding, chromatin accessibility, gene expression, non-coding variant prioritisation and

The hierarchical organization of the eukaryotic genome is crucial for nuclear activities and cellular development. Genetic aberrations can disrupt this 3D genomic architecture, potentially driving oncogenesis. However, current research often lacks a comprehensive perspective, focusing on specific mutation types and singular 3D structural levels. Here, pathological changes from chromosomes to nucleotides are systematically cataloged, including 10 789 interchromosomal translocations (ICTs), 18 863 structural variants (SVs), and 162 769 single nucleotide polymorphisms (SNPs). The multilayered ana

Parkinson's disease, currently the fastest-growing neurodegenerative disorder globally, has seen a 50% increase in cases within just two years. As disease progression impairs speech, memory, and motor functions over time, early diagnosis is crucial for preserving patients' quality of life. Although machine-learning-based detection has shown promise for detecting Parkinson's disease, most studies rely on a single feature for classification and can be error-prone due to the variability of symptoms between patients. To address this limitation we utilized the mPower dataset, which includes 150,000

The HIV-1 restriction factor, hSERINC3, functions as a lipid scramblase, translocating lipids across the bilayer in reconstituted proteoliposomes and the viral envelope. Phosphatidylserine(PS) scrambling and exposure at the outer leaflet are recognized to play important roles in several biological processes. To understand the mechanistic basis for hSERINC3-mediated PS lipid scrambling at atomistic resolution, we implemented the transition-tempered metadynamics (TTMetaD) enhanced sampling method. Our simulations sampled close-to-open hSERINC3 conformational transition during PS scrambling and d

Understanding the etiology of human neurological and psychiatric disorders remains challenging due to species-specific genomic architectures, prolonged developmental timelines, and the unique cytoarchitectural features of the human brain, which are inadequately replicated in conventional animal models. To address these limitations, human pluripotent stem cell (hPSC)-derived brain organoids have emerged as an ethically accessible experimental system that recapitulates critical aspects of early human neurodevelopment in vitro. These three-dimensional (3D) models mimic the generation of neural pr

Recent developments in artificial intelligence (AI) have introduced new technologies that can aid in detecting cognitive decline. This study developed a voice-based AI model that screens for cognitive decline using only a short conversational voice sample. The process involved collecting voice samples, applying machine learning (ML), and confirming accuracy through test data. The AI model extracts multiple voice features from the collected voice data to detect potential signs of cognitive impairment. Data labeling for ML was based on Mini-Mental State Examination scores: scores of 23 or lower
