Induction of heterotopic and orthotopic cartilage and bone formation in mice.

Heterotopic cartilage, bone and bone-marrow formation was achieved in mice by transplantation of a variety of xenogeneic established cell lines, by the transitional epithelium or by implants of demineralized bone matrix. The pattern and the sequence of events were always the same, regardless of the inducer used; viz., hyaline cartilage appeared 6-7 days after implantation, and endochondral bone formation followed. However, in cases of allogeneic implants of transitional epithelium into species other than the mouse, an intramembranous osteogenesis was the main mode of bone formation. When the y…

Morphologic and biochemical changes in cartilage of foals treated with dexamethasone.

Epiphyseal and articular cartilages were examined in pony foals treated with intramuscular injections of either 0.5 mg dexamethasone per 100 kg bodyweight daily for 3, 8 or 11 months, or 5.0 mg per 100 kg for 11 months, and in horse foals treated with 5.0 mg per 100 kg for 20 weeks. The proximal femoral growth plates exhibited increased spatial separation between chondrocyte columns, narrowed zones of disorganized columnar and hypertrophic cartilage, abnormal penetration of hypertrophic cartilage by metaphyseal capillaries, retained cartilage in the spongiosa, distal terminal plate formation, …

Observations on the senescence of cells derived from articular cartilage.

In the experiments described here, we have sought to determine whether primary cultures of cells derived from articular cartilage will, upon subsequent subculture, undergo in vitro senescence in a manner analogous to that described for several other types of diploid cell. Using cells from the articular cartilage of rabbits, dogs and man, we have established that the population doubling capacity of cultures of these cells is directly related to the specific lifespan of the donor organism. Furthermore, the doubling capacity of the initial cultures of lapine articular chondrocytes is inversely re…

Bilateral arytenoid cartilage lateralization for laryngeal paralysis in the dog.

Bilateral arytenoid cartilage lateralization was used successfully to treat 3 dogs with bilateral laryngeal paralysis. In our hands, this techniques was more effective than unilateral arytenoid cartilage lateralization in resolving the signs of laryngeal paralysis and in avoiding the complications that may follow partial laryngectomy.

In vitro cartilage degradation by Escherichia coli and Staphylococcus aureus.

Effects of Escherichia coli and Staphylococcus aureus on cartilage and chondrocytes in culture are reported. Under these conditions, bacterial effects on cartilage degradation and cell viability are measured in the absence of inflammation. E coli causes a 28% loss and S aureus an 83% loss of cartilage glycosaminoglycan within 48 hours. Collagen content is unchanged. Both bacterial species induce chondrocyte death in explants and in monolayers within 48 hours. Bacterial effects on glycosaminoglycans and cell viability do not result from depletion of nutrients from the culture medium. Serum in t…

Trisomy 9 mosaicism with punctate mineralization in developing cartilages.

A case of mosaic trisomy 9 (46,XX/47,XX,+9) is described. The main clinical findings included intrauterine growth retardation, failure to thrive, hydrocephalus, deeply set eyes, prominent antihelix, highly arched palate, subluxation of the interphalangeal joints of some fingers, hip dislocation, excessive sweating, and punctate mineralization in developing cartilages.

The role of link-protein in the structure of cartilage proteoglycan aggregates.

Proteoglycan fractions were prepared from pig laryngeal cartilage. The effect of link-protein on the properties of proteoglycan-hyaluronate aggregates was examined by viscometry and analytical ultracentrifugation. Aggregates containing link-protein were more stable than link-free aggregates at neutral pH, at temperatures up to 50 degrees C and in urea (up to 4.0M). Oligosaccharides of hyaluronate were able to displace proteoglycans from link-free aggregates, but not from the link-stabilized aggregates. Both types of aggregate were observed in the ultracentrifuge, but at the concentration inves…

[Cartilage of the head of the femoral in articular chondrocalcinosis (author's transl)].

A study of three femoral heads removed from patients suffering from articular chondrocalcinosis enabled the authors to identify the lesions of the cartilage in that disorder. They are mainly represented by crystals of calcium pyrophosphate. They appear within the cartilage which is macroscopically intact and histologically normal, but their location and the related tissue phenomena in their formation remain controversial and poorly understood. They accumulate in the various layers of the non-calcified cartilage within which they appear to hollow out genuine cavities by pushing back and then de…

Effect of salicylate therapy on cartilage destruction in experimental pyarthrosis.

An experimental rabbit pyarthrosis model was utilized to investigate the effects of sodium salicylate therapy on cartilage destruction. Animals were innoculated with 10(3) organisms of Staphylococcus aureus 502A. The opposite knee was injected with sterile saline. Infection was documented 48 hours post innoculation, procaine penicillin therapy initiated, and the adequacy of therapy evaluated at sacrifice. One half of the animals began salicylate feedings one week prior to injection and this continued until sacrifice at 14 or 49 days. Salicylate levels were 12.5 +/- 6.9 mg %. There was a signif…

Extrusion of pyrophosphate into extracellular media by osteoarthritic cartilage incubates.

The distribution of calcium pyrophosphate mineral phase, almost exclusively confined to articular cartilage in chondrocalcinosis, and the high level of pyrophosphate (PPi) ion relative to serum in synovial fluid in patients with either chondrocalcinosis or advanced osteoarthritis led to an investigation of whether cartilage cells elaborate PPi ions. Incubates of articular cartilage from young rabbits but not from mature rabbits, as well as growth plates cartilage, released PPi into incubation media during a 4h period. Control rabbit ear cartilage and synovial membrane elaborated negligible amo…
