Stroke drives glioma progression through the emergence of tumor-associated astrocytes with reduced Ca2+ activity.

Epidemiological studies suggest that persons with a history of stroke are at increased risk of developing brain tumors, yet the mechanisms linking brain injury to glioma progression remain unclear. Here we show that stroke promotes tumor infiltration into injured brain regions in human and mouse glioma models, accompanied by reduced overall survival. Stroke induces remodeling of the tumor microenvironment, including the emergence of a distinct population of tumor-associated astrocytes (TAAs) with diminished Ca2+ activity at the invasive front and enrichment of tumor-associated microglia and ma…

Central nervous system resilience to cyclophosphamide: preserved behavior, astrocytes and LGALS3 expression in immunosuppressed mice.

BACKGROUND: Cyclophosphamide is widely used as an immunosuppressive agent in experimental models. However, its potential effect on central nervous system (CNS) function and behavior may confound studies investigating neuroimmune interactions, interpretation of results, and translational relevance. OBJECTIVES: To characterize longitudinal behavioral outcomes and CNS-related molecular and histological changes following repeated cyclophosphamide-induced immunosuppression in mice. METHODS: Adult male C57BL/6 mice received an intraperitoneal injection of cyclophosphamide (100 mg/kg) for three or

IFNγ and proinflammatory cytokines cooperatively license astrocytes to support antigen-dependent CD4+ T cell restimulation.

We report that type 2 interferon with proinflammatory cytokines induces astrocytes to express multiple components required for attraction of and antigen presentation to CD4+ T cells via MHC-II molecules. Re-analysis of astrocyte-enriched single-cell sequencing from bacterial mimetic-injected mice revealed a transcriptionally distinct interferon-responsive astrocyte population enriched for antigen-presentation-associated genes 24 hours after peripheral immune challenge. In vivo, we identify MHC-II+GFAP+ astrocytes localized to the surface of the brain across multiple mouse models of neuroinflam…

Neurons derived from NeuroD1-expressing astrocytes transition through transit-amplifying intermediates but lack functional maturity.

In vivo conversion of nonneuronal cells into neurons is a proposed strategy to replace neurons lost to CNS injury or disease. Glia-to-neuron … t-amplifying OLIG2+ progenitors during early injury phase, but not in late phases or in nonreactive astrocytes. Neither a loss-of-function NeuroD1 mutant nor stemness-reprogramming factor SOX2 induces astrocyte-to-neuron conversion. However, contrary to previous reports, the neuronal-like cells generated by NeuroD1 lack mature neuroelectrical properties, lim …

Fibroblast Growth Factor 8 Suppresses Neurotoxic Astrocytes and Alleviates Neuropathic Pain via Spinal FGFR3 Signaling.

Astrocytes in the spinal dorsal horn (SDH) exhibit diverse reactive phenotypes under neuropathic conditions, yet the mechanisms driving this diversity and its implications in chronic pain remain unclear. Here, we report that spared nerve injury (SNI) induces marked upregulation of both complement component 3 (C3⁺, A1-like) and S100 calcium-binding protein A10 (S100A10⁺, A2-like) astrocyte subpopulations in the SDH, with elevated microglial cytokines including interleukin-1α, tumor necrosis factor-α, and complement component 1q. Transcriptomic, immunohistochemical, and Western blot analyses rev…

Metabolic reprogramming in astrocytes prevents neuronal death through a UCHL1/PFKFB3/H4K8la positive feedback loop.

Astrocytic metabolic reprogramming is an adaptation of metabolic patterns to meet increased energy demands, although the role after spinal cord injury (SCI) remains unclear. Analysis of single-cell RNA sequencing (scRNA-seq) data identified an increase in astrocytic glycolysis, while PFKFB3, a key regulator of glycolytic flux, was significantly upregulated following SCI. Loss of PFKFB3 in astrocytes prohibited neuronal energy supply and enhanced neuronal ferroptosis in vitro and expanded infiltration of CD68+ macrophages/microglia, exacerbated neuronal loss, and hindered functional recovery in…

Extracellular Vesicles Derived from FGF2-Primed Astrocytes Against Mitochondrial and Synaptic Toxicities in Parkinson's Disease.

PURPOSE: Mitochondrial dysfunction associated with neuronal degeneration and subsequent synaptic disconnection are essential for the development of Parkinson's disease (PD). Considering that astrocytes play key roles in synaptogenesis during development, we hypothesized that fibroblast growth factor - 2 (FGF2), a key factor for astrocyte development, could reverse the toxic phenotype of reactive astrocytes, and the extracellular vesicles (EVs) derived from FGF2-primed astrocytes would enhance synaptogenesis in PD model. The present study was to test this hypothesis. METHODS: EVs isolated from…

Lactate-mediated Ran lactylation at lysine 123 promotes astrocytes polarization after oxygen-glucose deprivation/reoxygenation.

Ischemia and hypoxia near the injury site following spinal cord injury (SCI) lead to early lactate accumulation. Reactive astrocytes polarize and migrate to the lesion site, sealing it off and restricting immune cell infiltration. However, the specific mechanisms through which lactate influences astrocyte polarization remain poorly understood. In this study, we found that lactate can promote astrocyte proliferation, migration and differentiation into A2 after oxygen-glucose deprivation/reoxygenation (OGD/R). Inhibiting signal transducer and activator of transcription 3 (STAT3) nuclear transpor…

Inhibition of hypoxic exosomal miR-423-3p decreases glioma progression by restricting autophagy in astrocytes.

The tumor microenvironment (TME) of gliomas comprises glioma cells and surrounding cells, such as astrocytes, macrophages, T cells, and neurons. In the TME, glioma cells can activate normal human astrocytes (NHAs) through the secretion of exosomes and the activation of astrocytes can further improve the progression of glioma, leading to a poor prognosis for patients. However, the molecular mechanisms underlying NHAs activation by gliomas remain largely unknown. It this study, glioma-derived exosomes (GDEs) play an important role in the modulation of autophagy and activation of NHAs. Compared w…

Effect of metformin on spontaneous calcium signaling in cultured astrocytes during normoxic and hypoxic conditions.

Astrocyte function is controlled by intracellular Ca2+ signaling. On the other hand, hypoxia influences calcium dynamics and its homeostatic range because of reduction of ATP synthesis, which inhibits ATP dependent processes. By using Ca2+ sensitive fluorescence dye, we studied how metformin changed spontaneous oscillating Ca2+ signals in soma of astrocytes in monocultures, prepared from rat brains. Mild hypoxic conditions (2 % O2) applied for 24 h had no effect on astrocyte viability; however, it reduced the relative amplitude of Ca2+ signals, slowed the decay of the signals, and increased th…

Silencing epileptic storms: targeting miRNA-lncRNA crosstalk in astrocytes and microglia to disarm neuroinflammatory triggers.

Epilepsy is a chronic neurological disorder characterized by abnormal synchronous discharges of neurons in the brain. It affects approximate … al process of epilepsy by regulating the phenotype, inflammatory response and metabolic function of astrocytes and microglia. In addition, long noncoding RNAs, as upstream regulators of miRNAs, influence miRNA function by acting as competitive endogenous RNAs, further regulating glial cell activation and inflammatory responses. Alzheimer's disease brain-derived tau extracts show differential processing and transcriptional effects in human astrocytes.

Post-translational modifications of tau, including phosphorylation at specific residues, are closely linked with tau seeding ability and clinical disease progression. While most previous evidence has focused on neuronal tau spread, evidence supports a similar role for astrocytes. Here, we demonstrate that well characterized tau aggregates isolated from postmortem Alzheimer's disease brain are internalized and processed by control human-induced pluripotent stem cell-derived astrocytes. Differences in the efficiency of tau internalization, clearance and/or seeding were noted, which reflect molec…

Hair follicle stem cell-derived secretome protects astrocytes in an in vitro ischemia/reperfusion model.

Ischemic stroke causes significant neuronal and glial cell damage. Recent studies suggest that stem cell-derived secretomes may offer therapeutic benefits for neural injuries. This study evaluates the protective effects of hair follicle stem cell (HFSC)-derived secretome on astrocytes subjected to oxygen-glucose deprivation (OGD), an in vitro model of ischemic stroke. In this regard, the primary astrocyte cultures were exposed to OGD conditions for 24 hours, followed by treatment with HFSC-derived secretome for 48 hours to create an environment rich in paracrine factors. The neuroprotective ef…

Three-Dimensional Bioprinting of Astrocytes and Endothelial Cells to Direct Retinal Axon Growth and Vascularization.

Retinal organoids (ROs) are currently used to study retinal development and diseases but cannot model glaucoma because they fail to form a nerve fiber layer (NFL) and optic nerve (ON). Utilizing three-dimensional bioprinting, ON head astrocytes (ONHAs) and vascular endothelial cells, both of which contribute to NFL development in vivo but are absent in ROs, were positioned at the center of scaffolds seeded with retinal ganglion cells (RGCs). In experiments using ONHAs isolated from developing retinas, polarization of RGC neurite growth increased by 43% while ONHA from adult retinas or astrocyt…

Agrin-Lrp4 pathway in hippocampal astrocytes restrains development of temporal lobe epilepsy through adenosine signaling.

BACKGROUND: Human patients often experience an episode of serious seizure activity, such as status epilepticus (SE), prior to the onset of t … idal neuron specific knockout mice (Nex-Lrp4-/-). Lrp4 was specifically knocked down in hippocampal astrocytes by injecting AAV virus carrying hGFAP-Cre into the hippocampus. The effects of agrin-Lrp4 pathway on the development of SE-induced TLE were evaluated on the chronic seizure model generated by injecting kainic acid (KA) into the amygdala. The spontan …

Astrocytes in Amyloid Generation and Alcohol Metabolism: Implications of Alcohol Use in Neurological Disorder(s).

As per the National Survey on Drug Use and Health, 10.5% of Americans aged 12 years and older are suffering from alcohol use disorder, with a wide range of neurological disorders. Alcohol-mediated neurological disorders can be linked to Alzheimer's-like pathology, which has not been well studied. We hypothesize that alcohol exposure can induce astrocytic amyloidosis, which can be corroborated by the neurological disorders observed in alcohol use disorder. In this study, we demonstrated that the exposure of astrocytes to ethanol resulted in an increase in Alzheimer's disease markers-the amyloid…

A Pvr-AP-1-Mmp1 signaling pathway is activated in astrocytes upon traumatic brain injury.

Traumatic brain injury (TBI) caused by external mechanical forces is a major health burden worldwide, but the underlying mechanism in glia remains largely unclear. We report herein that Drosophila adults exhibit a defective blood-brain barrier, elevated innate immune responses, and astrocyte swelling upon consecutive strikes with a high-impact trauma device. RNA sequencing (RNA-seq) analysis of these astrocytes revealed upregulated expression of genes encoding PDGF and VEGF receptor-related (Pvr, a receptor tyrosine kinase), adaptor protein complex 1 (AP-1, a transcription factor complex of th…
